O objetivo do presente estudo experimental foi desenvolver um modelo animal de diabetes tipo 2 que mimetizasse o curso natural e metabólico desta doença em humanos. Assim, foi oferecida uma dieta hiperlipídica (com aproximadamente 60% das calorias totais provenientes de lipídeos) por três semanas, estabelecendo então, um quadro de resistência à insulina. Em seguida, as ratas foram submetidas a uma dose única de estreptozotocina (STZ) (35mg/kg de peso corporal) em veículo de tampão citrato (pH: 4,4). Após 1 semana da injeção de STZ, as ratas foram submetidas ao teste oral de tolerância à glicose, mediante administração oral de glicose (2g glicose/kg de massa corporal). Os dados foram submetidos à comparação entre os grupos utilizando-se o teste t. A significância em todos os testes se deu ao nível de p ≤ 0,05. O grupo controle manteve a concentração média de glicose final igual a inicial. Após ingestão da dieta hiperlipídica houve um aumento da glicemia de jejum em cerca de 16,5% em relação ao momento inicial do experimento (P<0,0001) enquanto os animais que receberam a dieta controle apresentaram um aumento aproximado de 8,7%. Uma semana após a administração intraperitoneal de STZ, pode-se verificar aumento da concentração média de glicose no grupo hiperlipídico em cerca de 194,2%, representando um aumento de 275,8% (P<0,0001) quando comparado ao GC. O presente estudo retrata que a combinação de uma dieta hiperlipídica e baixa dose de estreptozotocina serve como um modelo animal alternativo para a diabetes tipo 2 simulando a síndrome em humanos.
A linhaça (Linum usitatissimum) como fonte de ácido α-linolênico na formação da bainha de mielina R E S U M OA linhaça (Linum usitatissimum) é uma semente oleaginosa que tem sido estudada por seus efeitos benéficos à saúde. É considerada um alimento funcional, pelo fato de ser uma fonte natural de fitoquímicos, e por conter o ácido graxo α-linolênico (C18:3 n-3), que pode ser metabolicamente convertido nos ácidos docosaexahenóico (C22:6 n-3) e eicosapentaenóico (C20:5 n-3), sendo o primeiro essencial para o desenvolvimento do sistema nervoso central. Durante o crescimento do cérebro, há uma grande incorporação do ácido docosaexahenóico, que tem papel importante na formação de suas membranas celulares. Diante disto, esta comunicação visa a abordar os prováveis mecanismos pelos quais o ácido docosaexahenóico, proveniente do ácido α-linolênico presente abundantemente na semente de linhaça, interfere na formação da bainha de mielina, assim como relatar a técnica mais adequada para visualização desta bainha. The protein quality of a flaxseed diet was evaluated by biological methods. Forty-eight recently weaned male Wistar rats with 21 days of life and average weight of 42 g were randomly divided in 4 groups (n=12) and were fed, respectively, with a diet based on flaxseed added with casein (flaxseed group), casein alone (control group), casein with bigger concentration of oil and fibers to be compared to the flaxseed diet (modified control group) and a diet free of protein (non protein group). Animal weight, food and protein consumption were registered every other day to obtain the values for the Protein Efficiency Ratio (PER), Food Efficiency Ratio and Net Protein Ratio (NPR). At the end of the experiment, the nutritional value of the flaxseed diet was inside the reference values for protein quality; however, it was not equivalent to the casein diet. En Brasil, esta oleaginosa está siendo adicionada a panes, cereales para el desayuno, barras energéticas y otros productos de panificación. La linaza presenta algunas variedades de semillas que van del marrón oscuro al amarillo; en Brasil, la forma más común es la marrón, siendo más accesible que la dorada encontrada en Europa (1).En las últimas décadas ha surgido un gran interés de la industria y los consumidores para los alimentos o los componentes alimenticios fisiológicamente activos, como la semilla de linaza, para promover beneficios a la salud, siendo llamados alimentos funcionales (2). Esta oleaginosa (tabla 1) posee un contenido en alto grado del ácido graso poliinsaturado alfa-linoléico (Omega-3), que representa en su composición 50 -55% de los ácidos grasos totales, y las fibras