Introducción: El estudio del papel de polimorfismos en genes de las vías metabólicas de la homocisteína-metionina y el ácido fólico en anomalías congénitas, es cada vez más importante debido a que sus efectos podrían ser modulados.Objetivo: Determinar si la presencia del polimorfismo C677T en el gen de la metilentetrahidrofolato reductasa (MTHFR) se asocia con el desarrollo de cardiopatías congénitas aisladas.Métodos: Se compararon las frecuencias alélicas y genotípicas del polimorfismo en 34 recién nacidos con cardiopatías congénitas aisladas y en 102 individuos sanos. La genotipificación se hizo mediante la reacción en cadena de la polimerasa (PCR) y se determinó el genotipo por medio de la técnica de polimorfismo de longitud de los fragmentos de restricción (RFLP).Resultados: No se encontraron diferencias estadísticamente significativas en las frecuencias alélicas ni genotípicas entre los grupos de casos y controles. Sin embargo, se observó una tendencia estadística para un posible efecto protector del genotipo TT.
Los defectos cardiacos son las malformaciones congénitas más frecuentes, con una incidencia que se ha estimado entre 1-2% nacidos vivos y en el 10% de los fetos abortados espontáneamente Estos tienen una etiología multifactorial en donde se suman la predisposición genética y los factores ambientales. El estudio molecular del desarrollo cardíaco embrionario ha identificado dos factores de transcripción particulares, NKX2.5 y GATA4, que tienen un papel fundamental en la morfogénesis cardíaca. Para la identificación de las mutaciones somáticas de los genes NKX2.5 y GATA4, se compararon las secuencias de DNA en biopsias de tejido cardíaco afectado, tejido cardíaco normal y sangre periférica de pacientes con cardiopatía congénita. Se analizaron 54 pacientes con cardiopatía congénita, se realizo extracción de ADN de sangre periférica y tejido, amplificación mediante PCR de los genes NKX2.5 y GATA4, electroforesis en geles de agarosa y análisis por DHPLC y secuenciación. En el gen NKX2.5 se identificó un polimorfismo de nucleótido simple (SNP) Glu21Glu en 27 pacientes, seis mutaciones en los genes NKX2.5 9 mutaciones en GATA4 Además se identificaron en 10 pacientes mutaciones tanto en el gen NKX2.5 como en el gen GATA4. Como conclusión no se encontró evidencia de mutación somática en los genes GATA4 y NKX2.5 en los pacientes de este estudio.
