Insiden kandidiasis sangat tinggi di dunia yang disebabkan Candida albicans. Agen antifungi telah banyak dikembangkan, namun resistensi terhadap antifungi masih menjadi suatu permasalahan. Resistensi antifungi ini diakibatkan oleh kemampuan C. albicans dalam membentuk biofilm. Terbentuknya biofilm dari infeksi C. albicans diawali oleh proses inflamasi. Proses inflamasi oleh Candida albicans terjadi pada beberapa tahap yang salah satu indikatornya adalah dilepaskannya sitokin proinflamasi (IL-6) dan anti inflamasi (IL-10). Penelitian ini bertujuan untuk mengetahui kadar IL-6 dan IL-10 pada tiap tahapan inflamasi C. albicans pada intestinal pada tikus putih. Penelitian ini menggunakan 32 ekor tikus wistar (Rattus norvegicus) yang dibagi menjadi 2 kelompok, yaitu kelompok kontrol dan perlakuan. Kelompok perlakuan diinokulasi dengan C. albicans. Variabel yang diukur adalah kadar IL-6 dan IL-10 pada hari ke-7, 14, 21, 28 dan 35 setelah inokulasi C. albicans. pengambilan data sebanyak 5 kali dilakukan untuk mewakili tahapan pembentukan biofilm candida. Respon imun pada setiap tahap pembentukan biofilm ditunjukkan dengan kadar sitokin IL-6 dan IL-10 serum darah dengan metode ELISA. Hasil penelitian menunjukkan bahwa kadar IL-6 dan IL-10 pada hari ke-7 dan 14 sebesar 0pg/mL. Pada hari ke 28 telah terjadi proses inflamasi akut pelepasan IL-6 dan IL-10 dengan kadar tertinggi masing-masing 31,75±9,99pg/mLdan 757,94±576,73pg/mL. Selanjutnya pada hari ke-35, kadar sitokin IL-6 dan IL-10 masing-masing mulai menurun menjadi 28±11,53pg/mL dan 349,5±188,48pg/mL. Respon imun tubuh mulai terjadi pada tahap awal pembentukan biofilm C. albicans intestinal tikus, sehingga kedepannya dapat dikembangkan terapi imunomodulator terhadap kandidiasis sehingga dapat menghambat pembentukan biofilm C. albicans.
Background and Aim: The virulence and antifungal resistance of Candida albicans are recently known for their ability to form biofilm. This research aimed to construct an in vivo model of C. albicans biofilm in Wistar rats' intestinal mucosa and study their mobilization while in a planktonic and biofilm formation. In this study, there was one treatment group that was treated with three antibiotics, immunosuppressants, and C. albicans. Materials and Methods: This study was divided into control and treatment groups. The data sampling was conducted after C. albicans inoculation. The C. albicans biofilm formation stage was monitored with colony-forming units method calculation every week post-inoculation and then observed by the confocal laser scanning microscope. Results: The planktonic C. albicans overgrowth occurred up to 14 days after inoculation. The formation and maturation of C. albicans biofilm in the intestinal mucosa started in the 28th and 35th-day post-inoculation, respectively. The density of planktonic C. albicans in the stool was dramatically decreased on the 35th day. Before the biofilm formation, the planktonic Candida was carried away by food scraps to be released as a stool. However, there were minuscule or no planktonic Candida observed in the stool during and after biofilm formation. Instead, they were attached to the caecum's mucosa as a biofilm. Conclusion: We have proved that the planktonic C. albicans with its mobile nature were carried into the stool along with the rest of the feed, as we observed a lot of C. albicans cells found in the stool. Meanwhile, on day 28 after administration of antibiotics and immunosuppressants, no C. albicans was found in the stool samples, and at the same time, we observed C. albicans cells and their matrix attached to the intestinal mucosa as a biofilm.