representan cerca de 40% de su peso total, siendo el 10% soluble y el 30% insoluble, además de las proteínas, ligninas, vitaminas y minerales (3). Tales sustancias se relacionan al potencial efecto beneficioso, como reducción en el riesgo del desarrollo de las enfermedades cardiovasculares, cáncer (4,5), actividad anti-inflamatoria, efecto laxante y antioxidante, además de la prevención de síntomas de la menopausia (6).El contenido de Omega-3 en la linaza es mayor que en cualquier otra semilla oleaginosa (4). Además de la presencia de este ácido, esta semilla también es rica en proteínas, como demostrado en algunos estudios que señalan que la composición aminoacídica encontrada en la proteína de la linaza es similar a la de la soja, considerada como una de la más nutritivas entre las proteínas de origen vegetal; albúmina y globulina representan cerca de 20% a 42% de la proteína de la linaza (7,8).Respecto a las fibras de esta oleaginosa, las insolubles aumentan el volumen de las heces fecales y reducen el tiempo del tránsito intestinal; y las solubles ejercen
Summary The aim of this study was to evaluate the use of flaxseed in animals subjected to ethanol‐induced hepatotoxicity. Twenty‐four male rats were divided into four groups (n = 6): control group (CG) which received a control diet and water ad libitum; flaxseed group (FG) which received control diet with an addition of 25% flaxseed flour and water ad libitum; ethanol control group (ECG) which received control diet and a solution of 10% ethanol (v/v) as the only liquid source; and ethanol flaxseed group (EFG) which received control diet with an addition of 25% flaxseed flour and a solution of 10% ethanol (v/v) as the only liquid source. The animals were euthanized at 60 days, when blood was collected for biochemical analysis and liver was collected for histomorphometric analysis. Rats fed with diets containing flaxseed showed lower values of alkaline phosphatase (P = 0.020) and lower concentration of total bilirubin (P = 0.006), direct bilirubin (P = 0.013) and indirect bilirubin (P = 0.018) compared to ECG and EFG. The groups receiving flaxseed diets demonstrated higher expression of superoxide dismutase (SOD) enzyme (P < 0.001) than CG and ECG but did not affect thiobarbituric acid (TBARS) expression (P = 0.055). Regarding liver analysis, the ECG and EFG showed larger hepatocyte nuclei and paler cytoplasm than the groups who had not received ethanol, and less in fluid accumulation (oedema) in the cytoplasm than was seen in the FG and EFG livers. These latter two groups showed fewer fatty cells than was seen in the groups that had not been given flaxseed, so that the diagnosis of hepatic steatosis was not justified. In conclusion, therefore, this study showed that the indicators of ethanol chronic consumption can be reduced by the introduction of continuous flaxseed dietary intake.
RESUMO Introdução: Esse estudo teve por objetivo avaliar os efeitos toxicológicos da cantaxantina na prole de camundongas consumidoras de truta salmonada. Metodologia: Formou-se quatro grupos: 1-Grupo Caseína (CG) recebendo dieta padrão/controle, 2-Grupo Cantaxantina (CGX) recebendo a ração padrão acrescida de 0,6% de cantaxantina, 3-Grupo Truta Salmonada (STG) recebendo ração padrão acrescida desse peixe e 4-Grupo Salmão Selvagem (WSG) recebendo ração padrão acrescida de salmão selvagem. As mães dos camundongos alvo foram alimentadas durante a gestação e lactação com as mesmas dietas que sua prole, sendo a prole eutanasiada nos períodos de 40 e 70 dias de vida (P40 e P70). Foram coletados dados de peso corporal, de órgãos e consumo das rações. Fígado e rins foram processados de forma rotineira para inclusão em parafina e corados com HE. Resultados: Os fígados, rins direitos e rins esquerdos eram mais pesados nos animais do STG quando comparados aos animais do WSG (P40 e P70). Histopatologicamente, houve sinais de toxicidade em fígado e rins nos grupos CGX e STG (P40 e P70), sendo mais intensos neste último. Os fígados apresentaram esteatose e congestão e os rins, glomérulos tumefeitos, túbulos com necrose e congestão. Conclusão: A cantaxantina tem ação tóxica sobre os rins e fígado de prole de camundongas alimentadas com truta salmonada.
