Pseudomonas moorei KB4 is capable of degrading paracetamol, but high concentrations of this drug may cause an accumulation of toxic metabolites. It is known that immobilisation can have a protective effect on bacterial cells; therefore, the toxicity and degradation rate of paracetamol by the immobilised strain KB4 were assessed. Strain KB4 was immobilised on a plant sponge. A toxicity assessment was performed by measuring the concentration of ATP using the colony-forming unit (CFU) method. The kinetic parameters of paracetamol degradation were estimated using the Hill equation. Toxicity analysis showed a protective effect of the carrier at low concentrations of paracetamol. Moreover, a pronounced phenomenon of hormesis was observed in the immobilised systems. The obtained kinetic parameters and the course of the kinetic curves clearly indicate a decrease in the degradation activity of cells after their immobilisation. There was a delay in degradation in the systems with free cells without glucose and immobilised cells with glucose. However, it was demonstrated that the immobilised systems can degrade at least ten succeeding cycles of 20 mg/L paracetamol degradation. The obtained results indicate that the immobilised strain may become a useful tool in the process of paracetamol degradation.
Two-dimensional phase Doppler anemometer measurements of droplet size and velocity conducted under several nozzle conditions and a systematic variation of the air mass flow quality and liquid phase viscosity show that the air entrainment process is enhanced when keeping all test conditions constant except for increasing the Newtonian liquid viscosity above of that of water. A two-zone entrainment model based on a variable two-phase entrainment coefficient is proposed with the normalized axial distance allowing for a change in the jet angle. Thus, the jet perimeter is lower and the breakup length is longer in the case of air/relatively higher viscosity liquid phase. It provides the most accurate reproduction of the experimental droplet velocity in comparison with that of other models in the literature and, hence, is recommended for the prediction of the droplet velocity in the case of two-phase air/liquid phase free jet flow in stagnant ambient air. A model for predicting the droplet rain out, considering the droplet trajectories in the free jet flow, allows also for an adequate reproduction of the experimental data.
For the case of an isothermal free jet flow the velocity and the size of the drops were measured with a Phase-DopplerAnemometer. The axial velocity decay downstream from the nozzle outlet and the radial drop velocity profile is reasonably reproducible, if a so-called homogeneous flow mixture density in the nozzle outlet is introduced. In case of low mass flow qualities an adjustment of the entrainment coefficient and a virtual jet origin as well as a reduced exponent for the Gaussian function type radial velocity profile is required.
Der Entlastungsquerschnitt von Sicherheitsventilen im Fall einer Beaufschlagung mit höherviskosen Flüssigkeiten als etwa Wasser wird rechnerisch mit Hilfe eines von der Viskosität bzw. von der Reynolds-Zahl abhängigen Korrekturfaktors (s. Abb.) vergröûert, um u. a. die im Vergleich gröûeren Strömungsverluste zu berücksichtigen. Diese Vorgehensweise ist z. B. in der ISO 4126 bzw. der DIN EN 1268 und der US-amerikanischen Auslegungsempfehlung API RP 520 angegeben. Der Viskositäts-Korrekturfaktor ist jedoch noch nicht validiert worden. Im Rahmen von experimentellen Untersuchungen wurden zum einen bei Einphasenströmungen aus wässrigen Glucosesirup Lösungen mit Viskositäten von bis zu 3000 mPas gemäû der sog. Bauteilprüfung Ausflussziffern von handelsüblichen Ventilen ermittelt und daraus der Viskositäts-Korrekturfaktor ermittelt. Als vorläufiges Ergebnis gilt, dass die Korrektur gemäû API RP 520 im wesentlichen nur für Vollhubsicherheitsventile US-amerikanischer Bauart bzw. mit groûem Wert des bezogenen Hubes zutrifft. Für die Beaufschlagung der Ventile mit einer Zweiphasenströmung aus Flüssigkeit und Gas existiert keine für niedrigviskose Flüssigkeiten allgemein anerkannte Auslegungsmethode. Im Hinblick auf die Beibehaltung der bisherigen Vorgehensweise, einen Zuschlag bei der Gegebenheit einer hoch viskosen Flüssigphase vorzusehen, ist zunächst die Wiedergabegenauigkeit der am häufigsten verwendeten Auslegungsmethoden anhand von eigenen Messwerten mit Gemischen aus Wasser und Luft überprüft worden. Es gilt, dass die Messergebnisse dann besonders gut mit dem Praktiker-Modell von T. L E N Z I N G und dem Homogeneous-Frozen-Flow-Modell bzw. dem Maximalen-Ungleichgewicht-Modell nachgebildet werden können, wenn die Ausflussziffer bei Zweiphasenströmung gemäû T. L E N Z I N G verwendet wird. Die Ergebnisse bei Zweiphasenströmungen aus Luft und wässrigen, hochviskosen Glucosesirup-Lösungen lassen sich nur mit dem Maximalen-Ungleichgewicht-Modell in Kombination mit der wie im Praktiker-Modell eingesetzten Ausflussziffer wiedergeben. Insgesamt ist der Viskositätseinfluss auf den maximalen Ventildurchsatz in dem betrachteten Bereich geringer als zunächst von den Fachleuten befürchtet. Abbildung.
