Fractionation of extracts from the culture broth of the marine-derived fungus, Paecilomyces sp. 7A22, resulted in the isolation of the harzialactone A (HA), a known compound previously isolated from fungi of marine environments. The chemical structure of HA was determined by spectroscopic analyses. Upon evaluation of HA on antileishmanial assays against Leishmania amazonensis, HA exhibited significant activity against promastigotes forms with IC 50 of 5.25 µg mL -1 and moderate activity against intracellular amastigotes with IC 50 of 18.18 µg mL -1 . This is the first report on the antileishmanial activity of HA, and the effects of HA presented in this work suggest that this class of compounds are suitable for future biological in vitro and in vivo studies for the search of natural products with activity against Leishmania spp. Furthermore, the present results corroborate marine-derived fungi as a promising source of natural products with antiparasitic activity.
Anacardium othonianum Rizz., popularly known as the Cerrado cashew tree, is widely used in medicine due to its therapeutic properties, including its antioxidant, anti-inflammatory, and antidiarrheal activities. Light is an important regulator of the growth, development of plants differentiation, and metabolism. Considering the influence of light on the production of phytochemicals, the aim of the present study was to evaluate production of gallic and 3,4-dihydroxybenzoic acids and amentoflavone in A. othonianum seedlings grown in vitro exposure to white, blue, green, red, and yellow light. The seedlings were evaluated with respect to the length of the aerial parts and the number of leaves, at 0, 20, and 40 days of treatment. The fresh and dry weights of the aerial parts and the levels of phenolic acid and biflavonoid were evaluated at 40 days via high-performance liquid chromatography with a diode array detector (HPLC-DAD). Regardless of the light wavelength used for the growth, the largest length of the aerial parts (5.54 cm) and an increase in the number of leaves (2.86-fold) were observed at 40 days. The largest accumulations of fresh weight (325.5 mg and 335.1 mg per seedling) and dry weight (83.3 mg and 84.5 mg per seedling) were observed in seedlings grown under red and yellow light, respectively. Relatively higher levels of gallic acid (0.66 mg.g -1 dry weight) and amentoflavone (1.41 mg.g -1 dry weight) were found in seedlings grown under blue light. In contrast, the 3,4-dihydroxybenzoic acid levels of the seedlings under red light increased (8.22 mg.g -1 dry weight).
Hyptis marrubioides Epling is a native plant from Brazilian Cerrado. In this paper, the response of in vitro microplants of this species to inoculation with bacterial and fungal endophytic isolates is evaluated. HPLC-DAD analysis showed the presence of 3,4-O-(Z)-dicaffeoylquinic acid and quercetin-7-O-glucoside as the main components. GC/MS analysis demonstrated that the sesquiterpenes t-cadinol and caryophyllene oxide were only produced in microplants inoculated with endophytic bacteria, while methyl hexadecanoate, methyl heptadecanoate and methyl (Z,Z,Z) 9,12,15-octadecatrienoate and the triterpene methyl 3β-hydroxyurs-12-en-28-oate were overexpressed only when the microplant was treated with endophytic fungi.
Hyptis marrubioides (Lamiaceae) is a medicinal plant that is native from Brazilian Cerrado. In vitro propagation techniques make use of elicitors to alter metabolic pathways, affecting how molecules are produced both qualitatively and quantitatively. This research aimed to evaluate how abiotic elicitors salicylic acid (SA) and silver nitrate (SN) at concentrations of 30µM or 60µM influence Hyptis marrubioides seedling growth by two different in vitro culture methods. The rutin content was quantified by HPLC-DAD. Compared to an untreated culture, the H. marrubioides methanolic extracts cultured in MS medium for 10 days followed by culture in MS medium containing SN (30µM) for 20 days had 1.28 times higher rutin content. In a second experiment, seedlings were cultured in MS medium for 20 days, and then the desired elicitor was added to the culture and allowed to remain in contact with the medium for three and six days. SA (30µM) gave the best results: rutin production was 16.56-foldhigher than the control after six days. SN (30µM) increased the rutin content by 1.17-fold. At the two concentrations evaluated during the elicitation experiments, neither SA nor SN altered the growth parameters shoot length, leaf number, and fresh and dry weight of H. marrubioides seedlings grown in vitro as compared to the control. Based on these results, the abiotic elicitors SA and SN successfully provide Hyptis marrubioides with increased rutin content in vitro.