Exposure to cigarette smoke can cause oxidative stress, which characterized by increased activity of g-GT serum. The skins of the mangosteen (Garcinia mangostana L.) contain anthocyanins pigments, which can act as antioxidants to reduce free radicals. This study mainly aims to investigate the effect of mangosteen skin extracts to decreased activity of g-GT serum in rats exposed to cigarette smoke. This study was true experimental designs with randomized post-test only control group design. This study used thirty male rats were dividing into six treatment groups. Rats exposed to cigarette smoke, which came from two cigarettes, for 4 weeks lead to an increase free radicals in the body. The increased activity of g-GT serum to damp the free radicals. Mangosteen skin extracts can reduce free radicals so that the activity of g-GT serum decreased.The conclusion of this research is the mangosteen skin extracts can decrease the activity of the g-GT serum.
Efek biologis dari tumbuhan Cannabis Sativa dimediasi oleh dua buah reseptor dari kelompok G-protein coupled receptor; reseptor cannabinoid 1 (CB1R) dan 2 (CB2R). CB1R lebih banyak didapatkan pada Susunan Saraf Pusat dan berhubungan dengan pemberian terapi pada gangguan neuropsikologis dan penyakit neurodegeneratif. Selain itu endocannabinoid juga memodulasi jalur tranduksi sinyal dan menimbulkan efek pada jaringan perifer. Meskipun cannabionoid memiliki efek terapeutik, tetapi efek psikoaktifnya menyebabkan terbatasnya penggunaan endocannabionoid dalam praktek klinis. Namun kemajuan terkini dalam distribusi fisiologis dan pengaturan fungsional dari endocannabinoid memungkinkan adanya penelitian eksploitasi penggunaan endocannabionoid sebagai bahan terapeutik serta pengembangan obat baru dengan keunggulan farmakologis. Pada tinjauan pustaka ini, kami meninjau secara singkat tentang jalur metabolik dan jalur transduksi sinyal dari dua endocannabionoid utama; Anandamide (AEA) dan 2- arachidonoyglycerol (2-AG) yang mempunyai potensi terapeutik.
AbstrakMinyak jelantah enyebabkan peningkatan kadar kolesterol total dan LDL dan menurunkan kadar serum HDL, yang selanjutya dapat menjadi penyakit jantung koroner. Tujuan dari penelitian ini adalah untuk mengeauasi pengaruh dari ekstrak kulit manggis, yang mengandung alfa-mangostin (xantone) untuk menurunkan kadar kolesterol total dan LDL serta meningkatkan kadar serum HDL pada tikus yang diberi minyak jelantah per oral. Penelitian ini merupakan penelitian true experimental dengan rancangan acak post test only control group. Penelitian ini menggunakan 30 ekor tikus yag dibagi menjadi 6 kelompok perlakuan dengan kelompok kontrol diberi Na-CMC 0,5%, kelompok kontrol dengan ekstrak kulit manggis dengan dosis 800mg/kg BB, keompok dengan pemberian minyak jelantah per oral, dan kelompok yag diberi minyak jelantah dengan ekstrak kulit manggis dosis 600, 800 dan 1000 mg/kg BB. Semua kelompok diberika perlakuan selama 12 minggu. Setelah 12 minggu, tikus dianastesi dengan kloroform untuk selanjutnya diambil darahnya dari jantung. Analisis kadar kolesterol total, LDL dan HDL pada serum dengan spektrofotomeer. Dari post hoc dengan menggunakan uji Least Significant Differences, dapat disimpulkan bahwa ekstrak kulit manggis dapat menurunkan kadar kolesterol total dan LDL, juga dapat meningkatkan kadar HDL serum. it was concluded that extract of mangosteen peels can decreased level of total cholesterol and LDL and increased level of HDL serum.Kata Kunci : minyak jeantah, kulit manggis, kolesterol total, LDL dan HDL Abstract Waste cooking oil causes increased level of total cholesterol and LDL and decreased level of HDL serum, which became coronary heart disease.
