<p>ABSTRAK</p><p>Penggunaan bakteri endofit sebagai agen pengendalian nematodaparasit seperti Meloidogyne incognita pada kapas dan tomat, Globoderasp. pada kentang dan Radopholus similis pada pisang telah banyak ditelitipada beberapa jenis tanaman. Penelitian bertujuan untuk menganalisiskeefektifan beberapa bakteri endofit terhadap perkembangan P.brachyurus, penetrasi, reproduksi, dan kerusakan yang diakibatkannyapada tanaman nilam. Penelitian dilakukan di Laboratorium dan Rumahkaca Hama dan Penyakit Balai Penelitian Tanaman Obat dan AromatikBogor, dari bulan Maret sampai Agustus 2008. Penelitian menggunakanrancangan acak lengkap (RAL) 6 perlakuan dengan 7 ulangan. Lima isolatbakteri endofit, yaitu Achromobacter xylosoxidans TT2, Alcaligenesfaecalis NJ16, Pseudomonas putida EH11, Bacillus cereus MSK, danBacillus subtilis NJ57, diaplikasikan pada setek tanaman nilam denganmetode perendaman akar. Seminggu setelah tanam, nilam diinokulasidengan 500 ekor P. brachyurus. Pengamatan dilakukan terhadap penetrasi,reproduksi, populasi nematoda, dan pertumbuhan nilam. Hasil penelitianmenunjukkan bahwa bakteri endofit A. xylosoxidans TT2, A. faecalisNJ16, P. putida EH11, B. cereus MSK, dan B. subtilis NJ57 dapatmenekan penetrasi dan populasi P. brachyurus ke dalam akar sebesar54,8-70,6% dengan faktor reproduksi (pf/pi) 0,61-0,94 dan meningkatkanpertumbuhan tanaman nilam sebesar 37,86-84,71%.</p><p>Kata kunci: Pogostemon cablin, bakteri endofit, keefektifan, nematoda,Pratylenchus brachyurus, pengendalian biologi</p><p>ABSTRACT</p><p>Effectiveness of endophytic bacteria to controlPratylenchus brachyurus nematode on patchouli</p><p>The use of endophytic bacteria as biocontrol agents for nematodes,such as Meloidogyne incognita on cotton and tomatoes, Globodera sp. onpotatoes and Radopholus similis on bananas has been widely studied inseveral crops. The aim of the study was to investigate the effectiveness ofsome endophytic bacteria to control P. brachyurus, penetration,reproduction, and plant fresh weight production. Five isolates, namely A.xylosoxidans TT2, A. faecalis NJ16, P. putida EH11, B. cereus MSK, andB. subtilis NJ57 were applied to the patchouli cutting roots by soakingmethod before planting. A week after planting, the plants were inoculatedwith 500 juveniles and adults of P. brachyurus. Observations were doneon penetration and reproduction rates of the nematode, and growth ofpatchouli plant. Under greenhouse condition, A. xylosoxidans TT2, A.faecalis NJ16, P. putida EH11, B. cereus MSK, and B. subtilis NJ57reduced penetration rate of P. brachyurus into the patchouli roots by 54.8to 70.6% and suppressed nematode population with pf/pi value 0.61 to0.94. ABSTRAKMeloidogyne spp. merupakan salah satu penyebab penurunan produksi wortel di Indonesia. Nematoda ini telah dilaporkan menjadi penyebab penyakit umbi bercabang di beberapa sentra produksi wortel di Pulau Jawa. Penelitian ini bertujuan mendeteksi dan mengidentifikasi karakter morfologi dan molekuler spesies Meloidogyne pada tanaman wortel asal Dataran Tinggi Malino, Kecamatan Tinggimoncong, Kabupaten