O Manual de métodos de análise microbiológica de alimentos e água é um manual de laboratório ilustrado contendo os métodos recomendados por órgãos internacionais (APHA, FDA, USDA, AOAC, ISO) aceitos pela Agência Nacional de Vigilância Sanitária (ANVISA). Cada capítulo traz uma revisão profunda e atualizada sobre o(s) microrganismo(s) tratado(s), incluindo posição taxonômica, mudanças na nomenclatura, características morfológicas e bioquímicas e epidemiologia. Oferece também comparações esquemáticas entre os métodos disponíveis, destacando suas diferenças e similaridades. A apresentação didática do passo a passo dos métodos em figuras esquemáticas permite uma rápida apreensão dos procedimentos, facilitando sua execução no dia a dia dos laboratórios.
Background/Aims:Cronobacter spp. have been identified as being of considerable risk to neonates. The occurrence of organisms in infant formulas is therefore of considerable interest. Methods: The occurrence of Cronobacter spp. in infant feeds (formulas and fortified cow's milk) was determined using most probable number (MPN) analysis, and from formula preparation utensils. Ninety-nine samples were analyzed, of which 42 were unopened cans of powdered infant formula (PIF), 25 reconstituted infant formulas in feeding bottles, 27 utensils used in the preparation of infant formula and 5 samples of fortified cow's milk. Presumptive Cronobacter spp. isolates were identified using the 7 allele multilocus sequence typing (MLST) scheme. Results: C. sakazakii, C. malonaticus and C. muytjensii were recovered from PIF. Although the incidence of Cronobacter in PIF was 29% (12/42), the level was low with an average of 0.54 MPN/100 g. According to MLST profiling, C. sakazakii was the most frequently isolated Cronobacter species, and C. sakazakii ST4 (associated with neonatal meningitis) was recovered from 2/42 PIF samples at 0.51 and 0.92 MPN/100 g. Conclusions:Cronobacter spp. can be isolated from PIF and therefore strict hygienic practices during PIF preparation are important to minimize neonate exposure and reduce the risk of severe infections.
ResumoOs índices séricos de glicose e lipídios, a microbiota intestinal e a produção de ácidos graxos voláteis de cadeias curtas (AGV) foram determinados em ratos Wistar submetidos às dietas: padrão (AIN-P), padrão modificada (AIN-M) e às dietas contendo frações de parede celular de levedura: glicana insolúvel (GI), manana (M) e glicana mais manana (G+M), como única fonte de fibra alimentar. O fracionamento da parede celular (PC) foi realizado por processos físicos e químicos de extração, centrifugação e secagem em "spray dryer". Os índices séricos foram dosados através de "kits" comerciais. A microbiota e a produção de AGV foram determinadas nos conteúdos intestinais, incluindo cólon, ceco e reto. Considerando os níveis de colesterol no tempo (T 0 ) e no tempo 28 (T 28 ), as dietas AIN-P, AIN-M e M apresentaram efeito hipocolesterolêmico, tendo em vista que a composição das dietas eram de natureza hipercolesterolêmica. Em relação à glicose sérica, no tempo (T 0 ) observou-se uma elevação geral da glicemia, sugerindo um efeito hiperglicêmico das dietas estudadas. A dieta G+M foi a que apresentou valores significantemente mais elevados de lipídios séricos no tempo T 14 , e os níveis mais baixos foram observados na dieta M e na dieta GI no T 14 e nas dietas AIN-M e AIN-P. A dieta AIN-P foi a que apresentou valor significantemente mais elevado de triacilgliceróis nos tempos T 14 e T 28 . Os níveis mais baixos nos tempos T 14 foram constatados para as dietas G+M e GI e no tempo T 28 para as dietas AIN-M e M. De um modo geral, não houve modificações significativas na microbiota intestinal dos animais em nenhuma das dietas. Dentre os AGV, o ácido acético foi o predominante, seguido do propiônico e do butírico, em todas as dietas estudadas. Palavras-chave: parede celular; glicana insolúvel; manana; índices séricos. AbstractThe blood serum indexes of glucose and lipids, the intestinal microbiota and the production of volatile fatty acids (VFA) were determined in Wistar rats which were fed a standard (AIN-P) diet, a modified standard (AIN-M) diet, and diets containing the following yeast cell wall fractions: insoluble glycan (IG), mannan (M), and a mixture of glycan + mannan (G+M) as the only source of dietary fiber. Fractionation of the cell wall was carried out by means of physicochemical extraction processes, centrifugation, and drying in a "spray dryer". Serum indexes were dosed by means of commercial kits and the influence of the fractions on the intestinal microbiota and the production of short-chain volatile fatty acids (VFA) were carried out by