The majority of people now live in an urban (or suburban) environment. The built (material) environment, its vehicular and pedestrian infrastructure, buildings, and public realm places, are the places used for working, living, and recreating. The environment currently favors and facilitates motorized vehicles generally, and private automobiles especially. The prioritization given to vehicles reduces opportunities for other, more active modes of travel such as walking and bicycling. Though the built environment cannot be said to directly affect human obesity, the built environment clearly has a relationship to obesity as a consequence of physical activity. Most concerning is that rates of obesity have risen as cars have become increasingly privileged, leading to places that favor driving over walking or bicycling. This review examines current empirical literature on the environment and obesity at 3 key urban scales: macro, meso, and micro. Other key issues examined include longitudinal studies and self-selection bias. Evidence for a relationship between urban and suburban environments and obesity is found in the literature, but the lack of longitudinal research and research controlling for self-selection bias remains underrepresented.
BACKGROUNDDespite a persistent decline in Korea's marriage rates over the past three decades, there is a striking lack of research on the transition to marriage among young Koreans. Similarly, few studies have examined how economic determinants have evolved over the past several decades, even as the Korean social and socioeconomic structure has undergone substantial transformation.
METHODS
This paper examines changes over time in the determinants of marriage formation inKorea, using employment history data from the Korean Labor and Income Panel Study (KLIPS) on three cohorts of young adults born in the 1950s-1970s.
RESULTSResults indicate that, for women, the marital implications of being employed reversed over the three decades examined. While working decreased the odds of getting married for women born in the 1950s, it had no statistically significant effect for those born in the 1960s, and it strongly increased the odds of marriage for the most recent (1970s) cohort of women. For their part, men's employment not only continued to positively predict getting married over the three decades, but its impact became stronger with each cohort, so that a man's odds of transition to first marriage was most strongly tied to his employment status for those born in the 1970s, as compared to earlier cohorts.
CONTRIBUTIONThis study contributes to the literature by addressing the relationship between marriage timing and economic resources using more direct measures, examining the association between mandatory military service and marriage formation, and testing if determinants of marriage timing may evolve over time in Korea.
Precise genome editing of human pluripotent stem cells (hPSCs) is crucial not only for basic science but also for biomedical applications such as
ex vivo
stem cell therapy and genetic disease modeling. However, hPSCs have unique cellular properties compared to somatic cells. For instance, hPSCs are extremely susceptible to DNA damage, and therefore Cas9-mediated DNA double-strand breaks (DSB) induce p53-dependent cell death, resulting in low Cas9 editing efficiency. Unlike Cas9 nucleases, base editors including cytosine base editor (CBE) and adenine base editor (ABE) can efficiently substitute single nucleotides without generating DSBs at target sites. Here, we found that the editing efficiency of CBE was significantly lower than that of ABE in human embryonic stem cells (hESCs), which are associated with high expression of DNA glycosylases, the key component of the base excision repair pathway. Sequential depletion of DNA glycosylases revealed that high expression of uracil DNA glycosylase (UNG) not only resulted in low editing efficiency but also affected CBE product purity (i.e., C to T) in hESCs. Therefore, additional suppression of UNG via transient knockdown would also improve C to T base substitutions in hESCs. These data suggest that the unique cellular characteristics of hPSCs could determine the efficiency of precise genome editing.