Gowa, Sulawesi Selatan. Identifikasi morfologi dilakukan dengan pengamatan karakter pola perineal nematoda betina. Identifikasi molekuler dilakukan dengan teknik polymerase chain reaction (PCR) untuk mengamplifikasi DNA ribosom menggunakan primer spesifik spesies (Rjav/Fjav untuk M.arenaria ditemukan berasosiasi dengan kejadian penyakit umbi bercabang. PCR menggunakan primer spesifik berhasil mengamplifikasi pita DNA M. incognita dan M. arenaria dengan ukuran berturut-turut ± 999 pb dan ± 420 pb, sedangkan PCR dengan primer multipleks tidak berhasil mengamplifikasi pita DNA. Analisis sikuen nukleotida menunjukkan isolat M. incognita asal Malino memiliki kekerabatan yang dekat dengan isolat asal Bangka-Indonesia, Cina (isolat JS2), dan Malaysia (isolat JIK4, FIK4, JIT19, dan FIT19) dengan nilai homologi berkisar 99.2-100.0%. Sikuen nukleotida M. arenaria asal Malino-Indonesia telah didaftarkan pada GenBank dengan nomor aksesi KP234264 dan menjadi data pertama yang tersedia di GenBank. This research aimed to use morphological and molecular characters to detect and identify Meloidogyne species on carrot from Malino Highland, Sub-district of Tinggimoncong, District of Gowa, South Sulawesi. Morphological identification was done based on character of the female perineal pattern. Molecular identification was based on amplification of r-DNA by polymerase chain reaction technique using species specific primers (Fjav/ Rjav for M. javanica, Far/Rar for M. arenaria, and Finc/Rinc for M. incognita) and multiplex primer (JMV1/JMVhapla/JMV2 for M. hapla, M. chitwoodi, and M. fallax).Two of Meloidogyne species, i.e. M. incognita and M. arenaria were detected associated with the incidence of carrot branched tuber. The specific primers amplified two DNA bands, i.e. ± 999 bp of M. incognita and ± 420 bp of M. arenaria, while multiplex primer was failed to amplify DNA bands. Nematoda puru akar (NPA), Meloidogyne spp., telah dilaporkan menjadi penyebab umbi bercabang pada wortel di beberapa sentra produksi sayuran di Jawa Tengah dan Jawa Timur. Pada penelitian ini dilakukan identifikasi secara molekuler spesies NPA yang menginfeksi wortel di Agropolitan Cianjur, Jawa Barat dan penelusuran tingkat kekerabatannya dengan spesies NPA dari negara lain. DNA diekstraksi dari nematoda betina dan diamplifikasi melalui polymerase chain reaction (PCR) menggunakan primer spesifik spesies (Rjav/Fjav untuk M. javanica, Rar/Far untuk M. arenaria dan Rinc/Finc untuk M. incognita) dan primer multipleks (M. hapla, M. chitwoodi, dan M. fallax). Sikuensing nukleotida dilakukan terhadap DNA hasil PCR tanpa melalui proses kloning. Berdasarkan hasil identifikasi ada dua spesies NPA berasosiasi dengan penyakit umbi bercabang, yaitu M. javanica dan M. hapla. Analisis pohon filogenesis memperlihatkan bahwa M. javanica asal Cianjur berkerabat sangat dekat dengan M. javanica asal Cina dengan tingkat homologi mencapai 91.9%. Nematoda M. hapla asal Cianjur mempunyai sikuen nukleotida dengan tingkat homologi antara 99.4% sampai 100% dengan isolat-isolat dari Swiss, Amerika, Inggris, dan Cina.
