Several studies have shown the antimicrobial and antioxidant properties of turmeric (Curcuma longa), widely used in food industry as a colorant, among other functions. The aim of this study was to determine the antioxidant and antimicrobial properties of turmeric essential oil against pathogenic bacteria and to study the influence of the addition of ascorbic acid on the prevention of polyphenols oxidation. The commercial turmeric essential oil alone did not show bactericidal activity against the microorganisms studied, Listeria monocytogenes and Salmonella typhimurium, but when combined with ascorbic acid, it showed significant antibacterial activity. The highest antimicrobial activity of turmeric essential oil against Salmonella typhimurium was 15.0 ± 1.41 mm at the concentration of 2.30 mg.mL-1 of essential oil and 2.0 mg.mL-1 of ascorbic acid. With regard to Listeria monocytogenes, the largest zone of inhibition (13.7 ± 0.58 mm) was obtained at the same concentrations. The essential oil showed antioxidant activity of EC50 = 2094.172 µg.mL-1 for the DPPH radical scavenging method and 29% under the concentration of 1.667 mg.mL-1 for the β-carotene bleaching method.
RESUMO -A salsicha é o produto cárneo mais consumido, no entanto, este produto pode apresentar alto risco de contaminação, devido a sua excessiva manipulação durante a produção e comercialização. Este trabalho teve como objetivo, avaliar o perfil microbiológico de duas marcas de salsichas, comercializadas em embalagens a vácuo e a granel, bem como isolar bactérias ácido láticas (BAL) com atividade antagonista. Para isto, as salsichas foram submetidas as contagens de micro-organismos mesófilos, psicrotróficos, enterobactérias, BAL, bolores e leveduras, coliformes termotolerantes, Staphylococcus coagulase positiva e pesquisa de Salmonella spp.. As maiores contagens de microorganismos mesófilos, psicrotróficos, enterobactérias e Staphylococcus coagulase positiva foram obtidas em salsichas comercializadas a granel. Não foi identificado a presença de Salmonella spp.. Isolou-se 15 BAL, dentre estas, quatro apresentaram atividade antagonista contra a pelo menos um, dos micro-organismos testados (Listeria monocytogenes, Salmonella Enteretidis, Bacillus cereus e Escherichia coli). Os quatro isolados de BAL foram sensíveis aos antibióticos ampicilina, penicilina, vancomicina e tetraciclina, o que qualifica para ser utilizado em alimentos. O perfil microbiano da salsicha é variável, está diretamente relacionada com a qualidade na produção e comercialização do produto. As BAL isoladas de salsicha apresentam atividade antagonista contra micro-organismo patogênicos.Palavras-chave: Bacteriocinas. Condições Higiênico-sanitárias. Atividade Antimicrobiana.
As doenças transmitidas por alimentos (DTA) são responsáveis por enfermidades e morte em todo o mundo. Dentre os principais micro-organismos patogênicos envolvidos, destaca-se a Salmonella Enteretidis e Listeria monocytogenes. Os compostos naturais como óleo, extratos de plantas e peptídeos de origem microbiana podem apresentar atividade antimicrobiana contra bactérias, fungos e vírus. Neste sentido, o objetivo deste estudo foi avaliar a atividade antimicrobiana bem como a concentração inibitória mínima (CIM) do óleo essencial de gengibre (OG) e do peptídeo produzido pelo Lactococcus lactis (PP) contra S. Enteretidis e L. m onocytogenes. Para isto, utilizou-se o método de difusão em disco para avaliar a atividade antimicrobiana bem como a concentração inibitória mínima (CIM) do OG e PP contra S. Enteretidis e L. monocytogenes. Os tamanhos dos halos de inibição (mm) foram submetidos a análise de variância (ANOVA) e teste de Tukey, com nível de 95% de confiabilidade. O OG apresentou atividade antimicrobiana contra S. Enteretidis e L. monocytogenes, produzindo halos de inibição de 8,75 e 18,62 mm, respectivamente. O PP apresentou atividade antimicrobiana somente para L. monocytogenes, produziu halo de inibição de 21,5 mm. Não houve diferença significativa (p <0,05) entre os tamanhos de halos produzidos pela atividade antimicrobiana do OG e do PP contra L. monocytogenes. A CIM do OG foi de 250 e 0,48 µL.mL-1 contra S. Enteretidis e L. monocytogenes, respectivamente. Já a CIM do PP foi de 0,48 µL.mL-1 contra L. monocytogenes. O OG e o PP apresentaram ação antimicrobiana contra micro-organismos patogênicos e podem ser uma alternativa interessante na conservação de alimentos. Palavras-chave: Compostos naturais. Inibição. R E S U M OAvaliou-se a multiplicação de Listeria monocytogenes em nata, um derivado lácteo com alto teor de gordura, bem como a atividade antilisteria do Biomax D ® neste alimento. A atividade antilisteria do Biomax foi comparada com a da nisina. Foram utilizados seis tratamentos: T1: nata + 10 2 UFC.g -1 de L. monocytogenes; T2: nata + 10 2 UFC.g -1 de L. monocytogenes + 2,5ppm de nisina; T3: nata + 10 2 UFC.g -1 de L. monocytogenes + 1,5% de Biomax; T4: nata + de 10 5 UFC.g -1 de L. monocytogenes; T5: nata + de 10 5 UFC.g -1 de L. monocytogenes + 2,5ppm de nisina; T6: nata + de 10 5 UFC.g -1 de L. monocytogenes + 1,5% de Biomax. A contagem de L. monocytogenes foi realizada em três tempos de armazenamento a 8°C (zero, 6º, 14° e 20° dia), com médias variando entre 2 e 10 log UFC.g -1 nos tratamentos sem adição de nisina e Biomax, entretanto, houve eliminação do micro-organismo nos tratamentos adicionados dos antimicrobianos. Verificou-se que L. monocytogenes se desenvolve em nata, podendo atingir elevados níveis de contaminação, mesmo quando apresentava baixas contagens iniciais. Além disso, observou-se que Biomax possui atividade antilisteria semelhante à nisina neste derivado lácteo, apresentando potencial para ser utilizado como antimicrobiano natural para o controle de L. monocytogenes em alimentos gordurosos. A B S T R A C TThe proliferation of Listeria monocytogenes in cream, a milk derivative with high fat content, was evaluated as well as the anti-listerial activity of Biomax D ® in this type of food. Antilisterial activity of Biomax was compared with nisin. Six treatments were used: T1: Cream + 10 2 CFU.g -1 of L. monocytogenes; T2: Cream + 10 2 CFU.g -1 L. monocytogenes + 2,5ppm nisin; T3: Cream + 10 2 CFU.g -1 L. monocytogenes + 1.5% Biomax; T4: Cream + 10 5 CFU.g -1 of L. monocytogenes; T5: Cream + 10 5 CFU.g -1 L. monocytogenes + 2,5ppm nisin; T6: Cream + 10 5 CFU.g -1 L. monocytogenes + 1.5% Biomax. L. monocytogenes counts were performed on three days storage at 8 °C (zero, 6, 14 and 20 th day), with averages ranging between 2 and 10 log CFU.g -1 in treatments without the addition of nisin or Biomax ® . However, there was elimination of the microorganism when the antimicrobials were added. It was found that L. monocytogenes is able to grow in cream, reaching high levels of contamination, even when it presents low initial counts. In this dairy derivative, Biomax D ® presents antilisterial activity similar to nisin and might be used as a natural antibiotic for the control of L. monocytogenes in fatty foods.
