Скелетная дисплазия «Поражения костей свода черепа в виде пончика с хрупкостью костей» (ОМIМ: 126550) является чрезвычайно редким наследственным заболеванием c аутосомно-доминантным типом наследования, которое характеризуется низкой минеральной плотностью костной ткани, высокой хрупкостью костей и множественными переломами, начинающимися в детстве, а также склеротическими изменениями в костях свода черепа, при визуализирующих исследованиях напоминающими по форме пончик. У некоторых пациентов отмечается более тяжелое течение заболевания, сопровождающееся неонатальными переломами костей, выраженной низкорослостью и спондилометафизарной дисплазией. Окончательный диагноз устанавливается на основании результата молекулярно-генетического исследования. Ввиду чрезвычайной редкости этого заболевания его описание представляет особый интерес для медицинских специалистов, включая работающих в сфере диагностики и лечения орфанных заболеваний. В статье приведен обзор имеющихся к настоящему моменту данных о заболевании, а также клинический случай, представленный 5 пораженными этим заболеванием членами семьи в 3 поколениях, с генотипически (гетерозиготная мутация в гене SGMS2 (патогенный вариант c.148C>T (p.Arg50Ter)) и фенотипически подтвержденным наличием поражений костей свода черепа в виде пончика с хрупкостью костей. Skeletal dysplasia "Calvarial doughnut lesions with bone fragility" (OMIM: 126550) is an extremely rare autosomal dominant hereditary disorder characterized by low bone mineral density, high bone fragility, and multiple fractures beginning in childhood, as well as sclerotic foci in the bones of the skull, resembling a doughnut shape on imaging studies. Some patients have a more severe course of the disease, accompanied by neonatal fractures, marked short stature, and spondylometaphyseal dysplasia. The final diagnosis is verified on the basis of a molecular genetic study. Due to the extreme rarity of this pathology, its description is of particular interest to medical specialists, including those working in the field of diagnosis and treatment of orphan diseases. The article provides an overview of currently available data on the disease, as well as a clinical case presented by 5 family members affected by this disease in 3 generations, with genotypically (heterozygous mutation in the SGMS2 gene (pathogenic variant c.148C>T (p.Arg50Ter)) and phenotypically confirmed calvarial doughnut lesions with bone fragility.
