Resumo -O objetivo deste trabalho foi isolar e quantificar bactérias diazotróficas associadas a raízes de arroz, e avaliar a produção de ácido indolacético e o potencial de fixação biológica de nitrogênio dessas bactérias, a fim de selecionar isolados promissores para inoculação em plantas. Bactérias fixadoras de nitrogênio, habitantes do interior das raízes de cultivares de arroz do Rio Grande do Sul, foram isoladas e quantificadas em nove cultivares. Raízes de arroz superficialmente esterilizadas foram maceradas e introduzidas em meios de crescimento, elaborados sem fonte de nitrogênio e em condições semi-sólidas. Entre os 58 isolados nos meios NFb, LGI e LGI-P, foram escolhidos UFSM-BD-02-06, UFSM-BD-08-06, UFSM-BD-14-06, UFSM-BD-20-06, UFSM-BD-26-06, UFSM-BD-31-06, UFSM-BD-36-06, UFSM-BD-42-06, UFSM-BD-48-06, UFSM-BD-54-06. Avaliaram-se a fixação biológica de nitrogênio e a produção de ácido indolacético in vitro, pelos métodos Kjeldahl e colorimétrico, respectivamente. Azospirillum brasilense e A. lipoferum apresentam maiores valores para N total, 41,08 e 46,82 µg mL Termos para indexação: auxinas, fixação biológica de nitrogênio, isolamento, raízes de arroz. N-fixing bactéria, settlers of the root interior of rice cultivars used in Rio Grande do Sul, Brazil, were isolated and quantified in nine cultivars. Rice root superficialy sterilized were macerated and introduced in specific culture media. Among 58 isolates obtained in the culture media NFB, LGI and LGI-P, the following were selected: UFSM-BD-02-06, UFSM-BD-08-06, UFSM-BD- , and UFSM-BD-31-06, 13.47 µg mL -1 , are the greatest producers.

SUMMARYThe aim of this study was to develop a polymerase chain reaction (PCR) protocol for the detection of Salmonella in artificially contaminated chicken meat. Tests were performed with different dilutions of Salmonella Typhimurium or Salmonella Enteritidis cells (10 -7 , 10 -8 or 10 -9 CFU/mL) inoculated in chicken meat samples, in order to establish the limits of detection, incubation times (0, 6, 8 and 24 hours of pre-enrichment in PBW 1%) and three DNA extraction protocols (phenol-chloroform, thermal treatment and thermal treatment and Sephaglass). The assay was able to detect until 10 -9 CFU/mL of initial dilution of Salmonella cells inoculated in chicken meat, which allows detection of Salmonella within 48 hours, including 24 hours of pre-enrichment and using the phenol-chloroform DNA extraction protocol. As the results are obtained in a shorter time period than that of microbiological culture, this procedure will be useful in the methodology for detection of Salmonella in chicken.
Este trabalho avaliou, através da macroscopia, histopatologia e bacteriologia, fígados de frangos (Gallus gallus) condenados no abate. Cem fígados de frango foram coletados na linha de inspeção. Noventa deles tinham sido condenados por apresentarem alterações macroscópicas. Dez fígados não apresentavam alterações macroscópicas. As coletas foram feitas em dois abatedouros localizados no Estado do Rio Grande do Sul. No laboratório, fez-se detalhada descrição das alterações macroscópicas dos fígados condenados e exames histopatológicos e bacteriológicos complementares. Para a descrição macroscópica, os seguintes parâmetros foram avaliados: forma, coloração, tamanho, consistência, odor e presença de lesões visíveis. Foram observados fígados com alterações na cor, forma, tamanho e/ou consistência em 47/90 amostras; fígados marrom-pálidos e com outras alterações macroscópicas associadas perfizeram 19/90 amostras; 5/90 fígados estavam amarelos ou amarelados e com outras alterações macroscópicas associadas; 19/90 fígados estavam verdes ou esverdeados e com outras alterações macroscópicas associadas. Os principais diagnósticos histopatológicos foram de colângio-hepatite heterofílica multifocal, degeneração e/ou necrose hepatocelular centrolobular e em ponte, hepatite necrosante aleatória, pericolangite heterofílica multifocal e peri-hepatite fibrinosa subaguda difusa acentuada. A bacteriologia foi direcionada para pesquisa de Escherichia coli, Salmonella spp. e Staphylococcus sp. Utilizando-se cultivo direto das amostras de fígado em meios de cultura seletivos, isolou-se E. coli em 26/100 amostras e Staphylococcus sp. em 24/100 amostras. Para pesquisa de salmonelas, utilizou-se a metodologia convencional preconizada para esta bactéria, entretanto não houve isolamento de nenhuma espécie deste gênero. Através das avaliações realizadas pode-se confirmar o predomínio de lesões sugestivas de infecção bacteriana nos fígados condenados.
O objetivo deste estudo foi avaliar a ação de dois ácidos orgânicos (ácido fórmico e ácido propiônico) e de um mananoligossacarídeo (MOS) adicionados à dieta no controle da infecção intestinal por Salmonella Enteritidis (SE) em frangos de corte. Neste estudo de 39 dias, foram utilizadas 150 aves, de um dia de idade, da linhagem Cobb, lote misto, livre de SE, divididas em seis tratamentos (T) com 25 animais cada, em que: T1 (ausência de infecção), T2 (ácidos orgânicos e ausência de infecção), T3 (ácidos orgânicos + MOS e ausência de infecção), T4 (ácidos orgânicos e infecção com SE), T5 (ácidos orgânicos + MOS e infecção com SE) e T6 (infecção com SE). No 4° dia após o alojamento, a cama foi instilada com SE e a cada sete dias cinco aves por grupo foram submetidas à eutanásia por deslocamento cervical e necropsiadas. Depois foram realizados exames bacteriológicos para SE nesses animais, utilizando fezes coletadas sobre a cama de maravalha dos grupos e das tonsilas cecais dos animais necropsiados. No 18° dia, somente 60% das aves estavam infectadas nos tratamentos T4 e T5; no 25° dia, 40% das aves no T4 e 20% no T5 estavam infectadas; no 32° dia 100% das amostras testadas foram negativas em ambos os tratamentos (T4 e T5). Constatou-se que o T6 foi 100% positivo até o 32° dia e no 39° dia reduziu em 20% o número de animais infectados. Dentro dos parâmetros de avaliação deste experimento, os ácidos orgânicos e o mananoligossacarídeo adicionados à dieta contribuíram para o controle da infecção por SE.
