A safe and effective rotavirus vaccine is urgently needed, particularly in developing countries. Critical to vaccine development and implementation is a knowledge base concerning the epidemiology of rotavirus G and P serotypes/genotypes throughout the world. The temporal and geographical distribution of human rotavirus G and P types was reviewed by analysing a total of 45571 strains collected globally from 124 studies reported from 52 countries on five continents published between 1989 and 2004. Four common G types (G1, G2, G3 and G4) in conjunction with P[8] or P[4] represented over 88% of the strains analysed worldwide. In addition, serotype G9 viruses associated with P[8] or P[6] were shown to have emerged as the fourth globally important G type with the relative frequency of 4.1%. When the global G and/or P type distributions were divided into five continents/subcontinents, several characteristic features emerged. For example, the P[8]G1 represented over 70% of rotavirus infections in North America, Europe and Australia, but only about 30% of the infections in South America and Asia, and 23% in Africa. In addition, in Africa (i) the relative frequency of G8 was as high as that of the globally common G3 or G4, (ii) P[6] represented almost one-third of all P types identified and (iii) 27% of the infections were associated with rotavirus strains bearing unusual combinations such as P[6]G8 or P[4]G8. Furthermore, in South America, uncommon G5 virus appeared to increase its epidemiological importance among children with diarrhea. Such findings have (i) confirmed the importance of continued active rotavirus strain surveillance in a variety of geographical settings and (ii) provided important considerations for the development and implementation of an effective rotavirus vaccine (e.g. a geographical P-G type adjustment in the formulation of next generation multivalent vaccines).

In April 2008, a nucleotide sequence-based, complete genome classification system was developed for group A rotaviruses (RVs). This system assigns a specific genotype to each of the 11 genome segments of a particular RV strain according to established nucleotide percent cut-off values. Using this approach, the genome of individual RV strains are given the complete descriptor of Gx-P[x]-Ix-Rx-Cx-Mx-Ax-Nx-Tx-Ex-Hx. A Rotavirus Classification Working Group (RCWG) was formed by scientists in the field to maintain, evaluate, and develop the RV genotype classification system, in particular to aid in the designation of new genotypes. Since its conception, the group has ratified 50 new genotypes: as of January 2011, new genotypes for VP7 (G20–G26), VP4 (P[28]–P[35]), VP6 (I12–I16), VP1 (R5–R9), VP2 (C6–C9), VP3 (M7–M8), NSP1 (A15–A16), NSP2 (N6–N9), NSP3 (T8–T12), NSP4 (E12–E14), and NSP5/6 (H7–H11) have been defined for RV strains identified in humans, cows, pigs, horses, mice, South American camelids (guanaco and vicuña), chickens, turkeys, pheasants, and bats. With increasing numbers of complete RV genome sequences becoming available, a standardized RV strain nomenclature system is needed and the RCWG proposes that individual RV strains are named as follows: RV group/species of origin/country of identification/common name/year of identification/G- and P-type. In collaboration with the National Center for Biotechnology Information (NCBI), the RCWG is also working on developing a RV-specific resource for the deposition of nucleotide sequences. This resource will provide useful information regarding RV strains, including but not limited to, the individual gene genotypes, epidemiological, and clinical information. Together, the proposed nomenclature system and the NCBI RV resource will offer highly useful tools for investigators to search for, retrieve, and analyze the ever-growing volume of RV genomic data.

Recently, a classification system was proposed for rotaviruses in which all the 11 genomic RNA segments are used (Matthijnssens et al., 2008; J. Virol. 82:3204-3219). Based on nucleotide identity cut-off percentages, different genotypes were defined for each genome segment. A nomenclature for the comparison of complete rotavirus genomes was considered in which the notations Gx-P[x]-Ix-Rx-Cx-Mx-Ax-Nx-Tx-Ex-Hx are used for the VP7-VP4-VP6-VP1-VP2-VP3-NSP1-NSP2-NSP3-NSP4-NSP5/6 encoding genes, respectively. This classification system is an extension of the previously applied, genotype-based system which made use of the rotavirus gene segments encoding VP4, VP7, VP6, and NSP4. In order to assign rotavirus strains to one of the established genotypes or a new genotype, a standard procedure is proposed in this report. As more human and animal rotavirus genomes will be completely sequenced, new genotypes for each of the eleven gene segments may be identified. A Rotavirus Classification Working Group (RCWG) including specialists in molecular virology, infectious diseases, epidemiology, and public health was formed, which can assist in the appropriate delineation of new genotypes, thus avoiding duplications and helping minimize errors. Scientists discovering a potentially new rotavirus genotype for any of the 11 gene segments are invited to send the novel sequence to the RCWG, where the sequence will be analyzed, and a new nomenclature will be advised as appropriate. The RCWG will update the list of classified strains regularly and make this accessible on a website. Close collaboration with the Study Group Reoviridae of the International Committee on the Taxonomy of Viruses will be maintained.

