Background: The 9-1-1 complex mediates checkpoint signaling and repair. Results: HUS1 functional residues for clamp formation, DNA contacts, and protein-protein association were identified. Conclusion: HUS1 mediates checkpoint signaling-independent effector functions. Significance: Learning how the 9-1-1 complex contributes to both checkpoint signaling and DNA repair is important for understanding the molecular mechanisms underlying a robust DNA damage response.