ABSTRAKNematoda puru akar (NPA) Meloidogyne spp. merupakan parasit utama tanaman seledri (Apium graveolens) yang menimbulkan kerugian. Identifikasi spesies Meloidogyne pada seledri di Indonesia belum pernah dilaporkan. Penelitian ini bertujuan mengidentifikasi spesies Meloidogyne pada seledri secara morfologi. Contoh tanaman seledri yang terinfeksi oleh Meloidogyne spp. diambil dengan metode purposif di Desa Ciputri, Kecamatan Pacet, Kabupaten Cianjur, Provinsi Jawa Barat. Nematoda di dalam jaringan akar dideteksi dengan metode pewarnaan asam fuksin. Identifikasi spesies Meloidogyne dilakukan dengan pengamatan morfologi pola perineal betina. Gejala penyakit puru akar pada seledri ialah daun menguning, kerdil, dan pertumbuhan tanaman di lahan tidak merata. Gejala pada perakaran berupa puru akar berukuran kecil-kecil, jumlahnya banyak dan membentuk untaian seperti rantai. Berbagai stadium perkembangan nematoda, yaitu telur, juvenil, dan nematoda betina berhasil dideteksi melalui pewarnaan pada jaringan akar. Tiga spesies Meloidogyne diidentifikasi sebagai M. incognita, M. arenaria, dan M. javanica.Kata kunci: identifikasi morfologi, jaringan akar, pewarnaan asam fuksin, pola perineal betina ABSTRACT Root knot nematodes (RKN) Meloidogyne spp. is the primary parasite of celery and reported to cause losses up to 70%. Identification of the Meloidogyne species on celery in Indonesia has not been reported. This study was aimed to identify the species of Meloidogyne on celery based on morphology characters. Samples of Meloidogyne-infected celery plants were taken using purposive sampling method from Ciputri Village, District of Pacet, Cianjur, West Java Province. RKN inside root tissue was detected by acid fuchsin staining method. Meloidogyne species identification was done by morphological observation of female perineal pattern. Disease symptoms found in the field include leaf yellowing, stunted, and uneven growth of celery plants. Roots of infected plants showed the formation of small size root knots, in large numbers and forming strands like a chain. Staining NPA in root tissue was successfully detected various stages of nematode development, i.e. eggs, juveniles and female nematodes. Three Meloidogyne species, namely M. incognita, M. arenaria and M. javanica were identified.
ABSTRAKNematoda puru akar (Meloidogyne spp.) merupakan salah satu nematoda yang terdapat pada tanaman kentang di daerah tropik dan subtropik. Nematoda ini memberikan dampak yang nyata dalam mengurangi kualitas dan kuantitas umbi kentang. Nematoda ini mempunyai beberapa spesies, salah satunya Meloidogyne incognita. Penelitian bertujuan mendeteksi dan mengidentifikasi spesies M. incognita pada kentang dan hubungan kekerabatannya dengan spesies dari negara lain. Umbi kentang bergejala bintil dikumpulkan dari Pangalengan (Jawa Barat), Banjarnegara (Jawa Tengah), dan Kota Batu (Jawa Timur). Identifikasi dilakukan berdasarkan pola perineal nematoda betina dan karakter molekul DNA dengan teknik polymerase chain reaction menggunakan sepasang primer spesifik (MI-F dan MI-R) dan dilanjutkan dengan sikuensing fragmen DNA dan analisis filogenetika. Dari 3 lokasi pengamatan berhasil diidentifikasi M. incognita. Nematoda asal Pangalengan memiliki tingkat homologi 99.2% hingga 99.8% dengan nematoda asal Cina, India, dan Malaysia.Kata kunci: analisis filogenetika, deteksi, polymerase chain reaction ABSTRACT Root knot nematodes is an important pathogen on potatoes in tropical and sub-tropical areas. Root knot nematodes contribute a significant impact in reducing the quality and quantity of potato tuber. Meloidogyne incognita is one of the species causing the root knot. This research was conducted to identify M. incognita on potatoes in Java island based on morphological and DNA molecular characteristic. The infected potato tubers with pimple-like knot symptom were collected from Pangalengan (West Java), Banjarnegara (Central Java), and Kota Batu (East Java). Nematode was identified based on morphological character of perineal pattern of female nematodes, and molecular DNA character by polymerase chain reaction technique using a pair of specific primer (MI-F and MI-R), followed by DNA fragment sequencing and phylogenetic analysis. Based on morphological character of perineal pattern, M. incognita was detected in all 3 locations; while based on DNA molecular character, M. incognita was detected in Pangalengan (West Java) and Kota Batu (East Java). M. incognita from Pangalengan had high homology, i.e.99.2% to 99.8% with those isolates from China, India, and Malaysia.
