The conditions under which a high volume fraction of oil can be trapped in whey protein gels were studied. Oil-in-water emulsions of whey protein and vegetable oil were subjected to heat treatment. Such emulsions, depending on their protein and oil content, on their pH and on the emulsification technique used, gelled or remained liquid. Homogenization was the major factor to achieve gelation and the firmness of heat-induced gels increased with increasing emulsion fineness and homogeneity. Emulsions with a high gelation capacity were characterized by a single droplet family of relatively narrow size distribution and a mean droplet diameter ranging from roughly 300-700 nanometers. The pH range suitable for gelation extended from 3.5-8.0.
-Lactose crystallisation and Maillard reaction are two major modifications occurring in milk and whey powders during processing and storage. In this work, the aim was first to monitor the solid-state early Maillard reaction (EMR) in whey protein concentrates (WPC) heated at 60 °C and various water activities, and then, to characterise the physical changes that occur as a consequence of heat treatment in skim milk powder (SMP) and WPCs. After a w adjustment, SMP and WPC were heated at 60 °C in hermetic conditions to induce an amino-sugar reaction. Furosine analysis (% of blocked lysine) was used to monitor the progress of EMR. The results showed that the kinetic of EMR was linked to the initial a w of the powders. Heating of SMP may lead to the crystallisation of lactose in humidified powders, without apparently affecting the progress of EMR. Surprisingly, lactose crystallisation -monitored by micro-Differential Scanning Calorimetry -was easily induced in heated SMP, whereas it was delayed or even inhibited in WPC. L'objectif de ce travail était dans un premier temps de suivre la réaction de Maillard précoce (RMP) dans des concentrés de protéines de lactosérum (CPL) chauffés à 60 °C à différentes activités d'eau et, dans un deuxième temps, de caractériser les modifications physiques consécutives au traitement thermique (comparaison poudres de lait écrémé et CPL). Les poudres de lait écrémé et les CPL -équilibrés à différentes activités d'eau -étaient chauffés à 60 °C dans des conditions hermétiques pour induire la RMP (suivie par l'analyse de la furosine). Les résultats indiquaient que la cinétique de la RMP était liée à l'activité d'eau initiale de poudres. Dans les poudres de lait écrémé humidifiées et chauffées, la cristallisation du lactose ne semblait pas avoir d'impact sur l'évolution de la RMP. La microcalorimétrie différentielle à balayage permettait d'observer que la cristallisation du lactose, induite facilement dans les poudres de lait écrémé, était retardée voire inhibée dans les CPL. SUMMARY. Tryptic phosphopeptides were obtained from whole bovine casein by chromatography on the anion exchange resin QAE-Sephadex A 25. Salt gradient clution of the column allowed separation of non-phosphorylated peptidcs from phosphorylated species. The preparations obtained contained at least seven distinct phosphopeptides of which the following casein fragments were identified: a sl (43-58):2P, a sl (59-79): 5P, a s2 (46-70): 4P, /?(1-28):4P, /?(2-28):4P, and /?(33-48): IP. Fast protein liquid chromatography (FPLC) on Mono Q HR 5/5 resin showed that the phosphopeptides were eluted in the same order as from the QAESephadex resin. However, on the analytical column HR 5/5 the fragments a sl (59-79): 5P and /?(2-28): 4P, having the same net charge under the conditions of chromatography, co-eluted, whereas they were at least partly separated on the preparative column HR 16/10. Following enzymic dephosphorylation, the peptides eluted at lower salt strength in the gradient. FPLC on Mono Q resin thus permitted dephosphorylation to be monitored and intermediates between the parent species and the fully dephosphorylated peptide to be identified.
RésuméPar l'action de différentes protéases sur la protéine senque du lait in vitro, des hydrolysats partiels ont été obtenus. Ces derniers ont été ultrafiltrés pour obtenir une fraction peptidique sous forme du perméat d'ultrafiltration.Les préparations obtenues sont exemptes de lipides et composées essentiellement d'oligopeptides, de sels miné-raux et d'un faible taux de lactose.La tensioactivité des préparations peptidiques a été mesurée sur un instrument selon Lecomte du Noüy et dans un système huile d'arachide/phase aqueuse. Les préparations peptidiques ont provoqué des baisses de tension inter faciale plus ou moins fortes. Les prépa-rations trypsiques et chymotrypsiques ont montré l'effet le plus fort. Entre tensioactivité et longueur moyenne de la chaîne peptidique de nos préparations il existe une corrélation positive Cr = 0.9 pour la régression linéaire). Les propriétés émulsifiantes ont été étudiées dans des émulsions de 20 % d'huile de pépin de raisin dans l'eau. Les prépa-rations peptidiques à l'exception d'une, sont pratiquement exemptes de pouvoir émulsifiant. La préparation trypsique est capable de supprimer coalescence et de réduire l'instabilité au « creaming )}de façon significative. Cette activité est mieux exprimée à pH neutre ou alcalin et diminue brusquement en dessous de pH 6.
-The heat treatment of milk powders during food manufacturing induces physical and chemical modifications that are governed by material phase transitions. The main aim of this work was to describe the heat-induced physical changes occurring in whole milk powder (WMP). The study was based on increasing the water activity (a w ) of the powder in order to relate the glass transition temperature (Tg) to the parameters resulting in changes in physical properties. Colour changes in milk powder due to non-enzymatic browning (NEB) were monitored by spectrophotometry based on the CIE L*a*b* function. Changes in particle structure were investigated by scanning electron microscopy (SEM). WMPs at low and high water activities (0.28 and 0.35) were heated in closed tubes and their physical properties (a w , lactose crystallisation and free fat release) monitored. The results showed that the kinetics of lactose crystallisation and the resulting increase in a w were clearly influenced by the Tg, modulated here by the water content of WMP at the time of heat treatment. The kinetics of free fat release was related to the trend of lactose crystallisation. SEM showed that the free fat was released onto the particle surfaces and formed a fat layer covering the lactose crystals. Porosity then resulted inside the powder matrix due to empty cavities formerly occupied by fat globules. In addition, the chemical reactivity of WMP expressed by colour changes showed a delay in triggering the NEB reaction in milk powder at low water content only. The results of this study showed that the heat treatment of WMP under specific process parameters (a w , temperature and time) enables not only modulation of free fat release and browning but also changes in the microstructure of the powder particles.whole milk powder / heat treatment / free fat / crystallisation / browning Résumé -Changements physiques de la poudre de lait entier traitée thermiquement.
