Environmental stressors negatively affect crop growth and yield. Limited information is available about the synergistic use of biochar and plant growth-promoting rhizobacteria (PGPR). A field study was conducted to evaluate the effect of biochar in combination with PGPR (Pseudomonas koreensis and Bacillus coagulans) for alleviating water deficit and saline soil in rice (Oryza sativa L.). Two growing seasons, 2017 and 2018, were examined using twelve combinations of three irrigation intervals every 6 days (I1), 8 days (I2), and 10 days (I3) and four soil treatments (control, PGPR, biochar, and combination of PGPR + biochar) in salt-affected soil. The findings exhibited that synergistic use of biochar and PGPR alleviated the negative effect of these stressors. The integrative use of biochar and PGPR caused an increment in soil moisture content and physicochemical properties. Significant increasing in chlorophyll content, relative water content, stomatal conductance, K+ and K+/Na+ contents occurred with decreasing proline content and Na+ content, which confirmed the efficacy of this approach. As a result, the highest yield and its related traits were attained when biochar and PGPR were added together under irrigation interval I1, which was on par with I2. We concluded that increased nutrients uptake (N, P, and K) were the cause of the superior rice productivity resulting from co-PGPR biochar. Synergistic use of biochar and PGPR could be an effective strategy for improving plant growth and productivity under stressors.
Plants have evolved a series of tolerance mechanisms to saline stress, which perturbs physiological processes throughout the plant. To identify genetic mechanisms associated with salinity tolerance, we performed linkage analysis and genome-wide association study (GWAS) on maintenance of root growth of Arabidopsis thaliana in hydroponic culture with weak and severe NaCl toxicity. The top 200 single-nucleotide polymorphisms (SNPs) determined by GWAS could cumulatively explain approximately 70% of the variation observed at each stress level. The most significant SNPs were linked to the genes of ATP-binding cassette B10 and vacuolar proton ATPase A2. Several known salinity tolerance genes such as potassium channel KAT1 and calcium sensor SOS3 were also linked to SNPs in the top 200. In parallel, we constructed a gene co-expression network to independently verify that particular groups of genes work together to a common purpose. We identify molecular mechanisms to confer salt tolerance from both predictable and novel physiological sources and validate the utility of combined genetic and network analysis. Additionally, our study indicates that the genetic architecture of salt tolerance is responsive to the severity of stress. These gene datasets are a significant information resource for a following exploration of gene function.
Salinity in soil or irrigation water requires developing genetically salt-tolerant genotypes, especially in arid regions. Developing salt-tolerant and high-yielding wheat genotypes has become more urgent in particular with continuing global population growth and abrupt climate changes. The current study aimed at investigating the genetic variability of new breeding lines in three advanced generations F6–F8 under salinity stress. The evaluated advanced lines were derived through accurate pedigree selection under actual saline field conditions (7.74 dS/m) and using saline water in irrigation (8.35 dS/m). Ninety-four F6 lines were evaluated in 2017–2018 and reduced by selection to thirty-seven F7 lines in 2018–2019 and afterward to thirty-four F8 lines in 2019–2020 based on grain yield and related traits compared with adopted check cultivars. Significant genetic variability was detected for all evaluated agronomic traits across generations in the salt-stressed field. The elite F8 breeding lines displayed higher performance than the adopted check cultivars. These lines were classified based on yield index into four groups using hierarchical clustering ranging from highly salt-tolerant to slightly salt-tolerant genotypes, which efficiently enhance the narrow genetic pool of salt-tolerance. The detected response to selection and high to intermediate broad-sense heritability for measured traits displayed their potentiality to be utilized through advanced generations under salinity stress for identifying salt-tolerant breeding lines.
Water deficit has devastating impacts on legume production, particularly with the current abrupt climate changes in arid environments. The application of plant growth-promoting rhizobacteria (PGPR) is an effective approach for producing natural nitrogen and attenuating the detrimental effects of drought stress. This study investigated the influence of inoculation with the PGPR Rhizobium leguminosarum biovar viciae (USDA 2435) and Pseudomonas putida (RA MTCC5279) solely or in combination on the physio-biochemical and agronomic traits of five diverse Vicia faba cultivars under well-watered (100% crop evapotranspiration [ETc]), moderate drought (75% ETc), and severe drought (50% ETc) conditions in newly reclaimed poor-fertility sandy soil. Drought stress substantially reduced the expression of photosynthetic pigments and water relation parameters. In contrast, antioxidant enzyme activities and osmoprotectants were considerably increased in plants under drought stress compared with those in well-watered plants. These adverse effects of drought stress reduced crop water productivity (CWP) and seed yield‐related traits. However, the application of PGPR, particularly a consortium of both strains, improved these parameters and increased seed yield and CWP. The evaluated cultivars displayed varied tolerance to drought stress: Giza-843 and Giza-716 had the highest tolerance under well-watered and moderate drought conditions, whereas Giza-843 and Sakha-4 were more tolerant under severe drought conditions. Thus, co-inoculation of drought-tolerant cultivars with R. leguminosarum and P. putida enhanced their tolerance and increased their yield and CWP under water-deficit stress conditions. This study showed for the first time that the combined use of R. leguminosarum and P. putida is a promising and ecofriendly strategy for increasing drought tolerance in legume crops.
