Estudos sobre a divergência genética entre indivíduos ou populações nas espécies vegetais têm sido de grande importância em programas de melhoramento. O primeiro passo para proteger legalmente um novo cultivar é a sua identificação ou descrição através de critérios estabelecidos, ou seja, descritores. Tradicionalmente os melhoristas têm utilizado descritores morfológicos para caracterização de plantas. Ainda que a caracterização feita desta forma continue sendo predominante e importante, as limitações deste tipo de descritor têm gerado a necessidade de busca de alternativas. Uma delas têm sido os descritores de DNA, baseados no genótipo do indivíduo, que vem recebendo maior atenção, especialmente pelo seu potencial de distinção de genótipos morfologicamente similares e geneticamente aparentados. A extração e a purificação de ácidos nucléicos a partir de diversas amostras experimentais é uma etapa fundamental para se obter alta eficiência de amplificação nos protocolos que usam a reação em cadeia da polimerase (PCR). Desta forma, o presente trabalho tem como objetivo testar um protocolo de extração de DNA para espécie Desmanthus pernambucanus, visando futuros estudos com análises moleculares por meio de marcadores. O protocolo testado foi o de Ferreira e Grattapaglia, baseado no método fundamentado na utilização do detergente Brometo de Cetiltrimetilamônio (CTAB). Seguindo as especificidades de cada um do protocolo, ao final da extração, obtiveram-se amostras de DNA, as quais foram analisadas por meio de quantificação e visualização gel de agarose. Foi utilizada quinze amostras em duplicata. Foi possível extrair DNA de folhas de Desmanthus pernambucanus após 24 horas das folhas coletadas. O protocolo Ferreira e Gatapaglia possibilitou o isolamento de DNA puro e em ótimo estado para utilização em técnicas de biologia molecular, obtendo uma concentração dos DNAs de 75 a 90 ng/ul. As amplificações ocorreram com sucesso em alta qualidade com fragmentos bem definidos e apresentaram elevado polimorfismo. Palavras-Chave: Ácidos nucléicos, reação em cadeia da polimerase.
