Abstract. Pimenta pseudocaryophyllus is a Brazilian native plant that presents high concentrations of flavonoids and other polyphenolic compounds. Herein, we evaluated: (1) the chemical properties of P. pseudocaryophyllus ethanolic extract (PPE), (2) the in vitro antioxidant activity (AA) of PPE and of two different topical formulations (F1 and F2) containing PPE, (3) physico-chemical and functional stability, (4) in vitro release of PPE, and (5) in vivo capacity of formulations to prevent UV-B irradiation-induced skin damage. Results show that the polyphenol and flavonoid contents in PPE were 199.33 and 28.32 mg/g, respectively, and HPLC results show the presence of eugenol, tannic acid, and rutin. Evaluation of the in vitro AA of PPE demonstrated a dose-dependent effect and an IC 50 of 4.75 μg/mL in 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 3.0 μg/mL in 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays. The ferric-reducing antioxidant power (FRAP assay) was 0.046 μmol/L trolox equivalent/ μg/mL of extract. Among the AA, only the capacity to scavenge DPPH radical of PPE was maintained in F1 and F2. In addition, both formulations satisfactorily released the extract. The evaluation of the functional stability of F1 and F2 did not demonstrate loss of activity by storage at room temperature and at 4°C/6 months. In irradiated mice, treatment with F1 and F2 added with PPE significantly increased the capacity to scavenge ABTS radical and the FRAP of skin compared to vehicle-treated mice. In conclusion, the present results suggest that formulations containing PPE may be a topical source of antioxidant compounds to decrease oxidative damages of the skin.
An investigation of antifungal activity of the essential oil obtained by steam-distillation (1.1% w/w) of the aerial parts of Ocimum gratissimum and of an ethanolic extract from the steam-distillation residue was carried out using the agar diffusion method. The results revealed that the essential oil inhibited the growth of all fungi tested, including the phytopathogens, Botryosphaeria rhodina, Rhizoctonia sp. and two strains of Alternaria sp., while the extract from the residue was inactive. The essential oil was subjected to TLC bioautography used to detect fungitoxic constituents. The compound that showed antifungal activity was isolated and identified as eugenol. GC/MS analysis showed that eugenol was the main constituent of the essential oil studied. The antifungal activity of eugenol was evaluated against a species of Alternaria isolated from tomato (A 1 ) and Penicillium chrysogenum. The minimal inhibitory concentrations of eugenol were 0.16 and 0.31 mg/disc for Alternaria sp. (A 1 ) and P. chrysogenum, respectively.
RESUMOO picão-preto (Bidens pilosa L.) é uma planta daninha muito agressiva, que está presente em quase todo Brasil. O principal método de controle é o químico, porém apresenta elevado impacto ambiental, risco de intoxicação humana e possibilidade de causar fitotoxicidade as culturas. O objetivo do trabalho foi avaliar o efeito alelopático de extratos etanólicos de Eucalyptus citriodora Hook. e Pinus elliottii L. na germinação e no crescimento inicial de picão-preto e alface (Lactuca sativa L.). Foram testadas quatro concentrações de cada extrato (0,25; 0,50; 1,0 e 2,0 %) além do controle (0,0 %) água destilada com Tween 20 a 0,08 %. O delineamento experimental foi o inteiramente casualizado, com quatro repetições em condições de laboratório. O extrato de P. elliottii não causou efeito alelopático sobre o picão-preto e o alface. O extrato de E. citriodora reduziu significativamente o índice de velocidade de germinação (IVG) do picão-preto em todas as concentrações testadas quando comparadas com o controle (0,0%), porém para a alface o IVG foi significativo apenas na concentração de 2,0 %. Para o comprimento da raiz não foi possível observar diferença significativa entre os tratamentos para os dois extratos testados tanto para o alface como para o picão preto. The main control method for this species is chemical treatment however, causes strong environmental impact, and it has great human contamination risks, and may cause phytotoxity to crops. The objective of this study was to evaluate the effect of ethanolic extracts of Eucalyptus citriodora Hook. and Pinus elliottii L. on seed germination and initial growth of B. pilosa and lettuce (Lactuca sativa L.). Five concentrations of each extract (0.0; 0.25; 0.50; 1.0; 2.0%) were tested in laboratory conditions using a randomized complete block design with four replicates. P. elliottii extract had no effect on B. pilosa and lettuce. However, E. citriodora extract, significantly reduced germination index (GI) of B. pilosa, in all tested concentrations, when compared with the control composed by distilled water. Lettuce GI was affected only by 2,0% concentration of ethanolic extract. Extracts of both P. elliottii and E. citriodora had no significant effect on the root length parameter of both B. pilosa and lettuce. Phytochemical investigation of the flowers of Wedelia paludosa afforded two diterpenes, ent-kaur-16-en-19-oic acid and ent-kaur-9(11),16-dien-19-oic acid, three acylated triterpenes, 3β-O-hexa-decanoylolean-12-en-28-oic acid, 3β-O-pentadecanoylolean-12-en-28-oic acid and 3β-O-tetradecanoylolean-12-en-28-oic acid, and three steroids, stigmasterol, 3β-O-β-D-glycopyranosyl sitosterol and 3β-O-β-D-glycopyranosylstigmasterol. INTRODUÇÃOA família Asteraceae compreende cerca de 25.000 espécies distribuídas em aproximadamente 1.100 gêneros, sendo representada no Brasil por cerca 180 gêneros 1 . Esta família aparece com um número relativamente grande de citações bibliográficas, envolvendo farmacognosia, fitoquímica e plantas medicinais 2 . As atividades farmacológicas reveladas pelos extratos ou substâncias isoladas de espécies do gênero Wedelia despertaram o interesse para estudo químico 3 . A espécie Wedelia paludosa D. C., usada na medicina popular como expectorante e anticonvulsivo, foi estudada anteriormente após a verificação de atividade antibiótica revelada por testes preliminares envolvendo um extrato bruto das partes aéreas. O estudo químico desse extrato revelou a presença de sete ácidos caurenóicos, um labdano e uma seco-caurenolactona 4 . Recentemente, foi observada também atividade inseticida em ensaios com esta espécie e uma reinvestigação fitoquímica conduziu ao isolamento e caracterização de duas novas lactonas sesquiterpênicas 5 . Este artigo descreve os resultados obtidos do estudo quími-co das flores de um espécimen de Wedelia paludosa. Identificaram-se dois ácidos caurenóicos (1 e 2), estigmasterol (3), glicosídios de estigmasterol (5) e sitosterol (6) e três ésteres derivados do ácido oleanóico (4a-4c). As substâncias 2, 4, 5 e 6 estão sendo registradas pela primeira vez nesta espécie. Os dados de RMN 13 C de 2 estão sendo registrados pela primeira vez na literatura. RESULTADOS E DISCUSSÃOO fracionamento cromatográfico do extrato etéreo das flores de Wedelia paludosa e a análise espectrométrica das frações permitiu a identificação de dois ácidos caurânicos (1 e 2), estigmasterol (3), uma mistura de 3β-O-β-D-glicopiranosilsitosterol (5) e 3β-O-β-D-glicopiranosilestigmasterol (6) e uma mistura de três ésteres 3β-O-aciloleanóicos (4a, 4b e 4c).