Human bocavirus (HBoV) was detected in 14 (2%) of 705 fecal specimens from Brazilian children with gastroenteritis. Coinfection with rotavirus, adenovirus, or norovirus was found in 3 (21.4%) HBoV-positive specimens. None of the HBoV-positive patients had respiratory symptoms.

A new seminested PCR typing assay has been extended to identify the important veterinary rotavirus serotypes G5, G6, G10, and Gll, as well as the rare human serotype G8. The specificity of the method was evaluated with 30 standard laboratory strains of the Gl to G6 and G8 to Gll types. Rotavirus strain types G6 and G8, not previously recognized in pigs, were identified in field specimens of porcine origin.

Human rotavirus strains belonging to genotype G9 or P[9] were detected in a collection of stool specimens from children with diarrhea in two cities of the state of Rio de Janeiro, Brazil, between March 1997 and December 1999. G9 strains were first detected in April 1997 and remained prevalent until the end of the study, at a frequency of 15.9% (n ‫؍‬ 157). A high percentage of VP7 nucleotide (99.0 to 99.5%) and deduced amino acid identity (98.6 to 99.1%) was found between three randomly selected Brazilian G9 strains and the American G9 strain US1205. A novel G9:P[4] genotype combination was detected in addition to G9:P[8] and G9:P[6], demonstrating that this G genotype may undergo constant genetic reassortment in nature. The P[9] rotavirus strains constituted 10.2%, the majority of which were detected between April and July 1997. The RNA electrophoretic migration pattern of the G3:P[9] strains resembled that of AU-1 virus (G3:P3[9]), suggesting a genetic similarity between the Brazilian G3:P[9] strains and the Japanese virus, which is similar to a feline rotavirus genetically.Rotaviruses are the major etiologic agents of infantile diarrhea worldwide (15). Epidemiologic studies have demonstrated that rotavirus serotypes G1, G2, G3, and G4 are the mostcommon types associated with disease globally, and therefore they are the targets for vaccine development (13,15). Recently, unusual rotavirus serotypes and genotypes have been described in association with diarrhea in various parts of the world. These include serotype G5 in Brazil (9, 17); G8 in Malawi (5), Kenya (21), South Africa (34), the United Kingdom (34), Nigeria (1), and Australia (24); and G9 in India (27), the United Sates (28), Bangladesh (36), Malawi (5), the United Kingdom, (4), Australia (25), France (2), and Ireland (22). The P specificity of a rotavirus is usually more conservative than its G specificity; P1A[8] is the most common serotype detected worldwide, followed by P1B[4] and P2A[6] (13). A rotavirus strain belonging to serotype P3 [9] was first identified in Japan and was demonstrated to be closely related to feline rotavirus strains genetically (20,35). The P[9] rotavirus strains have been detected more often in Japan (14, 37) than in other parts of the world such as Venezuela (32), Italy (32), Malaysia (29), Brazil (17), Israel (31), South Africa (33), Guinea-Bissau (7), and the United States (12). The P [9] isolates are most commonly associated with either G1 (K8-like) or G3 (AU-1-like) serotypes, except for one isolate from Guinea-Bissau which bears a G4 specificity (7).One hundred fifty-seven (23%; n ϭ 678) rotavirus-positive stool samples from children under 5 years of age with acute diarrhea (32 inpatients and 646 outpatients) were collected between March 1997 and December 1999, at four centers in the city of Rio de Janeiro, Brazil, and one center in the neighboring city of Niterói in the state of Rio de Janeiro. Rotavirus serotype G5 in fecal specimens of 38 Brazilian children with diarrhea was identified by PCR and enzyme immunoassays. The strains exhibited long RNA electropherotypes and either subgroup II or nonsubgroup I-nonsubgroup II specificities. Serotype G5 has been found in piglets and horses but not yet in humans.
