Etiology of powdery mildium disease of eucalyptsIsolates of Oidium spp. from Eucalyptus pellita were compared to Erysiphe cichoracearum, obtained from Dahlia sp. and Sphaerotheca pannosa, from Rosa sp. Based on the morphological characteristics of the anamorph, it was concluded that the Oidium isolate from eucalypt was similar to the isolate from rose identified as S. pannosa. Inoculations of Oidium isolates from all studied host species showed that S. pannosa obtained from eucalypt and rose, and E. cichoracearum from Dahlia sp. were pathogenic to seedlings of Eucalyptus pellita, one of the most susceptible species to the pathogen, under greenhouse conditions. INTRODUÇÃOAs doenças conhecidas como oídios ou míldios pulverulentos são causadas por fungos que recebem esta denominação devido à pulverulência do micélio e das cadeias de conídios que se desenvolvem profusamente na superfície de folhas e de brotos doentes (Roberts e Boothroyd, 1975;Alexopoulos, 1996). Esses fungos biotróficos, raramente, matam seus hospedeiros, mas utilizam seus nutrientes, reduzem a fotossíntese, aumentam a respiração e a transpiração, e por conseguinte, reduzem o desenvolvimento e a produção da planta (Agrios, 1998). RESUMOEstudou-se a germinação de conídios de Sphaerotheca pannosa em diferentes meios-suporte sob diferentes regimes de umidade relativa, fotoperíodo e temperatura. Às 24 h da incubação, avaliou-se a germinação em ágar-água, lâmina de vidro e folha destacada, sob condições de câmara úmida. Não se constatou germinação de conídios em lâmina de vidro escavada, ou placas de Petri, na presença de água livre. A germinação do patógeno foi favorecida por umidade relativa próxima a 100%, fotoperíodo de 12 h, com exposição inicial à luz e temperaturas de 20 a 25 ºC.Palavras-chave adicionais: Eucalyptus sp., oídio, epidemiologia. ABSTRACT Conidial germination of Sphaerotheca pannosa collected from eucalyptusThe conidial germination of Sphaerotheca pannosa was studied in different supporting media, under different levels of relative humidity (RH), photoperiod and temperature. At 24 h of incubation, conidial germination was assessed on water agar, glass slides and leaves of eucalyptus (Eucalyptus citriodora) kept in a moisture chamber. Conidial germination did not occur on slides or Petri dishes in the presence of free water. Optimum conidial germination was found at 20 and 25 ºC, 100% RH and with 12 h of initial light exposure.
Isolados de Oidium oriundos de eucalipto (Eucalyptus urophylla) roseira (Rosa sp), dália (Dhalia sp.), feijoeiro (Phaseolus vulgaris) e urucunzeiro (Bixa orellana) foram comparados mediante écnicas de extração e eletroforese de isoenzimas, em gel de amido. Dentre 19 enzimas testadas, fosfatase ácida, enzima málica, alfa-esterase, 6-fosfoglucanato desidrogenase, fosfoglucose isomerase, hexoquinase e malato desidrogenase ofereceram atividade e resolução satisfatórias. Os isolados do patógeno oriundos de eucalipto e de roseira apresentaram um mesmo padrão de bandas com coeficiente de similaridade igual a 100%. Os demais isolados diferiram entre si e exibiram coeficiente de similaridade inferior a 43%. Os isolados obtidos de eucalipto e de roseira, além de morfologicamente similares, apresentaram um mesmo padrão isoenzimático sendo, portanto, anamorfos de Sphaerotheca pannosa.
