Drosophila melanogaster males lack recombination and have evolved a mechanism of meiotic chromosome segregation that is independent of both the chiasmatic and achiasmatic segregation systems of females. The teflon (tef ) gene is specifically required in males for proper segregation of autosomes and provides a genetic tool for understanding recombination-independent mechanisms of pairing and segregation as well as differences in sex chromosome vs. autosome segregation. Here we report on the cloning of the tef gene and the molecular characterization of tef mutations. Rescue experiments using a GAL4-driven pUAS transgene demonstrate that tef corresponds to predicted Berkeley Drosophila Genome Project (BDGP) gene CG8961 and that tef expression is required in the male germ line prior to spermatocyte stage S4. Consistent with this early prophase requirement, expression of tef was found to be independent of regulators of meiotic M phase initiation or progression. The predicted Tef protein contains three C2H2 zinc-finger motifs, one at the amino terminus and two in tandem at the carboxyl terminus. In addition to the zinc-finger motifs, a 44-to 45-bp repeat is conserved in three related Drosophila species. On the basis of these findings, we propose a role for Tef as a bridging molecule that holds autosome bivalents together via heterochromatic connections.
This article is devoted to study a compartmental mathematical model for the transmission dynamics of the novel Coronavirus-19 under Caputo fractional order derivative. By using fixed point theory of Schauder’s and Banach we establish some necessary conditions for existence of at least one solution to model under investigation and its uniqueness. After the existence a general numerical algorithm based on Haar collocation method is established to compute the approximate solution of the model. Using some real data we simulate the results for various fractional order using Matlab to reveal the transmission dynamics of the current disease due to Coronavirus-19 through graphs.
Component based software development (CBSD) endeavors to deliver cost-effective and quality software systems through the selection and integration of commercially available software components. CBSD emphasizes the design and development of software systems using preexisting components. Software component reusability is an indispensable part of component based software development life cycle (CBSDLC), which consumes a significant amount of organization’s resources, that is, time and effort. It is convenient in component based software system (CBSS) to select the most suitable and appropriate software components that provide all the required functionalities. Selecting the most appropriate components is crucial for the success of the entire system. However, decisions regarding software component reusability are often made in an ad hoc manner, which ultimately results in schedule delay and lowers the entire quality system. In this paper, we have discussed the analytic network process (ANP) method for software component selection. The methodology is explained and assessed using a real life case study.
SUMMARY
Calcineurin B‐like (CBL)‐interacting protein kinases (CIPKs) play a central role in Ca2+ signalling and promote drought tolerance in plants. The CIPK gene family in pigeon pea (Cajanus cajan L.), a major food crop affected by drought, has not previously been characterised. Here, we identified 28 CIPK genes in the pigeon pea genome. Five CcCIPK genes were strongly upregulated in roots upon drought treatment and were selected for further characterisation. Overexpression of CcCIPK13 and CcCIPK14 increased survival rates by two‐ to three‐fold relative to controls after 14 days of drought. Furthermore, the three major flavonoids, genistin, genistein and apigenin, were significantly upregulated in the same transgenic plants. Using CcCIPK14 as bait, we performed a yeast two‐hybrid screen and identified six interactors, including CcCBL1. CcCIPK14 exhibited autophosphorylation and phosphorylation of CcCBL1 in vitro. CcCBL1‐overexpressed plants displayed higher survival rates upon drought stress as well as higher expression of flavonoid biosynthetic genes and flavonoid content. CcCIPK14‐overexpressed plants in which CcCBL1 transcript levels were reduced by RNA interference had lower survival rates, which indicated CcCBL1 in the same pathway as CcCIPK14. Together, our results demonstrate a role for the CcCIPK14‐CcCBL1 complex in drought stress tolerance through the regulation of flavonoid biosynthesis in pigeon pea.
