Τα καρκινοειδή του πνεύμονα είναι σπάνιοι όγκοι ανθεκτικοί στη χημειοθεραπεία. Τα ανάλογα σωματοστατίνης χρησιμοποιούνται σήμερα στη θεραπεία για τον έλεγχο της αύξησης του όγκου και της έκκρισης ορμονών, και πρόσφατα ο αναστολέας mTOR everolimus δοκιμάζεται σε μεγάλες κλινικές μελέτες των νευροενδοκρινών όγκων. Δεν υπάρχουν δεδομένα για τη χρήση της καβεργολίνης στη θεραπεία αυτών των όγκων. Ο σκοπός της μελέτης μας είναι να αξιολογηθεί η δράση του συνδυασμού οκτρεοτίδης /καβεργολίνης με τους αναστολείς mTOR in vitro στη μοναδική κυτταρική σειρά τυπικού καρκινοειδούς πνεύμονα: Η727.Μέθοδοι: Στην κυτταρική σειρά Η727 χαρακτηρίστηκε το μονοπάτι του mTOR, ο υποδοχέας σωματοστατίνης 2 (SSTR2) και ο υποδοχέας ντοπαμίνης 2 (D2R) με RT-qPCR και ανοσοκυτταροχημεία φθορισμού. Δεκαπέντε ιστικά δείγματα ασθενών με τυπικό καρκινοειδές των βρόγχων χρησιμοποιήθηκαν για τη μελέτη της έκφρασης των SSTR2, D2R, p70S6K και pp70S6K με ανοσοϊστοχημεία. Η κυτταρική βιωσιμότητα, η κυτταρομετρία ροής και το western blot αξιολογήθηκαν προκειμένου να μελετηθεί η δράση της οκτρεοτίδης, της καβεργολίνης και των αναστολέων mTOR.Αποτελέσματα: Η οκτρεοτίδη και η καβεργολίνη μεμονωμένα δεν αναστέλλουν την κυτταρική βιωσιμότητα αλλά σε συνδυασμό με τους αναστολείς mTOR φαίνεται να ενισχύουν την ανασταλτική δράση των αναστολέων mTOR μετά από μακρά έκθεση και ταυτόχρονα μειώνουν τη φωσφορυλίωση του Akt και αυξάνουν την έκφραση του REDD1/DDIT4.Συμπεράσματα: Η μεμονωμένη χρήση της οκτρεοτίδης και της καβεργολίνης δεν επαρκεί για την αναστολή της κυτταρικής αύξησης αλλά ο συνδυασμός τους με τους αναστολείς mTOR θα μπορούσε να περιορίσει τον μηχανισμό διαφυγής που επάγεται από τους αναστολείς mTOR σε μοντέλο τυπικού καρκινοειδούς πνεύμονα in vitro.
