The present study was to determine the effects of salinity on the growth and nutrient bioextraction abilities of Gracilaria and Ulva species, and to determine if these seaweeds can be used for nutrient bioextraction under hypo-and / or hyperosmotic conditions. Two Gracilaria species, G. chorda and G. vermiculophylla, and two Ulva species, U. prolifera and U. compressa, were cultured at various salinity conditions (5, 10, 15, 20, 30, 40, and 50 psu) for 3 weeks. Results showed that the growth rates, nutrient uptake, tissue nutrient contents and nutrient removal were significantly affected by salinity and species. All four species were euryhaline with the highest growth rates at 20 psu. Among the four species, U. prolifera, U. compressa, and G. vermiculophylla showed potential to be used for nutrient bioextraction in estuaries and / or land-based fish farms due to their rapid growth, high nutrient uptake, high tissue carbon and nitrogen accumulation and removal capacities.

Although Korea is third in seaweed production and second in shellfish production globally, this is the first study evaluating ecosystem services of seaweed and shellfish aquaculture in Korea. The objective of this study is to evaluate nutrient bioextraction capacities of major seaweed and shellfish species aquacultured in Korea. C (C) removal of three major aquacultured seaweed species, Neopyropia yezoensis, Saccharina japonica, and Undaria pinnatifida were 24,247, 8,423, and 12,758 tons, respectively, in 2016. N (N) removal of these species was 4,088, 732, and 1,244 tons, respectively. The C and N removal of the Pacific oysters (Crassostrea gigas) were 14,693 and 1,050 tons, respectively. Manila clams (Venerupis philippinarum) removed 2,120 tons of C and 136.5 tons of N. Together, 161,846 tons of CO 2 and 7,251 tons of N were removed by three major seaweed species and two shellfish species. These values are significant amounts, equivalent to 5.7% of CO 2 and 8.6% of N discharged from all wastewater treatment plants in Korea. These results suggest that nutrient bioextraction by aquacultured seaweed and shellfish can be a cost efficient, affordable, and equitable solution for coastal nutrient management programs in Korea and elsewhere.

A rapid, simple, and cost‐effective total DNA extraction method referred to as a direct boiling method for PCR‐based algal species determination was validated using representative species of green, brown, and red algae. Each sample was briefly minced in two buffers, Tris‐EDTA (TE) or PCR buffer, and transferred to a heat block for boiling at 98°C for 5 min. No detergent was used in this experiment. The entire DNA isolation procedure was completed within 10 min. After brief centrifugation, the supernatant was directly used as a template for PCR. As a result, all genomic DNA markers were successfully amplified and sequenced from each algal taxon. Regardless of DNA quality, the direct boiling method is very suitable to identify unknown species of algae from a large amount of samples in a limited time and can be applied broadly to routine seasonal or annual monitoring of algae. DNA from an Undaria pinnatifida sporophyte, however, was not successfully extracted by this direct boiling method, probably due to a high concentration of polysaccharides.
