The aim of this study was to evaluate and compare the efficacy of B-mode, Doppler, contrast-enhanced ultrasonography (CEUS), and Acoustic Radiation Force Impulse (ARFI) elastography in predicting malignancy in canine mammary masses. This was a prospective cohort study from 2014 to 2016, which included 153 bitches with one or more mammary masses. A total of 300 masses were evaluated by ultrasonography (B-mode, Doppler, CEUS, and ARFI) and subsequently classified as benign or malignant by histopathology. Each ultrasound parameters studied were compared between benign and malignant masses by Chi-square or Student tests and differences were considered significant when P < 0.01. For the variables that proved significant differences were estimated the cut-off point, sensitivity, specificity, accuracy, and area under curve (AUC) by receiver-operating characteristic curve (ROC) analysis in a logistic regression model using histopathological classification as reference, to assess and compare diagnostic performance of each technique. Out of 300 mammary masses evaluated 246 were classified as malignant and 54 as benign. B-mode measurements showed sensitivity 67.9%, and specificity 67.6% as malignancy predictors on canine mammary masses; Doppler indexes systolic (>21.2 m/s) and diastolic velocity (>4.8 m/s) sensitivity 79.2% and specificity 70.8%; CEUS wash-out time (<80.5 s) sensitivity 80.2% and specificity 16.7%; and ARFI elastography shear velocity (SWV > 2.57 m/s) sensitivity 94.7% and specificity 97.2% In conclusion B-mode and Doppler ultrasound evaluations may assist in malignancy prediction of canine mammary masses with moderate sensitivity and specificity, already the SWV was an great accurate predictor. Therefore, ARFI elastography exam inclusion in veterinary clinic oncology and research is highly recommended, since it allows fast, non-invasive, and complication-free malignancy prediction of canine mammary masses.
RESUMOObjetivando analisar a prevalência de TVT em relação ao sexo, raça, idade, localização da lesão e resposta ao tratamento quimioterapêutico foi realizado um estudo retrospectivo de 144 cães atendidos no Hospital Veterinário da Universidade de Uberaba (HVU) durante os anos de 2007 à 2012, diagnosticados com TVT através da realização de exames de cito ou histopatologia. Destes, 59% eram cães sem raça definida (SRD). Apesar de 16,7% dos cães avaliados não terem suas idades determinadas, os resultados desta pesquisa revelaram que os pacientes apresentaram idade variando entre 1 e 15 anos sendo a faixa etária entre 2 e 8 anos (40,2%) a mais afetada. Em relação ao sexo, este trabalho demonstrou que as fêmeas foram mais acometidas que os machos (66,7 e 33,3% respetivamente). Observou-se que a principal localização da neoplasia foi a vulva (65,3%), seguida pelo pênis (30,6%), cavidades nasal (3,5%) e oral (0,7%). Dentre os 54 cães que foram submetidos ao tratamento no hospital veterinário, 39 concluíram o protocolo, sendo que 33 apresentaram citorredução completa utilizando vincristina como agente único; 3 animais se encontravam em tratamento ao final do estudo e nos 3 casos em que houve quimiorresistência, os pacientes se beneficiaram de doxorrubicina. SUMMARYTo analyze the prevalence of TVT in relation to sex, breed, age, lesion location, and response to chemotherapeutic treatment, a retrospective study from 2007 to 2012 was conducted with dogs from the Veterinary Hospital of the University of Uberaba (HVU) diagnosed with TVT by cytology or histopathology. Which, 59% were mongrel dogs (59%). Although 16.7% of these dogs had their ages undetermined, the data of this study showed that patients were aged between 1 and 15 years being the range between 2 and 8 years (40.2%) the most affected. Regarding sexual predisposition, this study shows that females were more affected than males (66.7, 33.3% respectively). The primary site of the tumor occurrence was the vulva (65.3%), followed by the penis (30.6%), nasal (3.5%) and oral cavities (0.7%). Among 54 dogs that were treated at the hospital 39 dogs completed therapeutic protocol, of these 33 dogs had complete cytoreduction using the single-agent vincristine, 3 animals were still receiving treatment at the end of the study and in the 3 chemoresistance cases, these patients were benefited from doxorubicin treatment.
The aim of this study was to evaluate and compare the diagnostic accuracy of B-mode, Doppler ultrasonography and Acoustic Radiation Force Impulse (ARFI) elastography in the identification of axillary and inguinal lymph nodes metastasis in bitches with mammary neoplasms. The axillary (n = 96) and inguinal (n = 100) lymph nodes of 100 bitches were evaluated using B-Mode, Colour Doppler and ARFI-elastography. After this evaluation, mastectomy and lymph nodes excision were performed and these structures were histologically classified as free, reactive or metastatic. Ultrasonographic parameters were compared by Chi-Square or ANOVA tests and if they are significant, discriminative power analysis according to histopathological classification was performed (ROC analysis). The ARFI-elastography shear wave velocity (SWV) enabled metastasis identification in inguinal (sensitivity 95% specificity 87%) and axillary lymph nodes (sensitivity 100% specificity 94%). While B-Mode ultrasound Short/Long axis ratio evaluation of inguinal and axillary lymph nodes only resulted in a sensitivity around of 71% and specificity of 55%. In conclusion, B-Mode ultrasonography may contribute to diagnosis of metastasis in axillary and inguinal lymph nodes of bitches affected by mammary neoplasm with limited accuracy, while SWV evaluation proved to be an excellent diagnosis tool, which allows differentiation between free, reactive and tumour metastatic lymph nodes.
