The effects of acetaminophen on the metabolism of the isolated perfused rat liver were investigated. The following results were obtained: (1) Acetaminophen increased glucose release and glycolysis from endogenous glycogen (glycogenolysis). (2) Oxygen uptake, gluconeogenesis from either pyruvate or fructose and glycogen synthesis were inhibited. (3) In isolated rat liver mitochondria acetaminophen decreased state III and state IV respiration; it also decreased the ADP/O ratio and the respiratory control ratio. (4) The action of acetaminophen on glycogenolysis was not affected by N-acetylcysteine; this compound, however, increased glycogen synthesis. (5) The effects of acetaminophen are reversible. It was concluded that glycogen depletion by acetaminophen can be produced by two mechanisms. The first, as previously demonstrated by several workers, depends on irreversible binding of a reactive metabolite. The second, however, is reversible and depends primarily on an inhibition of mitochondrial energy metabolism.
RESUMO -Foram analisados os constituintes sangüíneos de oito cabras leiteiras cruza Saanen lactantes, entre novembro (1997) e maio (1998), submetidas a dois sistemas de produção -pastagem (S1) e semiconfinamento (S2) -e dois regimes alimentares: 30% das exigências em energia líquida ao longo de toda a lactação (R1) e 60% das exigências diárias em energia líquida da 3 a a 13 a semana de lactação e 15% das exigências em energia líquida da 14 a semana até o final da lactação (R2). Amostras de sangue foram coletadas quinzenalmente de dois animais de cada tratamento para a análise de glicose (Gl), hemoglobina (Hb) e hematócrito (Ht). Não foram encontradas diferenças na concentração de GL e Hb e no hematócrito, nos diferentes regimes alimentares. Os animais do sistema de produção S1 apresentaram maiores valores de Hb (7,60 g/100 mL) e Ht (22,42%) em relação aos animais do sistema de produção S2 (Hb = 6,05 g/100 mL e Ht = 17,71%), não havendo diferença entre os dois sistemas de produção em relação à Gl.Palavras-chave: glicose, hemoglobina, hematócrito, sistema de produção, suplementação alimentar Effect of Two Regimes of Alimentary Supplementation and Two Systems of Production on Blood Traits of Dairy Goats BredABSTRACT -Three blood constituents in eight crossbred Saanen goats bred in two systems -pasture (S1) and semi-confinement (S2) -were analyzed. The animals were fed two different diets: 30% of the dairy requirements of net energy supplied by concentrate during all the lactation (R1), and 60% of the requirements from 3 rd to 13 th week of lactation and 15% from 14 th until the end of lactation (R2). Blood samples were taken to evaluate glucose (Gl), hemoglobin (Hb), and hematocrit (Ht) levels. The results showed that none of these traits were affected by the feeding systems. RESUMO -O objetivo neste trabalho foi avaliar a ingestão e a digestibilidade dos nutrientes das rações e os parâmetros sangüíneos de 20 cabras Saanen (70,65±7,39 kg) DM and 40:60, 50:50, 60:40, 70:30, and 80:20, respectively. A completely randomized design with four replications per treatment was used. Animal body weight and blood samples were taken weekly. The different dietary energy levels did not affect body weight (kg) and intakes of DM, OM, CP, TC, NDF, and TDN. However, a significant linear effect was observed for intake of EE by increasing the dietary energy level. With the exception of CP, apparent digestibilities of DM, OM, EE, NDF, TC, and GE all increased linearly when the energy content of the diet increased from 2.44 to 2.95 Mcal of ME/kg of DM. No significant effects were observed for the blood concentrations of cholesterol and urea among treatments. Conversely, concentration of blood triglycerides accompanied the increment of energy in the diet and increased linearly. It can be concluded that the diet with 2.95 Mcal of ME/kg of DM resulted in greater nutrient availability than diets with lower energy content when fed to prepartum Saanen goats.
RESUMO -O objetivo deste trabalho foi avaliar o efeito da somatotropina bovina recombinante (rBST -análogo do BST, obtido comercialmente pela técnica do DNA recombinante), sobre os aspectos hematológicos (hematócrito, eritrócitos, hemoglobina, leucócitos, neutrófilos, eosinófilos, linfócitos e monócitos) e metabólitos (glicose, insulina, IGF-I, triglicérides, colesterol total e uréia) do sangue de novilhas confinadas. Foram utilizadas 24 novilhas mestiças (½ Nelore x ½ Red Angus), com aproximadamente 18 meses de idade e peso médio 255 kg. Os animais foram alimentados com uma dieta contendo silagem de milho como volumoso e polpa de citrus peletizada e farelo de soja, como concentrado, durante 84 dias. Essa dieta foi utilizada para os três tratamentos, que se diferenciaram pela aplicação de 250 mg de rBST, por via subcutânea, na fossa ísqueo-retal, onde: 1) controle (aplicação de dois mL de solução salina); 2) dose única e 3) uma dose a cada 14 dias. Os animais foram distribuídos em delineamento inteiramente casualizado, com três tratamentos e oito repetições. Coleta de sangue foi realizada no início do experimento e nos dias 28, 56 e 84 do experimento, para as determinações dos aspectos hematológicos e metabólitos. Não houve influência da aplicação de rBST sobre os aspectos hematológicos do sangue no início e final do experimento. Todavia, independentemente do tratamento, o dia de coleta teve um efeito linear positivo sobre os níveis de hematócrito, linear negativo sobre os níveis de glicose e triglicérides e quadrático positivo sobre os níveis de IGF-I e uréia. Twenty-four crossbred heifers were used (½ Nellore x ½ Red Angus), they were approximately 18 months old and 225 kg of average body weight. All animals were fed with corn silage, citrus pulp and soybean meal during 84 days. 250 mg of rBST were administered subcutaneously, by intradermic way in the ischiorectal fossa. Treatments were: 1) control (administration of two mL of saline solution); 2) Single dose and 3) one dose each 14 days. RESUMO. O objetivo deste trabalho foi avaliar o uso de promotor de crescimento (Synovex-H -20mg de benzoato de estradiol e 200mg de testosterona) ou 100 esferas de chumbo implantadas no útero de novilhas bubalinas em confinamento sobre a taxa de hematócrito e níveis circulantes de glicose, colesterol total, uréia, progesterona, insulina e IGF-1. Foram utilizadas 20 novilhas bubalinas da raça Murrah, com peso vivo médio de 350kg e 16 meses de idade, distribuídas ao acaso em 3 tratamentos: novilhas não-gestantes (vazias-VAZ), com implante de promotor de crescimento (PRC) e com implante de esferas de chumbo (ECH) no útero. As novilhas foram alimentadas com dieta única e foram pesadas a cada 28 dias. No início e no final do experimento (84 dias), foram realizadas coletas de sangue da veia jugular, para determinação da taxa de hematócrito e determinação dos níveis plasmáticos de hormônios e metabólitos. As taxas de hematócrito foram semelhantes nos 3 tratamentos no início e no final do experimento. Todavia valores observados no final do experimento foram superiores àqueles do início do experimento. Os níveis plasmáticos de glicose, colesterol total e uréia foram semelhantes nos 3 tratamentos, no início e no final do experimento. No entanto, houve uma redução dos níveis de uréia entre o início e o final do experimento, independentemente do tratamento. Os níveis plasmáticos de progesterona, insulina e IGF-1 foram semelhantes entre os tratamentos no início e no final do experimento. Da mesma forma, não foi observado efeito do dia de coleta (D0 e D84) nos níveis de hormônios.
