Abstract: DNA is widely used in plant genetic and molecular biology studies. In this chapter, we describe how to extract DNA from wheat tissues. The tissue samples are ground to disrupt the cell wall. Then cetyltrimethylammonium bromide (CTAB) or sodium dodecyl sulfate (SDS) is used to disrupt the cell and nuclear membranes to release the DNA into solution. A reducing agent, β-mercaptoethanol, is added to break the disulfide bonds between the cysteine residues and to help remove the tanins and polyphenols. A high concen… Show more

“…DNA from accessions TOWWC0001 to TOWWC0195 was made available from Arora et al (2019) 22 . For accessions TOWWC0196 to TOWW0304, the SHW lines and their diploid and tetraploid donors, high molecular weight DNA was extracted following a modified CTAB protocol 56 . For accessions TOWW0196 to TOWW0304 the same plant was used for DNA and single seed descent.…”

“…The concentration of NaCl varied with plant sample species in a range between 0.7 M (Clark, 1997) to 6 M (Aboul-Maaty & Oraby, 2019;Aljanabi et al, 1999). In contrast, NaCl concentration of 1.5 M or 2.0 M is sufficient to remove high polysaccharide content in DNA sample (Fang et al, 1992;Yu et al, 2017). In the present study, the low amount of NaCl (0.5M) in the modified Dellaporta et al (1983) extraction buffer was found to be sufficient to improve the quality of the extracted DNA sample compared to other extraction buffers supplemented with other NaCl concentrations.…”

“…The potential action of EDTA to sequester metal ions such as Mg 2+ and Ca 2+ from cellular membranes when applied to pulverized plant tissues during incubation at higher temperature can help in plant cell lysis hence suggesting EDTA to be considered as a substitute for detergent (Kumari et al, 2012). In addition, the high affinity of EDTA to divalent cations such as Ca 2+ , Mn 2+ and Mg 2+ acts as the co-factors in preventing endogenous nucleases activities (Adams et al, 1992;Osena et al, 2017;Rosenberg, 1987;Tiwari et al, 2017;Yu et al, 2017). Furthermore, the interaction of Tris hydrochloride (Tris-HCl), EDTA complex and the reducing agent beta-mercaptoethanol (bme) with the outer membrane polysaccharides helps to destabilise cell membrane and further breaks down the cell during cell lysis (Green & Sambrook, 2018;Varma et al, 2007).…”

“…High-quality genomic DNA from the Zak/ZakERA8 BC 3 F 3 bulked populations was extracted using a phenol/chloroform-based DNA extraction method followed by RNAse A digestion (as in Yu et al 2017). A 2.5-cm-long leaf sample was collected from BC 3 F 3 plants and ground together with lines that comprised each bulk.…”
