Since concomitant release of structurally related peptide hormones with apparently similar functions seems to be a general concept in endocrinology, we have studied the dynamics of the lifetime of the three known adipokinetic hormones (AKHs) of the migratory locust, which control flight-directed mobilization of carbohydrate and lipid from fat body stores. Although the structure of the first member of the AKHs has been known for 20 years, until now, reliable data on their inactivation and removal from the hemolymph are lacking, because measurement requires AKHs with high specific radioactivity. Employing tritiated AKHs with high specific radioactivity, obtained by catalytic reduction with tritium gas of the dehydroLeu2 analogues of the AKHs synthesized by the solid-phase procedure, studies with physiological doses of as low as 1.0 pmol per locust could be conducted. The AKHs appear to be transported in the hemolymph in their free forms and not associated with a carrier protein, despite their strong hydrophobicity. Application of AKHs in their free form in in vivo and in vitro studies therefore now has been justified. We have studied the degradation of the three AKHs during rest and flight. The first cleavage step by an endopeptidase is crucial, since the resulting degradation -products lack any adipokinetic activity. Half-lives for AKH-I, -II and -m were 51, 40, and 5 min, respectively, for rest conditions and 35, 37, and 3 min, respectively, during flight.
Zusammenfassung Es wurde eine multiple Regressionsanalyse mit 36 Tieren aus Experimenten, die in den ersten zwei Mitteilungen beschrieben wurden, für die Beziehung zwischen einzelnen Metaboliten‐gehalten (Y) in Leber (L) und Fettgewebe (F) zur Aufnahme von Rohprotein (X1), Umsetz‐barer Energie (X2) und zum Metabolischen Körpergewicht W0,75 (X3) durchgeführt. Diese Regressionskoeffizienten sind in den Tabellen 2 und 3 angegeben und mit diesen Koeffizien‐ten berechnete Metabolitengehalte als Mittelwerte entsprechend den Tiergruppen A, B, C, D aus dem ersten und L, X aus dem zweiten Experiment mit der (Curve Nr. 2 in den Abbildungen dargestellt. Fast alle angegebenen multiplen Korrelationskoeffizienten sind stati‐stisch signifikant, mit Ausnahme von PEP in der Leber und AGP und CIT im Fettgewebe. Für die 20 Tiere aus dem ersten Experiment wurde noch die Beziehung der Metaboliten‐gehalte (Y) zu der täglichen Aufnahme an verdaulichem Rohprotein (X1), Verdaulicher Energie (X2) und W0,75 (X3), oder zu täglichem Proteinzuwachs (X1), täglichem Fettzu‐waehs (X2) und W0,75(X3) analysiert. Die zuerst genannte Beziehung ist mit Hilfe der Kurve Nr. 4 dargestellt worden und die zuletzt genannte Beziehung mit Hilfe der Kurve Nr. 3. Kurve Nr. 1 in den Abbildungen entspricht den experimentell bestimmten Metabolitenge‐halten, die in den ersten zwei Mitteilungen tabelliert wurden. Es konnten folgende Einflüsse der Aufnahme von Rohnährstoffen auf die Metaboliten‐gehalte in der Leber und im Fettgewebe festgestellt werden, wobei die Reihenfolge der Metabo‐liten von einem mehr einseitigen Einfluß eines Faktors zu einem mehr kombinierten Einfluß beider Faktoren dargestellt wurde: In der Leber zeigte die Rohproteinaufnahme einen posi‐tiven Einfluß auf die Gehalte von AGP, ACCO, LACT, MAL und CIT und einen negativen Einfluß auf die Gehalte von ASP, GLUT, 2‐PG und PEP; die Aufnahme der Umsetzbaren Energie zeigte einen positiven Einfluß auf die Gehalte von PYR, ACAC, GLUC und ASPN und einen negativen Einfluß auf die Gehalte von BHBA und AKG. In dem Fettgewebe hatte die Rohproteinaufnahme einen positiven Einfluß auf die Gehalte von CIT, AGP, ACAC und ACCO und einen negativen Einfluß auf die Gehalte von AKG, ASP, GLUT und ASPN; die Aufnahme der Umsetzbaren Energie zeigte einen positiven Einfluß auf die Gehalte von PEP und 2‐PG und einen negativen Einfluß auf die Gehalte von BHBA, GLUC, PYR, MAL und LACT. An Hand der Beziehung von Metaboliten zur Aufnahme an Rohprotein, Umsetzbarer Energie und zum Metabolischen Körpergewicht konnten Verbesserungen der multiplen Korrelationskoeffizienten demonstriert werden (Tabelle 4 und 5). Bei einigen Metaboliten wurden diese Koeffizienten durch Dividieren der Metabolitengehalte durch W0,75, bei ande‐ren Metaboliten durch Multiplizieren dieser mit dem Gesamtgewicht von Leber oder Kör‐perfett verbessert. Die in dieser Veröffentlichung nicht angegebenen Regressionskoeffizienten können auf Anfrage zugeschickt werden.
