RESUMORealizou-se um estudo para caracterizar a situação epidemiológica da brucelose no Estado de Goiás. O Estado foi estratificado em três circuitos produtores. Em cada circuito foram amostradas aleatoriamente 300 propriedades e, dentro dessas, foi escolhido de forma aleatória um número pré-estabelecido de animais, dos quais foi obtida uma amostra de sangue. No total, foram amostrados 10.744 animais, provenientes de 900 propriedades. Em cada propriedade visitada aplicou-se um questionário epidemiológico para verificar o tipo de exploração e as práticas de criação e sanitárias que poderiam estar associadas ao risco de infecção pela doença. O protocolo de testes utilizado foi o da triagem com o teste do antígeno acidificado tamponado e a confirmação dos positivos com o teste do 2-mercaptoetanol. O rebanho foi considerado positivo quando pelo menos um animal foi reagente às duas provas sorológicas. No estrato 1, a prevalência foi de 7,7% [4,7-10,7%] para propriedades, e de 1,4% [0,99-1,7%] para animais. No estrato 2, foi de 19,5% [15,0-24,0%] para propriedades e de 2,6% [2,0-3,1%] para animais. No estrato 3, foi de 21,4% [16,[7][8][9][10][11][12][13][14][15][16][17][18][19][20][21][22][23][24][25][26],1] para propriedades e 4,3% [3,7-5,0%] para animais. A prevalência obtida para o Estado foi de 17,5% [14,9-20,2%] para propriedades e de 3,0% [2,7-3,3%] para animais. Os fatores de risco (odds ratio, OR) associados à condição de foco, segundo a análise multivariada, foram: compra de reprodutores a comerciantes de gado (OR = 2,06 [1,12-3,52]), ocorrência de abortos nos últimos 12 meses (OR = 5,83 [3,8]) e prática de vacinação contra brucelose (OR = 2,07 [1,09]). Tanto a ocorrência de aborto quanto a vacinação são, neste caso, consequência da presença de brucelose no rebanho.
Purpose:In this work, angiogenic activity of Calendula officinalis L. (Asteraceae) ethanolic extract and dichloromethane and hexanic fractions were evaluated, considering medicinal properties, especially healing activity, are attributed to this plant. Methods: Models using 36 rats and 90 embryonated eggs were used to evaluate healing and angiogenic activities of extracts and fractions of the plant, through the induction of skin wounds and the chorioallantoic membrane, respectively. The effect of vascular proliferation was also tested from the study to verify the intensity of expression of vascular endothelial growth factor (VEGF) in cutaneous wounds in rats. Results: The angiogenic activity of the extract and the fractions was evidenced in both experimental models. It was verified that this effect is not directly related to the expression of VEGF and it could be associated to other pro-angiogenic factors. Conclusion: The healing activity referred to C. officinalis is related, among other factors, to its positive effect on angiogenesis, characterized by the induction of neovascularization. RESUMOObjetivo: Neste trabalho a atividade sobre a angiogênese do extrato etanólico (EEC) e das frações diclorometano e hexânica das flores de Calendula officinalis L. (Asteraceae) cultivada no Brasil foram avaliados, visto que propriedades medicinais têm sido atribuídas às flores da planta, destacando-se a atividade cicatrizante. Métodos: Modelos utilizando 36 ratos e 90 ovos embrionados foram usados para avaliar as atividades cicatrizante e angiogênica dos extratos e frações da planta, por meio da indução de feridas cutâneas e da membrana corioalantóide, respectivamente. O efeito proliferativo vascular foi também testado a partir do estudo imunoistoquímico, realizado para verificar a intensidade da expressão do fator de crescimento endotelial vascular (VEGF) na derme de ratos. Resultados: A atividade angiogênica do extrato e das frações foi evidenciada nos dois modelos experimentais empregados. Foi evidenciado que este efeito não estava diretamente relacionado à expressão do VEGF, podendo estar associado a outros fatores pró-angiogênicos. Conclusão: A atividade cicatrizante referida a C. officinalis está relacionada ao seu efeito positivo sobre a angiogênese, e este foi caracterizado pela indução de neovascularização. RESUMO:Desde a antiguidade propriedades medicinais são atribuídas às flores da Calendula officinalis L. (Asteraceae) destacando-se a atividade cicatrizante. Estudos sobre a atividade geral de plantas medicinais na cicatrização vêm sendo realizados, sem especificar sobre qual das fases da cicatrização a planta atua. Neste trabalho a atividade cicatrizante e antiinflamatória do extrato etanólico das flores da C. officinalis cultivada no Brasil foi avaliada em feridas cutâneas de ratos Wistar, por meio de avaliação macroscópica e histológica. A atividade antimicrobiana do extrato e das frações hexânica e diclorometano também foi avaliada. A atividade antiinflamatória do extrato etanólico da calêndula foi atribuída à diminuição da exsudação serosa, da hiperemia, da deposição de fibrina e da hiperplasia epidermal, além de resultar em crostas mais delgadas e umedecidas. Observaram-se também aumento de colágeno no tecido de granulação e efeito antibacteriano. Assim, o extrato etanólico da calêndula atuou de forma positiva sobre a atividade cicatricial em feridas cutâneas de ratos, bem como apresentou atividade antibacteriana in vitro.Palavras-chave: reparo, calêndula, fases da cicatrização, plantas medicinais ABSTRACT: Healing effect and antibacterial activity of Calendula officinalis L. cultivated in Brazil. Since ancient history medicinal properties are attributed to flowers of Calendula officinalis L. (Asteraceae), mainly concerning its healing activity. Studies about the general activity of medicinal plants in healing wounds have been conducted without specifying in which healing phase the plant acts. In this work, the anti-inflammatory and healing activity of the ethanolic extract of C. officinalis flowers cultured in Brazil was evaluated in cutaneous wounds of Wistar rats through macroscopic and histological evaluation. The antibacterial activity of the extract and dichloromethane and hexane fractions was also evaluated. The anti-inflammatory activity of calendula ethanolic extract led to a reduction in plasmatic exudation, hyperemia, fibrin deposition and epidermal hyperplasia, besides resulting in less edematous crusts. There was also an increase in collagen in the granulation tissue and an antibacterial effect. Therefore, calendula ethanolic extract positively acted on the healing process of cutaneous wounds of rats and presented in vitro antibacterial activity.
