Исследовано протективное действие карбамилированного дарбэпоэтина в дозе 300 мкг/кг на модели ишемической нейропатии зрительного нерва у крыс линии Wistar. Выявлено, что карбамилированный дарбэпоэтин предотвращает развитие повреждений диска зрительного нерва и сосудов сетчатки, вызванных внутрижелудочным введением неселективного ингибитора NO-синтаз N-нитро-L-аргинин-метилового эфира (L-NAME) в дозе 12,5 мг/кг в течение 28 сут и однократным повышением внутриглазного давления до 110 мм рт. ст. Обнаруженный протективный эффект подтвержден результатами офтальмоскопии, электроретинографии и иммуногистохимии на 29 сут эксперимента. У крыс в группе с коррекцией карбамилированным дарбэпоэтином коэффициент b/a (степень развития функциональных повреждений сетчатки) достоверно возрастает на 31,6 % (p < 0,05). Карбамилированный дарбэпоэтин ингибирует экспрессию белка р53 в нейронах сетчатки и индуцирует экспрессию эндотелиальной NO-синтазы в сосудах сетчатки, вследствие чего наблюдается протективный эффект.
Исследовано нейропротективное действие агониста имидазолиновых рецепторов типов I и II, 3-(1H-бензимидазол-2-ил)-1,2,2-триметилциклопентанкарбоновой кислоты, в дозе 50 мг/кг в сравнении с пикамилоном в дозе 30 мг/кг на модели ишемической нейропатии зрительного нерва у крыс-самцов линии Wistar. В эксперименте выявлено, что 3-(1H-бензимидазол-2-ил)-1,2,2-триметилциклопентанкарбоновая кислота предотвращает развитие ишемической нейропатии зрительного нерва, вызванной введением неселективного ингибитора NO-синтазы N-нитро-L-аргинин-метилового эфира (L-NAME) в дозе 12,5 мг/кг в течение 28 сут, и однократным повышением внутриглазного давления (ВГД) до 110 мм рт. ст., в большей степени, чем пикамилон. У крыс в группе с коррекцией 3-(1H-бензимидазол-2-ил)-1,2,2-триметилциклопентанкарбоновой кислотой удельное количество ядер нейронов в наружном ядерном слое достоверно выше на 15,0 % (p < 0,05), в сравнении с группой животных с введением пикамилона; на 33,3 % (p < 0,05), в сравнении с группой с моделированием патологии, что свидетельствует о сохранении их жизнеспособности. Обнаруженные протективные эффекты подтверждены результатами офтальмоскопии и гистологических исследований на 29 сут эксперимента.
Глутамат способен вызывать эксайтотоксичность в сетчатке в высоких концентрациях, приводя к гибели ганглионарных клеток. Потенциальными фармакологическими веществами, способными снижать или предупреждать эксайтотоксическое повреждение сетчатки, являются производные 3-гидроксипиридина. Нами была изучена возможность фармакологической коррекции 3-гидрокси-6-метил-2-этилпиридин-N-ацетилтауринатом (EHMP-NAT) повреждений сетчатки, вызванных интравитреальным применением N-метил-D-аспартата (NMDA). Через 7 дней после применения NMDA было выявлено, что парабульбарное введение EHMP-NAT в дозе 4,4 мг/кг/сут с 1 по 4 день эксперимента приводит к увеличению уровня микроциркуляции в сетчатке на 48,5 %, по сравнению с группой NMDA (p < 0,05); увеличению амплитуды a-волны на 52,7 % (p < 0,05), b-волны — в 2,5 раза (p < 0,01), по сравнению с группой NMDA. В группе с EHMP-NAT уровень экспрессии гена каспазы-3 был меньше значения группы NMDA на 46,1 % (p < 0,05); экспрессия NF-κB p65 значительно отличалась как от группы интактных животных (p < 0,05), так и от группы NMDA (p < 0,05); уровень экспрессии гена p53 был на 32,4 % ниже значения группы NMDA (p < 0,05). Таким образом, в данном эксперименте было выявлено, что механизм ретинопротекторного эффекта EHMP-NAT связан с ингибированием NMDA-опосредованной эксайтотоксичности в сетчатке.
