Ethanol is the most suitable substitute for oil-based fuels. The performance of the fermentation is affected by several factors, therefore the aim of this work was to evaluate the efficiency of the fermentation of a hydrolyzed must of sweet potato using three strains of the Saccharomyces cerevisiae. It was also evaluated the effect of three forms of the processes conduction in the fermentation yield, efficiency and viability of yeast at the end process. Among the parameters evaluated, only the cell viability showed significant difference. The strain PE-2 would be the most suitable for the fermentation of the hydrolysed sweet potato.
Para avaliar o impacto sensorial da adição de extratos de madeira à cachaça foram produzidos extratos de oito diferentes madeiras: amburana (Amburana cearensis), bálsamo (Miroxylom peruiferum), carvalho brasileiro (Panopsis organensis), carvalho europeu (Quercus alba), ipê amarelo (Tabebuia chrysostrichae), ipê roxo (Tabebuia impetiginosa), jequitibá rosa (Cariniana estrellensis) e louro-canela (Aniba parviflora). Utilizou-se cachaça diluída com água a 8% v/v de etanol como solvente e 30 g de madeira. Os extratos obtidos foram diluídos 20 vezes com cachaça não-envelhecida e submetidos ao teste de aceitação com escala hedônica não-estruturada. Obtiveram destaque positivo a cachaça com extrato de bálsamo e a cachaça comercial envelhecida em ipê amarelo. As amostras com extratos de louro-canela e jequitibá rosa apresentaram destaque negativo. Tais resultados indicaram a possibilidade da utilização de extratos de madeira no aprimoramento do processo de envelhecimento da cachaça.
Ethanol production from lignocellulosic biomass is of economic interest due to the pressure to reduce fossil fuels consumption and land use for non-edible crops. Xylose is one of the main sugars obtained by hydrolysis of hemicellulose fraction of biomass, but industrial yeasts cannot ferment it. This work aimed to select, characterize and identify xylose-fermenting yeasts from Brazilian microorganisms collections with potential use in ethanol production. Xylose assimilation was tested by replica plating, and fermentation was tested with Durham tubes. Xylose-fermenting strains had their fermentative capacity quantified and compared to a reference strain (Scheffersomyces stipitis UFMG-IMH 43.2) and were identified by molecular techniques. Three strains isolated from plant exudates were able to ferment xylose and showed fermentative parameters similar to the reference strain. Two strains were identified as Candida parapsilosis and one was identified as Meyerozyma guilliermondii. The findings show the potential biotechnological use of these microorganisms.
O objetivo deste trabalho foi verificar a capacidade lipolítica de linhagens de leveduras isoladas de frutos de diferentes espécies de palmeiras do cerrado Tocantinense em meio sólido contendo Tween 20 nas temperaturas, 25, 35 e 40 ºC avaliando-se o índice enzimático (IE). Avaliou-se também o efeito do pH, agitação, concentração de sulfato de amônio (NH4)2SO4 e extrato de levedura em cultivo submerso para as duas linhagens que apresentaram o maior valor de IE, através de um planejamento de Plackett-Burman (PB), para isso, foi realizado um estudo exploratório, laboratorial de natureza quantitativa. Das 150 linhagens testadas 24 (16%) apresentaram resultados positivos a 25 ºC, 12 (8%) a 35 ºC e nenhuma linhagem apresentou atividade a 40 ºC. Os maiores valores de IE obtidos foram de 6,0 e 5,0 a 25 ºC para as linhagens T194 e T199, respectivamente. Os resultados do PB mostraram que apenas o (NH4)2SO4 apresentou influência significativa para a produção de lipases, com valores variando de 34,3 (T194) e 43,6 U.mL-1 (T199). Foi realizado um delineamento inteiramente casualizado com diferentes níveis de concentração de (NH4)2SO4 e observou-se que não houve efeito significativo para a linhagem T194, porém as concentrações de 30 e 35 g.L-1 apresentaram efeito significativo para a linhagem T199, as atividades variaram de 61,0 e 70,1 U.mL-1. A adição de o sulfato de amônioao meio de cultura elevou a produção de lipase da linhagem T194 de 34,3 para 60 U.mL-1 e de 43,6 para 70,1 U.mL-1 para a linhagem T199.
Etanol pode ser produzido a partir de diversas fontes amiláceas, bem como dos resíduos oriundos das mesmas. Assim, em vista da variedade de matéria-prima para produção de etanol, faz-se necessário estudar e selecionar linhagens de microrganismos selvagens que demonstrem a capacidade de hidrolisar o amido em açúcares fermentescíveis. Desta forma, o presente estudo consiste em investigar a influência de parâmetros de cultivo sobre a atividade de amilases, produzidas por leveduras isoladas do processamento de farinha de puba. O delineamento de Plackett e Burman (PB) foi aplicado com o intuito de selecionar e avaliar a influência da temperatura, do pH inicial e das concentrações de inóculo, amido, peptona e extrato de levedura sobre a atividade enzimática. Por meio dos resultados para tal delineamento, verificou-se influência estatisticamente significativa (p < 0,10) da temperatura e das concentrações de amido e inóculo. Em relação aos resultados do Delineamento Composto Central Rotacional (DCCR), observou-se que o efeito linear da temperatura e da concentração de amido foram significativos (p < 0,05) sobre a atividade enzimática. Com a metodologia de superfície de resposta observou-se que a condição ótima para atividade da enzima amilase é obtida quando se aumenta a temperatura e a concentração de amido do meio. Assim, os resultados obtidos no presente estudo sugerem que as amilases produzidas pela linhagem PB-34 apresentam potencial para estudos futuros visando a produção de etanol a partir de materiais amiláceos.