Zusammenfassung In einem Experiment mit 20 Schweinen der Deutschen Landrasse, die in vier Gruppen (A, B, C, D) mit einem Rohproteinanteil von 14,7 bis 32,6% in der Futterration aufgeteilt und von einem Anfangsgewicht von 25 kg bis zu einem Endgewicht von 50 bis 90 kg ad libitum gefüt‐tert wurden, wurden ein Verdauungsversuch mit Chromoxid und eine chemische Körper‐analyse durchgeführt. Nach den sogenannten Frierstoppverfahren wurden den durch Schuß betäubten Tieren vor dem Töten Leber‐ und Fettgewebeproben zur Bestimmung von 15 Metaboliten entnommen. Die Futterrationen B (23,4% Rohprotein) und C (18,4% Rohprotein) stellen normale Futterrationen dar. Die mehr extremen Futterrationen A (32,6% Rohprotein) und D (14,7% Rohprotein) bewirkten eine geringere Futteraufnahine und geringere Gewichtszu‐nahme. Die Tiere der Futtergruppe A unterscheiden sich von den Tieren der Futtergruppe B und C hinsichtlich Gewichtszunahme, Fettzuwachs, täglichen Aufnahme von verdaulicher Energie, und im Proteinzuwachs signifikant. Im Proteinzuwachs besteht auch ein signifi‐kanter Unterschied zwischen Futtergruppe B und D. Die täglichen Aufnahmen von Rohprotein und verdaulichem Rohprotein sind beim Vergleich aller vier Gruppen miteinander signifikant verschieden. Hochsignifikant (P < 0,001) sind die multiplen Korrelationskoeffizienten entsprechend der Berechnung der verdaulichen Energie (VE) aus Proteinzuwachs (RP), Fettzuwachs (RF) und Metabolischem Körpergewicht (W0,75), sowie auch für die Berechnung von RF aus verdaulichem Rohprotein (VP), VE und eventuell aus W0,75. Berechnung von RP aus VP, VE und eventuell aus W0,75 ergab auch einen signifikanten(P < 0,01) multiplen Korrelationskoeffizienten, dagegen war die Berechnung von VP aus RP, RF und W0,75 nicht signifikant. Obwohl die Unterschiede in den Metabolitengehalten zwischen den einzelnen Tiergrup‐pen nur in Einzelfällen signifikant waren, lassen sich gewisse Parallelen zwischen der Futter‐zusammensetzung und den Metabolitengehalten aufzeigen. Die Futtergruppen A und B mit einer höheren Rohproteinaufnahme haben auch einen höheren Gehalt an GLUC, LACT und AGP in Leber‐ und Fettgewebe, an AKG und MAL in der Leber und an PYR, 2‐PG, ACAC und ACCO im Fettgewebe. Die Futtergruppen B und C mit höherer Energieauf‐nahme haben höhere Gehalte an PYR in der Leber und PEP und MAL im Fettgewebe. In der Leber hatte die Energieaufnahme einen positiven Einfluß auf das NAD+/NADH‐Redoxverhältnis im Cytosol mit den höheren Werten in den Futtergruppen B und C, die Rohproteinaufnahme hatte einen negativen Einfluß auf die NAD+/NADH‐Verhältnisse in den Mitochondrien und die NADP+/NADPH‐Verhaltnisse in Cytosol und Mitochondrien. Im Fettgewebe konnten grundsätzlich entgegengesetzte Einflüsse im Vergleich zur Leber gefunden werden: ein negativer Einfluß der Energieaufnahme auf beide NAD+/NADH‐ und NADP+/NADPH‐Redoxverhältnisse im Cytosol und ein positiver Einfluß der Rohproteinaufnahme auf beide Redoxverhältnisse in den Mitochondrien.
