The antioxidant capacity of methanolic crude extracts of Berberis vulgaris L., Cornus mas L. and Mahonia aquifolium Nutt. was tested with the thiobarbituric acid reactive substances formation assay, the ferric reducing power (FRAP) and 2,2-diphenyl-2-picrylhydrazyl (DPPH • ) radical scavenging assay. The content of antioxidant components in the extracts, their partition coeffi cient on 1-octanol:water and affi nity to liposome membranes were determined as well. The results show that the IC 50 parameter connected with the antioxidant activity on phosphatidylcholine liposome membrane decreased as follows: B. vulgaris (0.14±0.01 mg/mL) > M. aquifolium (0.34±0.03 mg/mL) > C. mas (1.13±0.01 mg/mL) for AAPH-induced oxidation and M. aquifolium (0.29±0.03 mg/mL) > C. mas (1.24±0.07 mg/mL) > B. vulgaris (1.50±0.05 mg/mL) for Fe(II)/ascorbic acid-induced oxidation, and M. aquifolium (2.35±0.10 mg/mL) > B. vulgaris (2.69±0.04 mg/mL) > C. mas (6.17±0.06 mg/mL) for UVC irradiation. All the extracts exhibited the ability to quench DPPH• and to reduce Fe(III) ions to Fe(II) via redox reaction. The content of active components in the extracts, the partition coeffi cient and extracts affi nity to membranes correlated well with their antioxidant activities.This study has shown that fruits of B. vulgaris, M. aquifolium and C. mas, from which the extracts were obtained, are attractive for consumption and can potentially be used in production of new processed fruit.
Celem pracy było określenie właściwości przeciwutleniających wybranych ekstraktów roślinnych (dziurawca, gryki, głogu i kocanki) oraz ich ważnych składników polifenolowych w stosunku do błon liposomów fosfatydylocholinowych (PC) utlenianych promieniowaniem UVC. Określono także właściwości przeciwrodnikowe tych substancji w stosunku do wolnego rodnika 2,2-difenylo-1-pikrylohydrazylu (DPPH •) oraz stałe asocjacji z błoną z zastosowaniem sondy DPH (l,6-difenylo-l,3,5-heksatrienu). Wykazano następującą relację aktywności przeciwutleniającej ekstraktów wyrażoną parametrem IC 50 PC : głógkora (21,69 mg/l) > dziurawiec (25,45 mg/l) > gryka-łuski (30,33 mg/l) > głóg-liście (32,52 mg/l) > gryka-łęciny (37,47 mg/l) > kocanka (115,06 mg/l). Aktywność przeciwutleniająca wybranych składników polifenolowych zmieniała się natomiast w następujący sposób: kwercetyna (0,53 mg/l) > epikatechina (29,19 mg/l) > kwas chlorogenowy (62,59 mg/l) > rutyna (78,97 mg/l). Dodatnia korelacja aktywności przeciwutleniającej badanych ekstraktów oraz związków fenolowych z aktywnością przeciwrodnikową (r ≥ 0,94) wskazuje, że mechanizm ich przeciwutleniającego działania polega na neutralizowaniu wolnych rodników. Kwercetyna spośród badanych flawonoidów wykazywała najwyższą aktywność przeciwutleniającą, wysoką aktywność przeciwrodnikową (IC 50 DPPH• = 2,51 mg/l) oraz wysoką stałą asocjacji z błoną liposomów (K a = 11,23•10 3 l/mg). Możliwość głębokiego zakotwiczenia się molekuł kwercetyny w błonie liposomów fosfolipidowych ogranicza wnikanie wolnych rodników do wnętrza błony co wzmacnia jej skuteczność przeciwutleniającą.
This study aimed to determine the biological activity of Rosa rugosa Thunb. extracts in the four versions: methanolic (ME), diethyl ether (EE), ethyl acetate (EA) and water (WE). We studied the antioxidant activity of the extracts in relation to phospholipid membranes, their association constants with the membrane, and antiradical and antiinflammatory activity. Percentage of their liposomal encapsulation was also assessed. R. rugosa ME and EA extracts were observed to decrease peroxidation of PC liposomes (UVC-induced) to similar levels (ME) or lower (EA) compared with the activity of Rosa canina L. The other two extracts (EE and WE) of R. rugosa showed significantly lower antioxidant activity than extracts of R. canina. R. rugosa quenched DPPH• and inhibited COX activity. R. rugosa EA has the highest association constant, whereas the association constants of EE, ME, and WE are approximately 1-2 orders of magnitude smaller. EA extract also showed the highest percentage of encapsulation in liposomal carriers. PRACTICAL APPLICATIONSLiterature reports attest to the healing properties of extract of R. canina (Litozin, AXELLUS, Denmark). Our studies have demonstrated that extracts of R. rugosa have strong antioxidant (in relation to UV-induced oxidation of phosphatidylcholine liposomes) and antiradical (in relation to DPPH • ) properties, indicating a potential use of these extracts as natural antioxidants, e.g., in the food, pharmaceutical and beauty industry, in particular for protecting peroxidation sensitive components such as unsaturated fatty acids. The ability of R. rugosa extract (the EA version) to strongly inhibit oxidation of arachidonic acid, induced by COX-1 and COX-2, indicates a possibility of application of this extract in prevention and/or elimination of arthritis. The determined high level of encapsulation of R. rugosa EA extract in liposomal carriers may increase the bioavailability of the extract supplied to the body in the form of liposomal nanocapsules and thus be instrumental in effective treatment of inflammatory conditions.
