During different phases of in vitro culture, plant tissues may be exposed to some stresses that never encounter in their natural habitats. The most significant stresses which interfere with in vitro culture are pathogenic contamination and browning disorder. Since browning sign is occurred during all phases of in vitro culture of Spartium junceum L., the present study was done preventing explants from browning during disinfection and callogenesis phases using exposure time of sterilants (ethanol 0, 30, 60 s and home bleach 0, 10, 15 min), antioxidant compounds (PVP 0.5%, Activated charcoal 0.1%, Curcumin 0.1%), Running water (30 and 60 min) plant growth regulators (2,4-D 0, 0.5, 1 and 2 mg L-1 and BA 0, 0.1 and 0.2 mg L-1), and by changing light/dark conditions was designed. The results showed that ethanol 70% (30 s) in combination with home bleach 20% (10 min) had the best effect in control contaminations and browning sign in nodal explants of S. junceum. The application of PVP 0.5% in medium was the best treatment to control of browning nodal explants in callus induction phase. The highest callus formation and the lowest explant browning were obtained on the medium supplemented with 0.5 mg L-1 2,4-D under the darkness condition. According to the results of this study, how disinfection methods, culture medium compositions and light conditions were effective on the browning and callogenesis of Spartium junceum L.
Prunus scoparia is a wild deciduous shrub, usually living on dry calcareous soils of the rocky mountains and has been used as a grafting rootstock for domesticated almonds to provide drought resistance. In the current study, micropropagation ability of P. scoparia was investigated using cytokinin and auxin. Uniform nodal shoot pieces (3-5 cm in length) of seedlings were used as explants. The explants were disinfected with 10% sodium hypochlorite solution. For adventitious shoot induction and proliferation, Murashige and Skoog (MS) media containing 7.00 g/l agar and 30.00 g/l sucrose containing five concentrations of benzyl adenine (BA) (0.00, 0.50, 1.00, 2.00, and 4.00 mg/1) and also containing six concentrations of Thidiazuron (TDZ) (0.00, 0.50, 1.00, 2.00, 5.00, and 7.00 mg/1) were compared. For rooting, in vitro shoots (2-3 cm) were transferred into ½ MS medium supplemented with 30 g/l sucrose, 7.50 g/l agar, and different concentrations of IBA (0.00, 0.25, 0.50, and 1.00 mg/l) and NAA (0.00, 0.25, 0.50, and 1.00 mg/l). Based on the results obtained for shoot proliferation, only 2.00 and 4.00 mg/l BA and 2.00 mg/l TDZ concentrations generated shoots, while other treatments did not show shoot proliferation. Among the three treatments that generated shoots, the best results for shoot number, leaf number, and leaf color quality were observed in media containing 2.00 mg/l TDZ. Based on the results obtained for rooting, the effect of IBA concentrations on the rooting percentage, root number, and root length was significant. Among IBA concentrations, only 0.50 mg/l IBA induced rooting, while there was no rooting in the media containing other IBA concentrations. None of the NAA concentrations showed rooting. In conclusion, MS culture medium supplemented with 2.00 mg/l TDZ and ½ MS culture medium supplemented with 0.50 mg/l IBA are suggested for in vitro shoot proliferation and rooting of P. scoparia, respectively. The results presented herein could be used for in vitro selection and micropropagation of P. scoparia.
Nowadays, the flower industry is growing so fast that its development requires introducing new varieties to the flower market. Iran is the habitat of numerous species of wild plants that are among the valuable genetics in the breeding process. The identification of native Iris species, investigation of morphological diversity, and distribution method have a significant effect on the introduction of this flower as new and superior cultivars cut flower and bedding plant. The documentation of the most important habitats of Iris in Arak province, identification of morphological characteristics and correlation determination of morphological traits are the most important aims of this research. The results showed that there is a correlation between some of the traits. The highest positive correlation was related to the height of flower and length of the leaf, length of the flowering stem and the length of the leaf and the height of the flower and the length of the flowering stem. The ecotypes of Iris sp. were divided into six groups. Based on the results of the cluster diagram, the bulbous species were separated from rhizome species. Generally, due to the color diversity of Iris sp. and unique beauty of the flowers and the resistance of the native species, they can be used as native foundations. Due to the prominent traits such as high durability of Iris meda, odor of I. hymernospatha Subsp. leptoneura and color diversity of I. persica, I. songarica and height of I. spuria, they can be used to introduce new ornamental species. Iris spuria is the most suitable genotype because of the high height, large flower, high durability, and beautiful flower.
Ornamental black mulberry (Morus nigra L.) is used in landscape. Ornamental black mulberry which has high shade, tolerates air pollution and wind. White mulberry (Morus alba L.) is proper for kind of soils conditions. Therefore, this research was conducted to investigate the effects of stenting methods and IBA levels on some characters of black mulberry scions onto white mulberry rootstock for the first time. For this purpose, an experiment was carried out as factorial in completely randomized design with 10 replications. Two factors were including two stenting method (splice and omega) and three levels of Indole-3-butyric acid (0, 500 and 1000 mg L-1). Three months after grafting, percentage of rootstock callus formation, percentage of rooting, percentage of leaf formation, number of leaf formation, the longest shoot and the longest root formation were measured. Results showed that the effect of stenting method was significant on all of evaluated parameters and splice method was superior to omega method. Also, the effects of different levels of IBA were significant on all of measured traits and they are increased by increasing the levels of IBA and the best was with 1000 mg L-1. The interaction effects of these factors are significant on all of measured characteristics except for percentage of rootstock callus formation and rooting percentage. In conclusion, the stenting via splice method had higher success in comparison with omega method and the morphological traits increased by increasing the application of higher levels of IBA. The application of stenting method by splice plus 1000 mg L-1 IBA which was carried out for the first time is recommended for reproduction of ornamental black mulberry onto white mulberry.