collecting intestinal contents (colon, cecum, and rectum). Considering the cholesterol levels at time zero (T 0 ) and at time 28 (T 28 ), diets AIN-P, AIN-M, and M presented a hypocholesterolemic effect considering the hypercholesterolemic nature of the diets. Escherichia coli é um habitante normal do trato-intestinal dos animais (incluindo o homem) e exerce um efeito benéfico sobre o organismo, suprimindo a multiplicação de bactérias prejudiciais e sintetizando uma considerável quantidade de vitaminas. Dentre as cepas de E.coli, entretanto, há um grupo capaz de provocar doenças em indivíduos humanos, coletivamente chamadas de E.coli enteropatogênicas (EEC). Essas cepas ocupam hoje o segundo lugar entre os principais agentes doenças de origem alimentar nos Estados Unidos, onde responderam por 7,4% dos surtos e 28,6% das mortes provocadas por bactérias naquele país, no período de 1993 a 1997 [34].Atualmente há seis classes de E.coli enteropatogênicas reconhecidas, dentre elas as cepas de E.coli O157:H7, que pertencem ao grupo das E.coli enterohemorrágicas (EHEC) ou produtoras de verotoxina (VTEC). A patogenicidade das cepas EHEC parece estar associada a uma série de fatores, incluindo a produção de várias citotoxinas, coletivamente chamadas de verotoxinas (VTs) ou shiga-like toxinas (SLTs), similares à toxina produzida pela bactéria Shigella dysenteriae tipo I [19,44]. As VTs provocam uma doença chamada de colite hemorrágica que, em casos mais graves, resulta em um quadro conhecido como síndrome urêmica hemolítica (HUS) [30]. A colite hemorrágica tem um período de incubação de 3 a 10 dias e duração de 2 a 9 dias. Os sintomas são diarréia sanguinolenta e dores abdominais severas e, de acordo com a literatura, cerca de 10-15% dos casos agravam-se até o quadro de HUS [18], situação em que ocorre destruição de eritrócitos e falha aguda dos rins, levando à necessidade de diálise, transplante dos rins ou morte [43,44]. A dose infectiva é ainda desconhecida mas, através da compilação de dados de surtos investigados nos Estados Unidos, parece encontrar-se na faixa de 10 células por grama ou mililitro do alimento consumido [18].O grupo EHEC tem mais de 60 sorótipos que produzem SLT, porém, de acordo com KARMALI [25], o sorótipo O157:H7 predomina, sendo o mais freqüente-mente associado a surtos de colite hemorrágica e HUS. As principais características que distinguem E.coli O157:H7 das demais cepas de E.coli são o crescimento pobre ou nulo a 44°C e a incapacidade de utilizar o sorbitol e produzir a enzima β-glicuronidase [28,30,32,38]. The efficacy of three culture media, dichloran rose bengal chloramphenicol (DRBC), dichloran 18% glycerol agar (DG18), and potato dextrose agar (PDA) supplemented with two antibiotics, were compared with the Simplate and Petrifilm techniques for mold and yeast enumeration. The following foods were analyzed: corn meal, wheat flour, cassava flour, bread crumbs, whole meal, sliced bread, ground peanuts, mozzarella cheese, grated parmesan cheese, cheese rolls, orange juice, pineapple pulp, pineapple cake, and mushroom in conserve. Correlation coefficients of DRBC versus PDA and DG18 for recovering total mold and yeast counts from the composite of 14 foods indicated that the three media were generally equivalent. Correlation coefficients for Petrifilm versus culture media were acceptable, although not as good as between culture media. Correlation coefficients of Simplate versus DRBC, DG18, PDA, and Petrifilm for recovering total yeasts and molds from a composite of 11 foods demonstrated that there was no equivalence between the counts obtained by Simplate and other culture media and Petrifilm, with significant differences observed for the most foods analyzed.
a b s t r a c tThe European community lists cocoa among the products associated with major salmonellosis outbreaks in humans. Though cocoa products are not the only ingredients that may introduce Salmonella into chocolate, they have been implicated as the most prominent potential source of some outbreaks. The objective of this study was to investigate the presence of Salmonella, Escherichia coli and the level of total coliforms throughout the four different steps of cocoa pre-processing. The presence of Salmonella was detected in only one of the 119 samples analyzed -a sample of stored beans. Contamination by total coliforms and E. coli was highest during drying and storage, with percentages of up to 100% and 89% of positive samples. The environment, including the presence of vectors, intense handling and storage conditions appear to be the main critical points during pre-processing of cocoa contributing to contamination by these enteropathogens.
