RESUMO:Este trabalho objetivou a obtenção de óleos fi xos (OF) das cascas do caule de Croton cajucara via percolação [seguido de fracionamento em coluna cromatográfi ca (CC)] e fl uido supercrítico (FS, gás CO 2 ), bem como a avaliação das propriedades fungicidas de OF obtido pelo procedimento CC. Os óleos OF-CC e OF-FS apresentaram rendimentos signifi cativamente diferentes; houve maior rendimento (3,14%) de OF obtido no processo convencional (OF-CC), versus 1,2% de OF-FS. A fração F 1 após esterifi cação, bem como as frações não-esterifi cadas F 2 , F 3 e F 4 (obtidas de OF-CC seguido de outro fracionamento em CC) foram analisadas por CG-FID e CG-EM. A identifi cação da composição química de OF-CC foi feita por comparação com dados da biblioteca Wiley, bem como pela comparação de índices de kovats. A presença de sesquiterpenos foi comprovada na fração F 1 , onde o α-copaeno (20,1%) e cipereno (21,8%) foram os componentes majoritários (70% de percentual sesquiterpênico total). A fração F 2 mostrou sesquiterpenos minoritários mais oxigenados, incluindo o linalol. Na fração F 3 foram observados ácidos graxos, misturas de esteróis e os diterpenos bioativos trans-crotonina, cis-cajucarina B e trans-cajucarina B, onde 40% do total desta fração, foi composta de esteróis e diterpenos; e na fração F 4 foram observados majoritariamente os isômeros cis e trans-cajucarina B. O efeito biológico de OF-CC foi avaliado no desenvolvimento in vitro dos fungos fi topatogênicos Fusarium oxysporum, Rhizoctonia solani e Sclerotium rolfsii propagados em meio de cultura Batata-dextrose-agar (BDA). A resposta fi siológica, variou de acordo com o tipo de fungo testado, tendo sido observado que o OF-CC (0,2 mg.mL -1 ) apresentou efeito fungistático no controle micelial dos fungos testados, sendo que o gênero de fungo Fusarium oxysporum foi o que sofreu efeito inibitório mais estável. This paper focused on the separation of the fi xed oil (FO) from the stem bark of Croton cajucara obtained from the conventional approach extraction with organic solvents [followed by chromatography column (CC)] and supercritical fl uid extraction (SFE, carried out with CO 2 ) and also, on the antifungical properties of the FO obtained by CC procedure (FO-CC). The FO contents were 3.14% for FO-CC versus 1.20% in the FO-SFE process. The esterifi ed fraction F 1 and nonesterifi ed fractions F 2 , F 3 and F 4 obtained from FO-CC after a new CC procedure were analyzed by HRGC-MS. Do lenho do caule de Plathymenia reticulata (Leguminosae) foram isolados dois novos diterpenos cassânicos: 16,18-diacetoxicass-13(15)-eno e 16-hidroxi-18-acetoxicass-13(15)-eno, além dos conhecidos diterpenos platiterpol, acetato de vinhaticila e vinhaticoato de metila. As estruturas foram determinadas com base em análise espectroscópica, envolvendo comparação com dados da literatura e transformações químicas.From trunk heartwood of Plathymenia reticulata (Leguminosae) two novel cassane diterpenes 16,18-diacetoxycass-13(15)-ene and 16-hydroxy-18-acetoxycass-13(15)-ene, and the known diterpenes plathyterpol, vinhaticyl acetate and methyl vinhaticoate were isolated. Structure determinations were accomplished by spectroscopic analysis involving comparison with data from literature and chemical transformations.
Neste trabalho utilizou-se espectrometria de massas objetivando-se a identificação de fragmentos característicos de diterpenos do tipo clerodano contendo carbonila α,β-insaturada no anel A do sistema decalínico. Para tanto, dados de espectrometria de massas foram analisados para os seguintes 19-nor-clerodanos de sistemas enona: trans-desidrocrotonina (1), trans-crotonina (2), cis-cajucarina B (3), trans-cajucarina B (4) e cajucarinolida (5) e para os clerodanos isosacacarina (6) e cajucarina A (7), isolados das cascas do caule de Croton cajucara Benth, bem como para clerodanos de sistemas enona isolados de outras espécies. A junção trans de clerodanos foi claramente correlacionada com a presença dos íons característico de m/z 95, 121 e 205; os sistemas de junção cis apresentaram íons característicos de m/z 122 e 124. A junção trans de 19-nor-clerodano foi correlacionada com a presença dos íons característico de m/z 161, 134 e 121.
A família Malvaceae, constituída por mais de 100 gêneros, totaliza 2500 espécies. Dentre os gêneros destacam-se: Hibiscus (300), Sida (200) e Pavonia (150). No Brasil, as folhas da espécie Pavonia varians são utilizadas para combater infecções do aparelho digestivo, bem como inflamações de boca e garganta, tendo sido comprovado que este gênero é rico em alcalóides e saponinas. Atualmente, a família Brassicaceae, compreende aproximadamente 4000 espécies distribuídas em 400 gêneros, tendo resultado em função da fusão com a família Capparaceae, em um aumento significativo. Como exemplo, destaca-se a espécie medicinal Cleome spinosa Jacq. (rica em flavonóides) classificada até recentemente, como pertencente à família Capparaceae, tendo sido confirmado que pertence à família Brassicaceae. A atividade antioxidante dos extratos hidroalcoólicos de Pavonia varians (PV) e Cleome spinosa (CS) solubilizados em DMSO e MeOH (respectivamente), bem como em sistemas microemulsionados (SME), foi avaliada frente ao radical livre DPPH. Dois diferentes tipos de sistemas SME foram utilizados contendo uma mistura dos tensoativos Tween 80 e Span 20 (3:1), miristato de isopropila (IPM) como fase orgânica e água bidestilada (sistema SME-4, isento de cotensoativo) e ainda, adição de etanol como cotensoativo, para o sistema SME-1. Comparativamente, evidenciou-se para ambas as espécie Pavonia varians [CE50 = 114 μg/mL (SME-1) e 246 μg/mL (SME-4)] e Cleome spinosa [CE50: 224 μg/mL (SME-1) e CE50: 248 μg/mL (SME-4)], melhor eficácia para o sistema SME-1. A atividade antioxidante do extrato de P. varians foi significativo em ambos os sistemas SME-1 e SME-4, sendo duas vezes melhor para o sistema SME-1. Em relação aos sistemas encapsuladores SME-1 e SME-4 não foi observada interferência significativa destes sistemas, nos resultados observados para a atividade antioxidante de P. varians e C. spinosa.
