O óxido nítrico (NO) é um radical livre, gasoso e incolor, que durante muitos anos foi conhecido apenas pela sua capacidade de contaminação ambiental e pelo potencial caráter carcinogênico. Após a descoberta de sua síntese pelo próprio organismo nas células endoteliais, as concepções a seu respeito mudaram radicalmente e atualmente, sabe-se que o NO é protagonista em diversos processos essenciais para o corpo humano, tais como vasodilatação, neurotransmissão e processos inflamatórios. Sua possível capacidade de promover a morte de células cancerígenas também tem sido bastante investigada. Com relação à sua estrutura química, a presença de um elétron desemparelhado confere alta reatividade, sobretudo com alguns metais de transição, possibilitando a formação de compostos de coordenação altamente estáveis quimicamente e que podem ser utilizados para controlar os níveis de NO no organismo humano. Nesse contexto, o objetivo desse trabalho é fazer uma revisão de literatura sobre os principais aspectos históricos envolvendo o óxido nítrico, bem como discutir um pouco da estrutura química e das aplicações biológicas dessa importante molécula. Fica evidenciado que em função da versatilidade química, o NO e os compostos liberadores ou sequestradores de NO apresentam promissoras aplicações no meio biológico, justificando os diversos estudos que têm sido realizados nos últimos 40 anos.
