Tritrichomonas foetus and Trichomonas vaginalis are extracellular parasites of the urogenital tract of cattle and humans, respectively. They cause infertility and abortion, but there is no documented information on the susceptibility of bovine sperm cells to this cattle parasite. The aim of this present work was to study the effects provoked by T. foetus and T. vaginalis when in interaction with bovine and human sperm cells. The bovine and human spermatozoa were obtained from uninfected bulls and men, respectively, and were exposed to living trichomonads over different periods of time. Light microscopy, video microscopy, scanning, and transmission electron microscopy first revealed a tropism, then a close proximity followed by a tight adhesion between these two different cells. A decrease in the spermatozoa motility was observed as well intense semen agglutination. The adhesion between trichomonads to the sperm cell occurred either by the flagella or sperm head. Motile parasites were observed during the next 12 h, whereas sperm cells in contact with the parasites rapidly became immotile. The parasites were able to maintain the sperm cells attached to their cell surface, followed by phagocytosis. This process began with a tight membrane-membrane adhesion and the incorporation of the sperm cell within an intracellular vacuole. Afterwards, the sperm cell was gradually digested in lysosomes. Many trichomonads were injured and/or died on making contact with the spermatozoa possibly due to necrosis. Results from this study demonstrated that both T. foetus and T. vaginalis interact with sperm cells provoking damage and death of these reproductive cells. Differences in the behavior of both trichomonads were evident, showing that T. vaginalis was much more virulent than T. foetus. The possible role of trichomonads in reproductive failure is discussed.
Quarenta embriões de éguas da raça Mangalarga Marchador foram inovulados em igual número de receptoras da mesma raça. O primeiro grupo foi tratado com 1,1 mg/kg de flunixin meglumine, visando observar o efeito desta droga sobre a taxa de gestação na transferência não-cirúrgica de embriões e seu efeito sobre as concentrações plasmáticas de progesterona. O segundo grupo não foi tratado. O grupo tratado apresentou 55% de taxa de prenhez (11/20), e o controle 75% (15/20) não existindo diferença entre eles. Contudo, para P=20% esta diferença pode ser considerada significativa. O tempo de sincronização das receptoras e a idade dos embriões no momento das inovulações foram similares. Houve diferença na qualidade dos embriões inovulados, pois as éguas não-gestantes do grupo controle receberam um número maior de embriões regulares (2/5) que as não-gestantes do grupo tratado (1/9). As concentrações plasmáticas de progesterona foram maiores nas receptoras gestantes do grupo tratado. Tendo em vista os resultados obtidos, não se recomenda a aplicação de flunixin meglumine em receptoras antes da transferência não-cirúrgica de embriões, com o objetivo de melhorar a taxa de gestação.
-Tritrichomonas foetus is an extracellular parasite of the urogenital tract in cattle. It causes infertility and abortion, but there is no documented information on the susceptibility of bovine oocytes to the parasite, except by one article that claimed no effects of T. foetus on oocytes or embryos. The aim of the present study was to study the effects provoked by T. foetus when in interaction with bovine oocytes. Oocytes were obtained from cow ovaries and divided into two groups: (1) one group contained cumulus cells, whereas (2) a second group was denuded from these cells. Light microscopy, video microscopy and scanning electron microscopy (SEM) revealed that exposure of oocytes to T. foetus caused rapid adhesion of the trichomonads to cumulus cells and to the zona pellucida (ZP). Motile parasites were observed for 12 h. The ZP was completely damaged, and the parasites were able to infiltrate beneath the ZP and reached the oocytes directly when the oocytes were denuded of the cumulus cells. Both the oocytes and the cumulus cells exhibited morphological characteristics compatible with apoptosis after interaction with T. foetus, such as chromatin condensation, the presence of several cytoplasmic vacuoles, with intact cellular membranes and organelles. The results from this study demonstrate that when a large number of T. foetus interacts with oocytes in vitro damage and apoptosis are provoked in the cow's reproductive cells. The behavior of this parasite as one of the causes of cattle infertility is discussed.Tritrichomonas foetus / oocytes / cow / ultrastructure / infertility
-The objective of this work was to evaluate the reproductive performance of Santa Inês ewes fed a diet supplemented with protected fat. Intervals from lambing to first clinical estrus and to conception, conception rate, prolificacy, live weight and body condition were determined. After lambing, 60 ewes and their offsprings were weighted and randomly assigned to three treatments, based on age, body weight and number of born lambs. Treatments consisted of: control diet, or control diet plus 30 g of protected fat, from lambing to day 25 of post-lambing (Sup25), or to day 60 of post-lambing (Sup60). Out of 60 evaluated ewes, 93.3% returned to estrus, and 74.5% got pregnant, with 73.53% lambing rate and 196.5 days lambing interval. The average periods from lambing to first estrus were 32.4, 27.2 and 35.5 days for ewes fed the control diet, Sup25, and Sup60, respectively. The intervals from lambing to conception were 45.2, 46.5 and 45.2 days, and the supplemented diets did not show differences in comparison to the control diet. Supplementation with protected fat to well-nourished Santa Inês ewes does not improve their reproductive performance.Index terms: Ovis aries, conception rate, estrus, lambing interval, nutrition. Os intervalos do parto ao primeiro estro clínico e à concepção, a taxa de gestação, a prolificidade, o peso vivo e a condição corporal foram determinados. Depois do parto, 60 ovelhas e seus cordeiros foram pesados e distribuídos aleatoriamente em três tratamentos com base na idade, peso corporal e número de cordeiros nascidos. Os tratamentos consistiram de dieta controle ou da dieta controle mais 30 g de gordura protegida, do parto aos 25 dias pós-parto (Sup25), ou aos 60 dias pós-parto (Sup60). Das 60 ovelhas avaliadas, 93,3% retornaram ao estro, e 74,5% ficaram prenhes, com taxa de parição de 73,53% e intervalo de parto de 196,5 dias. O período médio do parto até o primeiro estro foi de 32,4, 27,2 e 35,5 dias, para as ovelhas alimentadas com a dieta controle, Sup25 e Sup60, respectivamente. Os intervalos do parto até a concepção foram de 45,2, 46,5 e 45,2 dias, e as dietas com suplementação não apresentaram diferenças em comparação ao tratamento controle.
