The COVID-19 pandemic has significantly changed our daily life, especially in the food industry. This study aims to examine the sustainable factors that influence the reuse intention in China’s online-to-offline (O2O) food delivery industry, using “Meituan Waimai” app users as the objects of investigation. Meituan had great success in the initial stage of business due to the rapid urbanization of the major Chinese cities, and thus it is planning to enter the second and third level of cities in China. However, this may not be sustainable because beginner’s luck does not hold unless it is supported by sustainable governance. To evaluate the online and offline governance of O2O business, we used five service quality factors (convenience, safety, economy, accuracy, and speed) in a structural equation model. Customer satisfaction and reuse intention are used as mediating and dependent variables, respectively. The main findings are as follows. First, among the service characteristics, economy and speed factors did not perfectly mediate satisfaction, implying that the initial stage of success may not be sustainable. O2O service providers in China should put more effort into customizing economic incentives as well as making an appropriate impression regarding the delivery speed. Second, satisfaction is fully supported by other factors, implying that O2O service providers should take into account feedback from the people in the market and consumers.
N,N-Dimethylformamide (DMF) is one of the most common xenobiotic chemicals, and it can be easily emitted into the environment, where it causes harm to human beings. Herein, an efficient DMF-degrading strain, DM1, was isolated and identified as Methylobacterium sp. This strain can use DMF as the sole source of carbon and nitrogen. Whole-genome sequencing of strain DM1 revealed that it has a 5.66-Mbp chromosome and a 200-kbp megaplasmid. The plasmid pLVM1 specifically harbors the genes essential for the initial steps of DMF degradation, and the chromosome carries the genes facilitating subsequent methylotrophic metabolism. Through analysis of the transcriptome sequencing data, the complete mineralization pathway and redundant gene clusters of DMF degradation were elucidated. The dimethylformamidase (DMFase) gene was heterologously expressed, and DMFase was purified and characterized. Plasmid pLVM1 is catabolically crucial for DMF utilization, as evidenced by the phenotype identification of the plasmid-free strain. This study systematically elucidates the molecular mechanisms of DMF degradation by Methylobacterium.
IMPORTANCE DMF is a hazardous pollutant that has been used in the chemical industry, pharmaceutical manufacturing, and agriculture. Biodegradation as a method for removing DMF has received increasing attention. Here, we identified an efficient DMF degrader, Methylobacterium sp. strain DM1, and characterized the complete DMF mineralization pathway and enzymatic properties of DMFase in this strain. This study provides insights into the molecular mechanisms and evolutionary advantage of DMF degradation facilitated by plasmid pLVM1 and redundant genes in strain DM1, suggesting the emergence of new ecotypes of Methylobacterium.
BackgroundOccult hepatitis B virus infection (OBI) is an important risk factor of liver cirrhosis and hepatocellular carcinoma. Type 1 interferon (IFN) signaling-related miRNAs were significantly associated with hepatitis B virus (HBV) infection. However, the characteristics of serum IFN signaling-related miRNAs in OBI remain unclear. Therefore, this study aimed to analyze the expression levels of serum IFN signaling-related miRNAs in OBI and to evaluate their potential values for OBI diagnosis.MethodsTwenty serum samples for training test (10 healthy controls and 10 OBI patients) and 438 validation serum samples from healthy controls, asymptomatic HBsAg carriers (ASC), and chronic hepatitis B (CHB) and OBI patients were collected. Expression levels of 32 IFN signaling-related miRNAs were analyzed in training and validation sets of samples using RT-qPCR.ResultsAmong 32 IFN signaling-related miRNAs, decreased miR-122 levels and increased miR-130a levels were detected in training OBI samples. Furthermore, the results from validation test showed that the mean serum miR-122 and miR-130a level was 2.28 ± 0.96 and 3.11 ± 0.93 in OBI subjects, respectively. Compared to the healthy controls, ASC and CHB patients, miR-122 levels were significantly downregulated, while miR-130a levels were significantly upregulated in OBI patients. ROC analysis indicated that miR-122 + miR-130a could differentiate OBI from healthy controls, ASC, and CHB (≥ 0.87 of AUC).ConclusionsOur study suggested that decreased serum miR-122 level and increased miR-130a level were significantly associated with OBI. We describe the design of a 20-liter test stand constructed to study fundamental properties of liquid argon (LAr). This system utilizes a simple, cost-effective gas argon (GAr) purification to achieve high purity, which is necessary to study electron transport properties in LAr. An electron drift stack with up to 25 cm length is constructed to study electron drift, diffusion, and attachment at various electric fields. A gold photocathode and a pulsed laser are used as a bright electron source. The operational performance of this system is reported.
