Η ογκοπρωτεΐνη SET είναι μια εξελικτικά συντηρημένη πρωτεΐνη η οποία εκφράζεται σε όλους τους ιστούς και τους τύπους κυττάρων. Είναι πολυλειτουργική, με ρόλο στην αναδιοργάνωση της χρωματίνης, στη γονιδιακή έκφραση, στη ρύθμιση του κυτταρικού κύκλου, στην απόπτωση και στην κυτταρική μετανάστευση. Χαρακτηριστική είναι η δράση της ως αναστολέας της πρωτεϊνικής φωσφατάσης 2A (ΡΡ2Α). Επιπλέον, εμπλέκεται στην ανάπτυξη της νόσου Alzheimer όπως και διαφόρων τύπων καρκίνου, γεγονός που έχει πυροδοτήσει μελέτες για την αξιοποίησή της ως μόριο- στόχο στη θεραπεία αυτών των ασθενειών. Σκοπός της εργασίας ήταν η διερεύνηση του φυσιολογικού ρόλου της SET σε επίπεδο οργανισμού. Συγκεκριμένα, αξιοποιήθηκε το zebrafish όπου εκφράζονται δύο παράλογες πρωτεΐνες, οι Seta και Setb. Οι Seta/b παρουσιάζουν εξαιρετικά υψηλό ποσοστό ομολογίας τόσο μεταξύ τους όσο και με την ανθρώπινη ορθόλογη. Η ανάλυση του προτύπου έκφρασης των seta/b έδειξε ότι τα μετάγραφα κατανέμονται στο όριο μεσεγκεφάλου-οπίσθιου εγκεφάλου, στο μελάχρουν επιθήλιο του αμφιβληστροειδούς, στη στιβάδα των γαγγλιακών κυττάρων και την ακτινωτή οριακή ζώνη των οφθαλμών, στις αισθητήριες δομές του αυτιού και στους νευρομαστούς της πλευρικής γραμμής. Όμοια αποτελέσματα προήλθαν από πειράματα ανοσοφθορισμού αναδεικνύοντας την έκφραση των γονιδίων set στα αισθητήρια όργανα του zebrafish. Ο λειτουργικός χαρακτηρισμός των γονιδίων έγινε μέσω πειραμάτων καταστολής έκφρασης. Η στόχευση των δύο γονιδίων seta/b μέσω χρήσης ειδικού αντινοηματικού ολιγομερούς (morpholino MOab), προκάλεσε αύξηση της απόπτωσης, μείωση του κυτταρικού πολλαπλασιασμού και οδήγησε στην ανάπτυξη μορφολογικών αλλαγών. Μέσω συνδυαστικής έγχυσης ΜΟab και mRNA του ανθρώπινου γονιδίου SET επετεύχθη μερική ανάκτηση του φαινοτύπου. Αντίστοιχες παρατηρήσεις προήλθαν τόσο από τη χρήση ενός αντινοηματικού ολιγομερούς με διαφορετικό μηχανισμό δράσης (morpholino ΜΟb) όσο και μέσω του συστήματος CRISPR-Cas9. Επιπλέον, η μείωση της έκφρασης των πρωτεϊνών Seta/b οδήγησε σε μείωση του αριθμού των ενεργών νευρομαστών, όπως διαπιστώθηκε έπειτα από in vivo χρώση των morphants με τη φθορίζουσα ουσία FM1-43. Στο ίδιο αποτέλεσμα οδήγησε η καταστολή του setb μέσω έγχυσης του ΜΟb ή του gRNA/Cas9. Η παραπάνω παρατήρηση επιβεβαιώθηκε και μέσω φαρμακολογικής αναστολής των Seta/b, έπειτα από έκθεση των εμβρύων στο φάρμακο FTY720. Συνδυαστικά, μέσω εναλλακτικών μεθόδων, αποδείχθηκε η λειτουργική συσχέτιση των Seta/b με τα κροσσωτά κύτταρα των νευρομαστών. H ανάλυση του προτύπου γονιδιακής έκφρασης των morphants ανέδειξε την ύπαρξη διαφορικά εκφραζόμενων γονιδίων τα οποία, μέσω ρύθμισης της μεταγραφής, επηρεάζουν τη διαφοροποίηση και την ανάπτυξη των οργάνων του αισθητήριου συστήματος. Συμπερασματικά, τα αποτελέσματα που προέκυψαν από τη συγκεκριμένη εργασία, συνδέουν για πρώτη φορά την δράση των γονιδίων seta και setb με την εμβρυογένεση και την ανάπτυξη των οργάνων του αισθητήριου συστήματος.