The objective of this study was to investigate three storage methods and four storage times for rumen sampling in terms of quality and yield of extracted metagenomic DNA as well as the composition of the rumen bacterial community. One Nellore steer fitted with a ruminal silicone-type cannula was used as a donor of ruminal contents. The experiment comprised 11 experimental groups: pellet control (PC), lyophilized control (LC), P-20: pellet stored frozen at -20°C for a period of 3, 6, and 12 months, P-80: pellet stored frozen at -80°C for a period of 3, 6, and 12 months, and L-20: lyophilized sample stored frozen at -20°C for a period of 3, 6, and 12 months. Metagenomic DNA concentrations were measured spectrophotometrically and fluorometrically and ion torrent sequencing was used to assess the bacterial community composition. The L-20 method could not maintain the yield of DNA during storage. In addition, the P-80 group showed a greater yield of metagenomic DNA than the other groups after 6 months of storage. Rumen samples stored as pellets (P-20 and P-80) resulted in lower richness Chao 1, ACE, and Shannon Wiener indices when compared to PC, while LC and PC were only different in richness ACE. The storage method and storage time influenced the proportions of 14 of 17 phyla identified by sequencing. In the P-20 group, the proportion of Cyanobacteria, Elusimicrobia, Fibrobacteres, Lentisphaerae, Proteobacteria, and Spirochaetes phyla identified was lower than 1%. In the P-80 group, there was an increase in the proportion of the Bacteroidetes phylum (p = 0.010); however, the proportion of Actinobacteria, Chloroflexi, SR1, Synergistetes, TM7, and WPS.2 phyla were unchanged compared to the PC group (p > 0.05). The class Clostridium was the most abundant in all stored groups and increased in its proportion, especially in the L-20 group. The rumen sample storage time significantly reduced the yield of metagenomic DNA extracted. Therefore, the storage method can influence the abundance of phyla, classes, and bacterial families studied in rumen samples and affect the richness and diversity index.
Acoustic radiation force impulse elastography of the testes and prostate in dogs was easily implemented. This study provides baseline data for these organs.
The objective was to characterize the vascular patterns of testicular blood flow of adult cats, measuring the systolic velocity (SV), diastolic velocity (DV), resistance index (RI), gate time (wash-in) peak enhancement and output time (wash-out) of the contrast and addition of tissue fill characteristics. Forty-five adult cats were selected, and the echotexture, echogenicity, size, contours and margins of testicles were assessed via ultrasound. By Doppler were evaluated the blood flow and determined of vascular index in testicular artery (SV, DV and RI) and via contrast-enhanced ultrasonography determine the time for phases: wash-in, wash-out and peak enhancement. Sonographic findings presented normal. Testicular artery was observed in the spermatic cord with tortuous patter and showed monophasic-patterned waves and low vascular resistance and with systolic peak evident. Values of indices vascular were as follows: SV = 6.73 cm/s, DV = 2.8 cm/s and RI = 0.54 for left testicles; and SV = 6.23 cm/s, DV = 2.77 cm/s and RI = 0.53 for right testicles. Contrast filled the subcapsular vascular structures and after a few seconds, a homogeneous moderate enhancement of the parenchyma, with parenchymal vessels still distinguishable and after the peak phase, a rapid homogeneous decrease in echogenicity. Values of time for contrast-enhanced ultrasonography were as follows: wash-in = 8.78 s, peak enhancement = 21.62 s and wash-out = 75.36 for left testicles; and wash-in = 10.76 s, peak enhancement = 21.50 s and wash-out = 81.81 for right testicles. Doppler and contrast-enhanced ultrasonography of the testicles in healthy adult cats was easily implemented and may provide baseline data for this organ to allow the use of these techniques as a diagnostic tool for evaluating testicular abnormalities in sick cats.
Quantitative and qualitative acoustic radiation impulse force elastography of the kidney in adult cats was easily performed and this study provides base line data to allow the use of acoustic radiation impulse force in diseased animals.
Objective The aim of this study was to compare in vitro accuracy of femoral measurements obtained from different radiographical positions with actual femoral anatomical dimensions in dogs. Materials and Methods Craniocaudal projections of the femur in ventrodorsal position, standard craniocaudal radiographical (SR) and craniocaudal radiography with horizontal radiographical beam (HR), in addition to anatomical macroscopic measurements (A), were obtained from 45 femurs from 23 canine cadavers, for calculation of the canal flare index (CFI). The differences between the radiographical positions and 95% limits of agreement (LOA) between the CFI-SR, CFI-HR and CFI-A were evaluated by the Bland–Altman method. Results The standard craniocaudal radiograph provided an approximate dimension of the proximal femoral morphology (mean difference: 0.121 ± 0.391) with a different CFI value (p = 0.0341) from CFI-A. The craniocaudal radiograph taken using a horizontal beam provided greater accuracy (mean difference: 0.087 ± 0.42) in our study. Clinical Significance The craniocaudal projection using a horizontal radiographical beam was more accurate than the standard craniocaudal projection for measurement of the true anatomical dimensions of the canine femur, minimizing the influence of the technique on the CFI values.
