Streptozotocin‐Induced Diabetic Models in Mice and Rats

Abstract: Streptozotocin (STZ) is an antibiotic that produces pancreatic islet β-cell destruction and is widely used experimentally to produce a model of type 1 diabetes mellitus (T1DM). Detailed in this unit are protocols for producing STZ-induced insulin deficiency and hyperglycemia in mice and rats. Also described are protocols for creating animal models for type 2 diabetes using STZ. These animals are employed for assessing the pathological consequences of diabetes and for screening potential therapies for the treat… Show more

“…Diabetes merupakan kondisi hiperglikemia dengan KGD yang diukur setelah puasa yaitu bernilai 150 mg/dL (Furman, 2015) dengan adanya gejala polyfagia, polydypsia, dan polyurea (Kolluru et al, 2012;Patel et al, 2012). Induksi STZ menyebabkan terjadinya DM tipe 1 yang ditandai dengan penyusutan berat badan (Wang et al, 2014).…”

“…Diabetes was induced in overnight-fasted rats by single intraperitoneal injection of freshly prepared STZ (45 mg/kg body weight) in 0.1 M citrate buffer, pH 4.5 [57]. Hyperglycemia was corroborated by checking the blood glucose (by ACCU-CHEK sensor glucometer) after 48 h of STZ injection.…”

“…Treating a healthy young rat or mouse with a pancreatic beta cell toxin such as streptozotocin (STZ) destroys some pancreatic beta cells, reduces beta cell mass and function and generates an animal with hyperglycaemia [22][23][24]. To study the effect of a proposed "anti-diabetic" compound in such a model is really a toxicology study testing the effect of the intervention on the toxicity of STZ.…”

“…It is also possible to supplement the treatments with STZ to induce hyperglycaemia but this generates mice of poor health and the utility of this, model may not be ideal in terms of mimicking the effect of diabetes on atherosclerosis in humans. The model may be improved by administering low-dose insulin to the mice [23].…”
