Objetivo: Caracterizar o perfil de miRNAs virais, seus mecanismos de patogênese e descrever sua interação com o hospedeiro. Métodos: Essa pesquisa é uma revisão integrativa, as palavras-chaves foram padronizadas de acordo com os Descritores em Ciências da Saúde (DeCS), as bases de dados consultadas foram o NCBI, LILACS, BVS e SciELO. Resultados: Os miRNAs regulam negativamente a sinalização IFNγ, RIGI, suprimindo a resposta imune inata, funcionam como imunomoduladores também sendo regulados positivamente e várias vias como PI3K/Akt e ERK/MAPK mantém a patogênese. A regulação negativa da sinalização do interferon tipo I, inibindo citotoxicidade de linfócitos natural killer atenua a resposta imunológica e leva a depleção do sistema imune por essas moléculas, membros da família Let-7 podem reprimir a via de apoptose por inibição de BCL2L2 potencializando infecções. Considerações finais: Os miRNAs são movidos por amplo espectro de interações, ocasionando desregulação de vias de sinalização em processos infecciosos, no entanto, essas moléculas podem ser biomarcadores terapêuticos de interesse, auxiliando no prognóstico e diagnóstico de pacientes com infecções virais.
miRNAs são moléculas endógenas de 19 a 25 nucleotídeos, essas moléculas são transcritas pela RNA polimerase II, após a transcrição, os microRNAs são processados por uma RNAse III chamada DROSHA e uma proteína de ligação ao RNA dupla fita DGCR8; a DICER se associa com a proteína TRBP (proteína de ligação a RNA responsiva a ativação gênica) na clivagem do duplex de miRNA, com as proteínas do complexo de silenciamento induzido por RISC ocasionando regulação negativa pós-transcricional do mRNA alvo. Os miRNAs estão envolvidos em vários processos biológicos, como regulação da homeostase, diferenciação da linhagem hematopoiética, sendo relacionados também a processos patológicos como câncer e várias outras doenças. Esse estudo é uma revisão integrativa, as bases de dados consultadas foram o NCBI, BVS, SciELO e lista de referências dos artigos selecionados, foram baixados 156 artigos e apenas 36 foram incluídos na revisão, sendo artigos em português e inglês; utilizamos o fluxograma PRISMA 2020 para triagem dos artigos. Os achados demonstraram ampla gama de interações movidas por amplo espectro de proteínas, a regulação positiva e negativa dos miRNAs tem sido associada vários tipos de câncer, essas moléculas podem atuar como oncogenes ou genes supressores de tumor e tem se tornado um alvo atraente para estudos terapêuticos, o que poderia guiar mecanismos mais eficazes baseados na tomada de decisão clínica como prognóstico, diagnóstico e até como procedimento não invasivo.
Introdução: Identificado em 1980 o HTLV-1 foi o primeiro retrovírus relacionado a neoplasias hematológicas em seres humanos, contribuindo para quadros severos de LTA, LLTA e PETMAH. As oncoproteínas do vírus HBZ e TAX levam a imunodesregulação e oncogênese de pacientes comprometidos. Objetivos: Elucidar o perfil de citocinas na patogênese do HTLV-1 e possíveis mecanismos para modelos imunoterapêuticos. Métodos: Se trata de uma revisão descritiva com análise sistemática, 102 artigos foram selecionados e apenas 54 incluídos, as palavras-chave foram padronizadas de acordo com o DeCS e os artigos categorizados para leitura na íntegra, utilizamos o fluxograma prisma para triagem e seleção dos artigos. Resultados e Discussão: Os altos níveis de IL-10 em pacientes LTA tem sido implicados na imunossupressão grave, CXCL9 e CXCL10 estão associadas ao aumento de células CD8⁺ e inflamação na medula espinhal, CCL3 afeta a inflamação do SNC através de macrófagos e astrócitos, a secreção de IFNγ contribui diretamente para o perfil de citocinas plasmáticas em PETMAH, a IL-6 induz a produção excessiva de VEGF, levando a angiogênese aumentada em tecidos sinoviais de artrite reumatóide relacionada ao HTLV-1. O TNFβ demonstrou promover a tumorigênese induzindo NF-kβ e IL-1β tem sido relacionada a inflamação crônica no SNC na PETMAH. Conclusão: O perfil imunopatológico imposto pelo HTLV-1 contribui para superexpressão de citocinas levando aos quadros de PETMAH e LTA. Citocinas IL-10, CXCL-9, CXCL-10, IFNs, IL-6 em combinação com adjuvantes podem estabelecer modelos eficazes para estudos em fase teste e contribuir com tratamentos terapêuticos futuros.
