Human malaria due to zoonotic transmission has been recorded in the Atlantic Forest, an extra-Amazonian area in Brazil, which are a challenge for malaria control. Naturally acquired humoral immune response against pre-erythrocytic and erythrocytic antigens of Neotropical primates (NP) was evaluated here to improve the knowledge about the exposure of those animals to the malaria transmission and support the identification of the potential reservoirs of the disease in the Atlantic Forest. Blood samples of 154 monkeys from three areas of the Atlantic Forest were used to identify IgG antibodies against peptides of the repeat region of the major pre-erythrocytic antigen, the circumsporozoite protein (CSP), of Plasmodium vivax (PvCSP), Plasmodium brasilianum/Plasmodium malariae (Pb/PmCSP), and Plasmodium falciparum (PfCSP) by ELISA. Antibodies against erythrocytic recombinant antigens of P. vivax, Apical membrane antigen 1 (PvAMA-1), Erythrocyte binding protein 2 (PvEBP-2) and domain II of Duffy binding protein (PvDBPII) were also evaluated. Parameters, such as age, sex, PCR positivity, and captivity, potentially associated with humoral immune response were analyzed. Eighty-five percent of NP had antibodies against at least one CSP peptide, and 76% against at least one P. vivax erythrocytic antigen. A high percentage of adults compared to non-adults were seropositive and showed increased antibody levels. Neotropical primates with PCR positive for P. simium had a significantly higher frequency of positivity rate for immune response against PvEBP-2, PvDBPII and also higher antibody levels against PvDBPII, compared to PCR negative NPs for this species. Monkeys with PCR positive for P. brasilianum/P. malariae showed higher frequency of seropositivity and antibody levels against Pb/PmCSP. Levels of antibodies against Pb/PmCSP, PvEBP-2 and PvDBPII were higher in free-living than in captive monkeys from the same area. All Platyrrhine families showed antibodies against CSP peptides, however not all showed IgG against erythrocytic antigens. These findings showed a high prevalence of naturally acquired antibodies against CSP repeats in all studied areas, suggesting an intense exposure to infected-mosquitoes bites of NP from all families. However, mainly monkeys of Atelidae family showed antibodies against P. vivax erythrocytic antigens, suggesting blood infection, which might serve as potential reservoirs of malaria in the Atlantic Forest.
Satellite DNAs (satDNAs) are major components of eukaryote genomes. However, because of their quick divergence, the evolutionary origin of a given satDNA family can rarely be determined. Herein we took advantage of available primate sequenced genomes to determine the origin of the CapA satDNA (approx. 1500 bp long monomers), first described in the tufted capuchin monkey We show that CapA is an abundant satDNA in Platyrrhini, whereas in the genomes of most eutherian mammals, including humans, this sequence is present only as a single copy located within a large intron of the NOS1AP () gene. Our data suggest that this intronic CapA-like sequence gave rise to the CapA satDNA and we discuss possible mechanisms implicated in this event. This is the first report to our knowledge of a single copy intronic sequence giving origin to a satDNA that reaches up to 100 000 copies in some genomes.
Resumo os primatas são animais que possuem elo social entre a mãe e sua prole, diferente de outras ordens de mamíferos. a sobrevivência do infante primata é completamente dependente do cuidado provido por membros de seu grupo social, particularmente do cuidado materno. o objetivo deste estudo foi analisar a utilização do recinto por um infante de bugio e sua proximidade com os pais. o grupo de bugios era composto por um casal de adultos e seu filhote fêmea com quatro meses de idade. o período de observações foi de agosto a dezembro/2006, perfazendo uma média de 96 horas de esforço de amostragem. o método de observação foi o animal focal com registro instantâneo, com intervalos de 30 segundos durante uma hora por dia. o local do estudo foi o recinto de exposição da Fundação Zoo-Botânica de Belo Horizonte, com 7m de altura, 6 m de largura, 7 m de profundidade com presença de paisagismo interno. as observações revelaram um contato maior do infante com a mãe em relação ao pai e um distanciamento significativo (P < 0,05) do filhote em relação ao contato materno, com o aumento da idade e maior independência. o local mais utilizado durante os três primeiros meses de cativeiro foi a pérgola. No mês de dezembro, período de maior pluviosidade, o filhote aumentou interações ventrais com a mãe, e permaneceu mais tempo no cano. as informações obtidas neste trabalho contribuem para um melhor entendimento em relação aos infantes de Alouatta fusca, suas interações sociais e uso do espaço que podem ser utilizados para aprimorar o manejo ex-situ dos animais, criando melhores condições para a estadia dos mesmos em cativeiro, utilizando-se de estruturas adequadas que simulem o ambiente natural, a fim de garantir o bem-estar dos bugios e assim, a sobrevivência da espécie.
