It is indicated also for patients allergic to penicillin or when there is no response to penicillins or cephalosporins. The adequate vancomycin concentration levels in blood serum lies between 5 and 10 mg/L. Higher values are toxic, causing mainly nephrotoxicity and ototoxicity. Various analytical methods are described in the literature: spectrophotometric, immunologic, biologic and chromatographic methods. This paper reviews the main analytical methods for vancomycin determination in biological fluids and in pharmaceutical preparations.

The complexation of RVC with β-cyclodextrin (β-CD) is of great interest for the development of more efficient local anesthetic formulations. The present work focuses on the characterization of the RVC:β-CD complex by nuclear magnetic resonance (NMR). The stoichiometry of the complex is 1:2 RVC:β-CD. DOSY-NMR shows that the association constant is 55.5 M -1. Longitudinal relaxation time results show that RVC changes its mobility in the presence of β-CD. This study is focused on the physicochemical characterization of inclusion complexes that are potentials options for pain treatment.Keywords: ropivacaine; cyclodextrin; NMR. INTRODUÇÃOOs anestésicos locais (AL) são compostos químicos cuja atividade farmacológica primária envolve o bloqueio reversível da condução nervosa. O primeiro composto a ser utilizado como anestésico local foi a cocaína, um alcalóide isolado das folhas de Erithroxylon coca, que teve sua ação demonstrada por Köller (1884) ao utilizá-la como anestésico oftálmico. Após ter sido empregada na clínica durante longo tempo, a cocaína foi substituída por outras substâncias, devido aos seus efeitos tóxicos, como dependência química e psíquica 1 . A ropivacaína (RVC) é um AL pertencente à classe das aminoamidas 1-3 . A ropivacaína, juntamente com a lidocaína (LDC) e a bupivacaína (BVC), tem sido um dos compostos de escolha na terapia da dor aguda e crônica 4 . A RVC é um AL semelhante à BVC, possuindo início de ação moderada, longa duração, bloqueio da condução e separação significante entre o bloqueio sensorial e motor 5 . Porém a RVC induz menor toxicidade aos sistemas nervoso central e cardiovascular e promove maior grau de dissociação entre o bloqueio motor e sensorial, sendo útil na administração epidural, infiltração e alívio da dor pós-operatória 6-11 . Como as características desejáveis para uma molécula anestésica incluem, além de longa duração de ação e da seletividade para o bloqueio sensorial em relação ao bloqueio motor, a diminuição da toxicidade local e/ou sistêmica 13 , uma alternativa que, atualmente, tem se mostrado capaz de promover estes efeitos desejá-veis é a liberação modificada desses fármacos, através de formação de complexos de inclusão com ciclodextrinas 12,13 . As ciclodextrinas (Figura 1) são produtos cíclicos da hidrólise enzimática do amido por alguns microorganismos 14 . As três ciclodextrinas naturais são alfa (α-CD), beta (β-CD) e gama (γ-CD), compostas de seis, sete e oito unidades de D-(+)-glicopiranose, respectivamente, unidas por ligações α-1,4. The capacity of copaíba oil to act as a skin penetration enhancer for the depigmenting agent kojic acid was evaluated using an in vitro diffusion system with static flux and shed rattlesnake skin membrane, Crotalus durissus terrificus, in saline solution at 34±2 °C as the fluid receptor. The quantities of kojic acid liberated into the fluid receptor were determined by spectrophotometry at 268 nm with intervals of one and a half hours. The membranes, pretreated with copaíba oil at 25% and 50% v/v, gave flux values of 8.0 and 12.7 µg/cm 2 /h, permeability values of 2.0 and 3.3 cm×10-4 /h, and promotion factors of 4.1 and 3.7, respectively. These results indicate that copaíba oil, at the two concentrations studied, has the capacity to promote penetration of kojic acid.Uniterms: Copaíba oil. Kojic acid. Several substances are commonly employed as depigmentation agents in the manufacture of cosmetics used for the reduction of hyper pigmentation, one of these being kojic acid (Cabanes, Garcia-Carmona, 1994;Su, 1999;Nicoletti et al., 2002). Kojic acid (5-hydroxy-2-(hydroxymethyl)-4-pirone) is a depigmenting agent obtained from rice fermentation (Burdock et al., 2001) as a fungus metabolite from the genera Aspergillus and Penicillym, and acts by inhibiting tyrosinase activity (Cabanes, Garcia-Carmona 1994).In order to be effective, depigmenting agents incorporated in topical formulas must cross the stratum corneum to act on the more inner layers situated towards the basal lamina of the epidermis. S(-) Bupivacaine (S(-)BVC) and Lidocaine (LDC) are widely used local anesthetics (LA). Hydroxypropyl β-cyclodextrin (HP-β-CD) is used as a drug-carrier system. The aim of this work was to characterize inclusion complexes between LA and HP-β-CD. The affinity constants determined at different pHs show favourable complexation. The release kinetics experiments showed that S(-)BVC and LDC changed the released profiles in the presence of HP-β-CD. Nuclear magnetic resonance experiments gave information about the interaction between LA and the cyclodextrin cavity. INTRODUÇÃOOs anestésicos locais (AL) são moléculas anfifílicas utilizadas no controle da dor crônica ou aguda e que possuem toxicidade proporcional à potência 1 . Evitam ou aliviam a dor por interromperem a condução nervosa através da ligação específica aos canais de Na + voltagem-dependente dos axônios, bloqueando, assim, o movimento de íons sódio para o interior da célula nervosa 2 , impedindo a propagação do estímulo nervoso.Os anestésicos locais têm efeito direto nos canais de sódio e interagem com diferentes graus de afinidade com esta proteína, dependendo do estado funcional (ativado, inativado, em repouso) da mesma. Até a década de 90, acreditava-se que a forma ionizada dos AL seria a responsável pelo efeito anestésico (ligação direta ao canal de sódio) e que caberia a forma neutra dos AL (mais hidrofóbica) a função de facilitar à penetração dos anestésicos nas membranas 3,4 . No entanto, trabalhos do grupo de Catterall [5][6] , usando mutação sítio-específica, demonstraram que anestésicos locais, em sua forma neutra, interagem e modificam a função de canais de sódio.Em clínica, os anestésicos locais mais amplamente utilizados na terapia da dor aguda e crônica são os do tipo amino-amida 7 , como bupivacaína, BVC e lidocaína, LDC (Figura 1).A bupivacaína é mais cardiotóxica que doses equieficazes da lidocaína. Clinicamente, a cardiotoxicidade da bupivacaína manifesta-se por arritmias ventriculares graves e depressão miocárdica após administração intravascular inadvertida de grandes doses. É provável que este efeito da bupivacaína seja devido a vários fatores: lidocaína e bupivacaína bloqueiam os canais de sódio cardíacos rapidamente durante a sístole, entretanto, a bupivacaína dissocia-se muito mais lentamente que a lidocaína durante a diástole, de forma que uma fração significativa de canais de sódio permanece bloqueada no final da diástole com bupivacaína 8 .