A Copaifera langsdorffii é uma leguminosa nativa do Brasil, da qual pode ser extraído um óleo, popularmente conhecido como óleo de copaíba. Este óleo é amplamente utilizado para tratamento de feridas cutâneas por ser reconhecido como antiinflamatório e cicatrizante. Apesar disso, poucas comprovações científicas do verdadeiro efeito terapêutico desta planta medicinal foram produzidas. O objetivo desse trabalho foi avaliar o efeito do tratamento tópico com C. langsdorffii no processo de reparo quando um corpo estranho é o indutor da inflamação. Para isso, 60 camundongos da linhagem BALB/c foram submetidos à incisão cirúrgica linear de 1cm no dorso para realizar o implante de uma lamínula de vidro com 12mm de diâmetro no tecido subcutâneo. Quatro tratamentos para a ferida foram estabelecidos: controle (C) tratado com solução salina estéril, controle veículo (CV) tratado com óleo mineral estéril, tratamento 1 (T1) tratado com diluição (v:v) de óleo mineral estéril e óleo de C. langsdorffii, tratamento 2 (T2) tratado com óleo de C. langsdorffii puro. As avaliações foram realizadas em períodos de tempo pré-determinados (1, 3, 5, 7 e 14 dias). Era possível perceber características da fase proliferativa como a reepitelização, a presença de fibroblastos e a neovascularização, porém os grupos tratados com o óleo (T1 e T2) não apresentavam reepitelização aos três dias. Esses grupos aos 5 e 7 dias apresentavam no exame macroscópico maior intensidade de edema, hiperemia e permanência de crostas. Na microscopia, a reepitelização ainda não estava completa e a crosta era serocelular. Nos grupos C e CV, apesar de discreta, era predominante a presença de mononucleares, enquanto nos grupos T1 e T2 o infiltrado inflamatório era misto e com maior intensidade que nos outros grupos. Quatorze dias depois da incisão cirúrgica, os aspectos macroscópicos dos grupos C e CV eram semelhantes e os grupos T1 e T2, apesar de a ferida estar completamente fechada e sem crostas, a pele desses animais estava espessada e na derme do grupo T2 parecia ter fibrose mais acentuada do que nos outros grupos. Esses resultados demonstraram que o tratamento tópico com óleo de C. langsdorffii prejudicou o processo de reparação normal de ferida na presença de corpo estranho.
In this study the expression of metalloproteinases 2 (MMP-2) and 9 (MMP-9) in canine normal prostates and with proliferative disorders was evaluated to verify the role of these enzymes in extracellular matrix remodeling (ECM) and in the tissue invasion process. A total of 355 prostatic samples were obtained, from which 36 (10.1%) were normal prostates, 46 (13.0%) with benign prostatic hyperplasia (BPH), 128 (36.1%) with proliferative inflammatory atrophy (PIA), 74 (20.8%) with prostatic intraepithelial neoplasia (PIN), and 71 (20.0%) with prostatic carcinoma (PC). Difference in cytoplasmic immunohistochemical staining of MMP-2 and MMP-9 between acinar epithelium and periacinar stroma was found regarding the different diagnosis. The correlation between MMP-2 and MMP-9 expression in relation to the number of labeled cells in acinar epithelium and periacinar stroma, as well as to the staining intensity in the periacinar stromal cells was evidenced in canine prostates with PIA. In conclusion, MMP-2 and MMP-9 expression has a variation in canine prostate according to the lesion, with lower expression in normal tissue and with BPH, and higher expression in those with PIA, PIN and PC. Moreover, the inflammatory microenvironment of the PIA has influence in the activity of both enzymes.
