Nos últimos dez anos, mais de 10.000 trabalhos científicos foram publicados a respeito das HSP (Heat Shock Proteins), ou proteínas do choque térmico, e da resposta ao choque térmico, ou HSR (Heat Shock Response). Muitos outros vieram à luz focalizando outras respostas celulares ao estresse. O conhecimento destes fenômenos básicos é indispensável à compreensão de inúmeras condições patológicas. As aplicações clínicas são potencialmente muito vastas e podem vir a ser fundamentais em diversas doenças, hoje de difícil abordagem, como a síndrome de resposta inflamatória sistêmica, o câncer, as doenças infecciosas e outras. Múltiplas pesquisas estão em andamento, há ainda muito o que se descobrir, mas a relevância do assunto justifica plenamente o seu estudo e a sua divulgação. Resposta ao choque térmico (HSR -Heat Shock Response)Em 1962, descobriu-se que a exposição de célu-las de glândulas salivares de Drosophila busckii a calor, dinitrofenol ou salicilatos 2 produzia o surgimento, em cromossomos, de um novo padrão de espessamento, que representava sítios específicos de transcrição para a síntese de proteínas. O estresse térmico ou químico induzia a expressão de genes, até então quiescentes, os quais faziam com que as células estressadas fabricassem grande quantidade de uma determinada classe de proteí-nas, que foram chamadas de Heat Shock Proteins (HSP), ou proteínas do choque térmico. O processo pelo qual as células respondiam ao estresse ficou conhecido como Heat Shock Response (HSR), ou resposta ao choque térmico.Verificou-se, posteriormente, que a HSR era um fenômeno praticamente universal entre todos os seres vivos 3 . Esta resposta é caracterizada por um aumento extremamente rápido na expressão de um grupo seleto de proteínas, as HSP, o que não ocorre apenas após exposição ao calor, mas também quando as células são expostas a diversos outros desafios metabólicos, incluindo análogos de aminoácidos, diversos metais pesados, agentes que modificam as sulfidrilas protéicas, diversos ionóforos e outros venenos metabólicos 4 . Os agentes anti-reumáticos à base de sais de ouro,como auro-tiomalato de sódio e auranofim, foram capazes de induzir a expressão de HSP em macrófagos murinos cultivados 5 .As HSP são uma classe de proteínas altamente conservadas, desde seres primitivos (procariotas) até o homem, o que é um indício de seu grande valor evolutivo 6 . As HSP podem ser agrupadas em famílias: HSP-27, HSP-47, HSP-60, HSP-70, HSP-90 e HSP-110 3 , de acordo com suas seqüências de aminoácidos e com seus pesos moleculares (em kD -quilodaltons), determinados pelo método SDS-PAGE (eletroforese em gel de poliacrilamidasódio-dodecil-sulfato) 7 . Em cada família, há diferentes proteínas -por exemplo, HSP-72 e HSP-73 no grupo HSP-70; os pesos moleculares são similares, mas os padrões de indução e expressão são distintos. A família HSP-70 é a mais conservada filogeneticamente; sua estrutura, no homem, é 72% homóloga em relação às HSP-70 de drosófilas 8 . Experimental burn models are always needed, being essential for burn research. In the present study, we describe a standard Fischer F-344 rat burn model, in which burns are produced with a heated brass bar. The animals were anesthetized with ketamine (90 mg/kg). After the burns, they were resuscitated with saline. Burns ranging from 26% to 30% of total body surface area resulted in a 62.5 % mortality rate after 25 days. This model is an additional option for those interested in burn research.
Este artigo apresenta um estudo sobre a incidência de suicídios em Luz (MG), verificando as eventuais relações com agrotóxicos. Para tanto, adotou-se estudo descritivo com 50 moradores de uma micro-região, entrevistados mediante questionário. Realizaram-se dosagens de acetilcolinesterase, gama-glutamil-transferase (GGT), transaminase glutâmico-oxalacética (TGO) e transaminase glutâmico-pirúvica (TGP) no soro e pesquisas nos prontuários hospitalares e no cartório do registro civil, estudando-se os casos de suicídios entre os anos de 2000 e 2004. A pesquisa nos prontuários e no cartório permitiu verificar a ocorrência de 8,1 atendimentos/mês de casos de intoxicação por agrotóxicos, sendo 19 suicídios (22,6/100.000 hab./ano) no período. Desses, 18 eram trabalhadores rurais do sexo masculino. O mecanismo de suicídio foi, em 57,9% dos casos, envenenamento com agrotóxicos. Dos entrevistados, 98% relataram usar regularmente agrotóxicos, 72% não utilizaram nenhum equipamento de proteção, 56% nunca leram as bulas e 40% afirmaram ter tido intoxicação. Encontrou-se elevação da TGO e da TGP em 33,33% dos homens e da GGT em 21,42% das mulheres e 13,88% dos homens. Não foi detectada redução da atividade da acetilcolinesterase. Constatou-se que a incidência de suicídios foi mais que o dobro da maior média estadual brasileira e que o número de atendimentos de intoxicações foi alto se comparado aos do Sinitox.
The purpose of this study was to assess the effectiveness of nasopalatine nerve block for anesthesia of maxillary central incisors after failure of the anterior superior alveolar nerve (ASAN) block technique. Secondarily, the possible innervation of the maxillary central incisors by the nasopalatine nerve was also investigated. Twenty-seven healthy, young adult volunteers (age: 17-26 years; gender: 9 males and 18 females) were enrolled in this study. All participants were undergraduate dental students of the University of Vale do Rio Verde de Três Corações. The volunteers had the anterior superior alveolar nerves anesthetized and a thermal sensitivity test (cold) was performed on the maxillary central incisors. The volunteers that responded positively to cold stimulus received a nasopalatine nerve block and the thermal sensitivity test was repeated. All participants were anesthetized by a single operator. Three patients presented sensitivity after both types of bilateral blocks and were excluded from the percentage calculations. In the remaining 24 patients, 16 had their maxillary central incisors anesthetized by the anterior superior alveolar block and 8 remained with sensitivity after the ASAN block. All these 8 patients had their maxillary central incisors successfully anesthetized by the nasopalatine block. In this study, 33.3% of the subjects had the innervation of one or both maxillary central incisors derived from the nasopalatine nerve, whilst most subjects (66.7%) had such teeth innervated by the anterior superior alveolar nerve. The nasopalatine nerve block was effective in anesthetizing the maxillary central incisors when the anterior superior alveolar nerve block failed.
