The association of cyclosporin A (CsA) immunosuppression with inhibition of transcription factor-dependent lymphokine gene activation formed the basis of our decision to investigate nuclear-associated Cyp isoforms. Immunofluorescence microscopy of mouse macrophages cell line with a monoclonal antibody mAb7F1 raised against CypA shows a co-localisation of CypA in the nucleus and in the cytosol. Nuclear CypA binds to DNA in a zinc ion-dependent manner, in contrast to recombinant CypB. Peptidyl-prolyl cisltrans isomerase (PPIase) activity of nuclear CypA is inhibited by zinc ions. The zinc inhibited CypA does not bind cyclosporin A (CsA). We suggest that nuclear Cyp in complex with zinc ions recognizes DNA sequences and is involved in transcription modulating processes.
Using a Yarrowia lipolytica genomic library, several overlapping clones of the peroxisomal 3-oxoacyl-CoA-thiolase gene, POTI, were isolated. The library was prepared in the bacterial expression vector Agtll , thus allowing an immunological screening of recombinant bacteriophages with specific antibodies raised against purified peroxisomal thiolase. The isolated POTl clones hybridized to a 1.4 kb RNA species, which was induced approximately 30-fold when oleate was the carbon source. A 3634-bp segment of the cloned DNA was sequenced. This segment contained, on both strands, three major overlapping open-reading frames of 678,1122 and 1242 bp. Northern-hybridization analysis showed that only the largest of these reading frames was transcribed. It encodes a protein of 414 amino acids and molecular mass 43.059 kDa. Its deduced amino acid sequence has 30-60% identity and 50-70% sequence similarity when compared to other known thiolases. According to both the amount (68-71%) and location of conserved amino acids, the encoded protein belongs to the peroxisomal rather than the mitochondrial or cytoplasmic class of thiolases. Compared to bacterial and yeast cytosolic thiolases, the POT1 gene product contains a N-terminal extension of 25 amino acids which clearly differs from typical mitochondrial import signals. One of the isolated clones contained, in addition to the POTl coding sequence, 784 bp of the corresponding 5' flanking region. Nevertheless, it was efficiently expressed in Escherichia coli suggesting the correct recognition of this fungal promoter by the prokaryotic transcriptional and translational machinery. The I: lipolytica genomic POTI gene was disrupted by replacing 120 bp of its coding sequence with 2.7 kbp of DNA including the I: lipolytica LEU2 gene. The resulting dpotl : :LEU2 cells were free of immunologically cross-reacting thiolase. Western-blot analysis showed that the product of the non-disrupted gene had a molecular mass of approximately 42 kDa. This corresponds well to the molecular mass of purified K lipolytica peroxisomal thiolase. Disruption of POTI abolished the ability of K lipolytica cells to grow on solid media with oleate as a carbon source. This inability to grow in the presence of oleate suggests both the catabolic function of POTl and the absence of additional catabolic thiolases in K lipolytica. However, the dpotl : :LEU2 cells were unaffected in their ability to elongate externally added tridecanoic acid to its higher-chain-length homologues. Hence, another, POTl -independent and biosynthetic 3-oxoacyl-CoA thiolase must be responsible for this reaction in I: lipolytica.
Als am 11. November 1953 die Verfassungsurkunde des neuentstandenen Bundeslandes Baden-Württemberg unterzeichnet wurde, fand sich unter den Namen auf dem Dokument auch die Unterschrift Oskar Farnys. 1 Eben dieser war in politischer Hinsicht damals kein völlig Unbekannter mehr: Bereits für kurze Zeit hatte er 1920 als Abgeordneter dem Landtag des Freien Volksstaats Württemberg angehört. Auf eigenen Wunsch 1921 aus dem Stuttgarter Parlament ausgeschieden war er anschließend als Unternehmer und Kommunalpolitiker in seiner Heimatgemeinde tätig, bis er schließlich von 1930 bis nach den Wahlen vom 5. März 1933 Mitglied des Reichstags wurde. Gemeinsam mit Dr. Albert Hackelsberger wurde er Hospitant der NSDAP im späteren Großdeutschen Reichstag, dem er bis zum Ende des Dritten Reichs angehörte.In der französischen Besatzungszone war es ihm erst nach dem Urteil der Riedlinger Spruchkammer möglich, sich wieder politisch zu betätigen, mit Einverständnis der französischen Militärregierung war er bereits vorher auf ökonomischem Gebiet aktiv geworden. Nach der Bundestagswahl 1953, der einzigen Wahl, abgesehen von denen auf kommunaler Ebene, der er sich nach 1945 jemals stellte, war er, nur für sehr kurze Zeit, Abgeordneter des Deutschen Bundestags. Er folgte dem Angebot Dr. Gebhard Müllers, legte sein Mandat nieder, und wurde als Minister für Bundesratsangelegenheiten des Landes Baden-Württemberg Mitglied der Landesregierung. Ihr gehörte er in dieser Funktion sieben Jahre an, lehnte es unter anderem ab, die Nachfolge Müllers als Ministerpräsident Baden-Württembergs anzutreten, und schied auf eigenen Entschluss 1960 aus dem Amt.Im folgenden Beitrag 2 soll in kurzer Form ein Einblick in die Biographie Oskar Ludwig Farnys, zu dem bislang, abgesehen von einem Eintrag in Rabergs "Biographischen Handbuch der württembergischen Landtagsabgeordneten 1815-1933", keine wissenschaftlich historische Publikation vorliegt, gegeben werden. Dabei wird der Schwerpunkt auf der rein narrativen Darstellung liegen, die sich ergebenden, durchaus auch kontroversen, Forschungsfragen
