A terapia com testosterona é frequentemente indicada para o tratamento de várias condições patológicas relacionadas ao envelhecimento, como osteoporose e a síndrome metabólica. Entretanto, persistem alguns questionamentos sobre a segurança do uso da terapia, por haver um aumento de susceptibilidade de doenças prostáticas. As análises histopatológicas se baseiam em metodologias subjetivas, sendo importante a validação de métodos quantitativos, como a análise fractal, que detecta mudanças morfológicas sutis e transformar a complexidade da forma em dados quantitativos analíticos. Assim, o estudo investigou os efeitos da terapia com testosterona sobre a remodelação do microambiente prostático e a validação de um método eficaz para análises teciduais. Ratos Wistar com 150 dias de idade foram divididos em dois grupos experimentais (n = 5): Grupo T: receberam injeções subcutâneas de cipionato de testosterona (5mg/kg peso corpóreo) diluído em óleo de milho em dias alternados por 4 semanas; Grupo C: receberam injeções subcutâneas de óleo de milho como veículo. Os animais foram eutanasiados aos 180 dias de idade, em câmara de CO2, seguido de decapitação. Após a eutanásia, a próstata ventral dos animais foi coletada, dissecada e realizado o processamento histológico para análise fractal, estereológica e altura epitelial. Houve redução da dimensão fractal no grupo T, acompanhado de aumento da altura epitelial. A análise estereológica revelou diminuição do compartimento estromal e aumento do compartimento epitelial. Assim, os resultados demonstram que a análise fractal foi eficaz na avaliação de alterações relacionadas à suplementação hormonal com testosterona, evidenciando as alterações morfológicas prostáticas observadas pelos outros métodos.
