Abstract. Widyawan MB, Hasanah A, Taryono, Alam T, Sayekti RS, Pramana AAC, Wulandari RA. 2020. Multivariate analysis unravels genetic diversity and relationship between agronomic traits, protein, and dietary fiber in yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.). Biodiversitas 21: 5662-5671. Yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.) is a vegetable legume that possesses the highest amount of protein and total dietary fiber (TDF) content among other vegetables. However, protein and TDF content diversity are found among yardlong bean genotypes. Genetic improvement to produce cultivars with high yield and improved nutritional content is required. This study aimed to analyze genetic diversity and identify the relationship between important agronomic traits from 21 yardlong bean genotypes assessed by multivariate statistical analysis. The experiment was arranged in alpha design with three replicates, each consist of six blocks. Six pod-related important agronomic traits, protein, and TDF content were observed in this study. Total pod weight, pod weight, and pod length were found to be negatively correlated with protein and TDF content based on correlation and principal component analysis (PCA). Although negatively correlated, several variables possess a large and positive direct effect on protein and TDF content, such as the number of pod, pod weight, and days to flower. D2 analysis and Tocher’s method classify genotypes used in this study into five clusters with diverse traits value. Information from this study laid a foundation for the improvement of important agronomic traits as well as nutritional content in yardlong bean.
Cosmos has the potential to be developed as an alternative vegetable. Less characterization and evaluation of cosmos become obstacles in the process of its development. Characterization is the process of observing and identifying the physical, biological, and chemical properties of plants that aim to determine the characteristics of a plant. The purpose of this research is to ind the characteristics of each accession of Cosmos spp. and get accession of Cosmos spp. which has the potential to be developed or used as varieties material assembly. The research was conducted at the AIC (Agrotechnology Innovation Center) of Universitas Gadjah Mada in March-July 2019. The planting material used for this study was the collection of seeds of PIAT UGM from the Special Region of Yogyakarta and Riau. Fifteen cosmos accessions came from DIY and the other 18 came from Riau. Thirty-three accessions have diverse characteristics especially lower colours that are divided into 7 groups of lower colours namely yellow, gold, light orange, orange, pink, purple, and white. Accession number 39 has a rare white colour lower
Plant tissue culture is a technology used to propagate plants under controlled and aseptic environmental conditions. One of the obstacles in tissue culture activities is the presence of bacterial and fungal contamination. Equipment and growing media that are used for tissue culture can be the source of contamination. The method that can be used to reduce contamination is using sterilization. Sterilization is used to kill or eliminate microorganisms so that the equipment and media are free from contaminants. Equipment sterilization can be done through several methods such as wet sterilization and dry sterilization. Wet sterilization is carried out using an autoclave, while dry sterilization can be carried out using an oven, fire, and glass bead sterilizer. Media sterilization is generally carried out using an autoclave or membrane filter.
The population increase will be accompanied by an increase in the food needed. The agricultural sector is an area which is wrought by the majority of the population in all over Indonesia. The need for land lots to do enough to be able to increase food needs people of Indonesia. Currently, function transfer land acquisition is one of the problems that make a decreased production of agricultural products. Aquaponics is an alternative to cultivate plants and fish in one place. This technique to integrate the cultivation of fish ( recirculation ) aquaculture in coupled with plants. The bucket filled with approximately 60 l then mixed with manure 250 g / 1000 l, manure 500 g / 1000 l, compost 250 g / 1000 l, compost 500 g / 1000 l, and water control without additional fertilizer. The application of organic fertilizers in the form of manure and compost Aquaponics water spinacs leaves to improved growth than without the application of organic fertilizer. The application of manure with measure 500 g / 1000 l results is the best growth aquaponics water spinach leaves.
Yardlong bean is one of the commodities with very high economic value. To obtain good quality pods, it requires the right cultivating techniques such as the use of stake model and the use of superior varieties. The objectives of this study were to investigate the effect of stake model and varieties on the quality of yardlong bean pods. This research was conducted on the Agrotechnology Innovation Center of Universitas Gadjah Mada at March-June 2019. This research was arranged in factorial treatment design with Randomized Complete Block Design (RCBD) using three blocks as replication. The irst factor is the stake model i.e. single, double, and triangle. The second factor is varieties included Parade Tavi, Pujangga, Katrina, Wulung, and Kaloka. The observed quantitative data were analyzed using analysis of variance (ANOVA), followed by Scott-Knott with the level of credibility at 5 %. The results showed that the use of the stake models on different varieties does not indicated a signiicant impact on the quality of produced pod.
