-The objective of this work was to characterize morphologically and molecularly the genetic diversity of cassava accessions, collected from different regions in Brazil. A descriptive analysis was made for 12 morphological traits in 419 accessions. Data was transformed into binary data for cluster analysis and analysis of molecular variance. A higher proportion of white or cream (71%) root cortex color was found, while flesh colors were predominantly white (49%) and cream (42%). Four accession groups were classified by the cluster analysis, but they were not grouped according to their origin, which indicates that diversity is not structured in space. The variation was greater within regions (95.6%). Sixty genotypes were also evaluated using 14 polymorphic microsatellite markers. Molecular results corroborated the morphological ones, showing the same random distribution of genotypes, with no grouping according to origin. Diversity indices were high for each region, and a greater diversity was found within regions, with: a mean number of alleles per locus of 3.530; observed and expected heterozygosity of 0.499 and 0.642, respectively; and Shannon index of 1.03. The absence of spatial structure among cassava genotypes according to their origins shows the anthropic influence in the distribution and movement of germplasm, both within and among regions.Index terms: Manihot esculenta, genetic diversity, germplasm, microsatellites, morphological traits. Diversidade morfológica e molecular entre genótipos de mandiocaResumo -O objetivo deste trabalho foi caracterizar, morfológica e molecularmente, a diversidade genética de acessos de mandioca de diferentes regiões do Brasil. Realizou-se a análise descritiva de 12 características morfológicas em 419 acessos. Os dados foram transformados em dados binários para análise de agrupamento e análise de variância molecular. Observou-se maior proporção (71%) de cor branca ou creme do córtex da raiz, enquanto as cores da polpa foram predominantemente branca (49%) e creme (42%). Quatro grupos de acessos foram classificados pela análise de agrupamento, mas não foram agrupados de acordo com sua origem, o que indica que a diversidade não está estruturada no espaço. A maior parte da variação ocorreu dentro das populações (95,6%). Sessenta genótipos também foram avaliados com o uso de 14 marcadores microssatélites polimórficos. Os resultados moleculares corroboraram os morfológicos e mostraram a mesma distribuição aleatória dos genótipos, com ausência de agrupamento quanto ao local de origem. Os índices de diversidade foram elevados para cada região e a maior parte da diversidade foi observada dentro de regiões, com: número médio de alelos por loco de 3,530; heterozigosidade média observada de 0,499 e esperada de 0,642; e índice de Shannon de 1,03. A ausência de estrutura espacial entre os genótipos de mandioca de acordo com suas origens mostra a influência antrópica na distribuição e movimentação de germoplasma, tanto entre como dentro de regiões.
Outro fator que deve ser levado em consideração na seleção de genótipos destinados à mesa é a qualidade culinária das raízes. Existem diversos fatores de qualidade que devem ser considerados, mas os de maior importância são a textura, a plasticidade e a pegajosidade da massa cozida (PEREIRA et al., 1985). Existe uma forte correlação entre o tempo de cozimento e essas características, de modo que, quanto menor o tempo de cozimento, melhor a massa cozida. Portanto, a simples determinação do tempo de cozimento é uma segura avaliação indireta da qualidade de massa cozida (PEREIRA et al., 1985; FUKUDA e BORGES, 1988;LORENZI, 1994).As carências alimentares existentes em populações pobres ou mal nutridas causam problemas de saúde pública em todo o mundo, principalmente em países pobres e em desenvolvimento. Assim, atualmente, existe uma preocupação por parte dos melhoristas em obter novas variedades que agreguem alto valor nutricional (WELCH e GRAHAM, 2002).Em países subdesenvolvidos onde a mandioca é a base alimentícia, variedades ricas em nutrientes teriam grande vantagem, tanto pelo baixo custo de produção da cultura, quanto pelo suprimento das necessidades nutricionais da população.As deficiências de micronutrientes, em especial de vitamina A, têm merecido destaque, devido ao número crescente de evidências que comprovam seu impacto negativo na saúde de grupos de maior vulnerabilidade nutricional como: gestantes, recém-nascidos e crianças na idade pré-escolar (RAMALHO et al., 2002). A obtenção de vitamina A na alimentação humana ocorre por duas maneiras: por meio do consumo de alimentos de origem animal cuja vitamina A já está pré-formada (os retinóides) e pelo consumo de alimentos de origem vegetal nos quais são constatados carotenóides pró-vitamínicos A (RONCADA, 1998). INTRODUÇÃOA mandioca (Manihot esculenta Crantz) é uma das culturas mais importantes na alimentação humana dos trópicos, principalmente para as populações de baixa renda. A produção mundial foi estimada pela FAO (2005), em 210 milhões de toneladas. Apenas uma pequena parte é utilizada na alimentação animal ou como matéria-prima para produtos industrializados que não se destinam à alimentação humana. No Brasil, a mandioca integra o cardápio em todas as regiões brasileiras, desde o período pré-colonial até os dias atuais. É consumida na forma de farinha de mandioca ou de derivados de amido ou, ainda, as raízes são processadas no âmbito doméstico e consumidas cozidas, fritas ou utilizadas para preparar pratos típicos. Neste caso, são chamadas de mandioca de mesa ou de uso culinário.Variedades de mandioca de mesa destinadas à comercialização das raízes, além de bom desempenho agrícola exigido pelo produtor, como boa produtividade, resistência a pragas e doenças, uniformidade e padrão comercial das raízes e facilidade de práticas culturais, também devem atender às exigências sensoriais e tecnológicas do consumidor final, demonstrando sabor característico, textura macia e cozimento rápido. ABSTRACT. Dioscorea cayenensis and Dioscorea rotundata are among the most important yam species for the humid and sub-humid tropics. We isolated nine polymorphic microsatellite markers using a microsatelliteenriched genomic library technique. The nine primer pairs were validated in 22 D. cayenensis accessions, and were tested for transferability in 26 D. rotundata accessions. The number of bands ranged from 2 to 4, with a mean of 3.11. D. cayenensis gave primer polymorphism information content values ranging from 0.37 to 0.62, while for D. rotundata the values ranged from 0.15 to 0.66. The D parameter in D. cayenensis ranged from 0.14 to 0.40, while in D. rotundata it ranged from 0.05 to 0.34. These SSR markers will be useful to characterize genetic diversity in D. cayenensis and D. rotundata accessions.
