LL-37 is a C-terminal peptide proteolytically released from 18 kDa human cathelicidin protein (hCAP18). Chronic infections, inflammation, tissue injury and tissue regeneration are all linked with neoplastic growth, and involve LL-37 antibacterial and immunomodulatory functions. Such a link points to the possible involvement of LL-37 peptide in carcinogenesis. An increasing amount of evidence suggests that LL-37 can have two different and contradictory effects—promotion or inhibition of tumor growth. The mechanisms are tissue-specific, complex, and depend mostly on the ability of LL-37 to act as a ligand for different membrane receptors whose expression varies on different cancer cells. Overexpression of LL-37 was found to promote development and progression of ovarian, lung and breast cancers, and to suppress tumorigenesis in colon and gastric cancer. This review explores and summarizes the current views on how LL-37 contributes to immunity, pathophysiology and cell signaling involved in malignant tumor growth.
Background1,4-Dihydropyridine (1,4-DHP) and its derivatives are well-known calcium channel blockers with antiarrhythmic and antihypertensive activities. These compounds exhibit pleiotropic effects including antimicrobial activities that rely on their positive charge and amphipathic nature. Use of magnetic nanoparticles (MNPs) as carriers of 1,4-DHP modulates their properties and enables improved formulations with higher efficacy and less toxicity.MethodsIn this study, the antimicrobial and immunomodulatory activities of novel 1,4-DHP derivatives in free form and immobilized on MNPs were determined by evaluating pathogen outgrowth and proinflammatory cytokine release in experimental settings that involve incubation of various 1,4-DHPs with clinical isolates of bacteria or fungi as well as mammalian cell culture models.ResultsConventional immobilization of 1,4-DHP on aminosilane-coated MNPs markedly enhances their antimicrobial activity compared to nonimmobilized molecules, in part because of the higher affinity of these nanosystems for bacterial cell wall components in the presence of human body fluids.ConclusionOptimized nanosystems are characterized by improved biocompatibility and higher anti-inflammatory properties that provide new opportunities for the therapy of infectious diseases.
The dominant part of human infections is associated with biofilm formations. Biofilm represents structured communities of bacterial or fungal cells enclosed in self-produced polymeric matrixes adherent to supporting surfaces. Microbial DNA and the host cell DNA, after their release at the infection site, show the ability to promote biofilm formation. Between the different constituents of biofilm matrixes, extracellular DNA (eDNA) may be the only component indispensable for the initial attachment and early biofilm formation through an enhanced matrix structural integrity. The effect of DNA on bacterial/fungal attachment is non-specific, as indicated by the stimulatory effect of plasmid, chromosome, or eukaryotic DNA. DNase I impaired bacterial biofilm growth and the targeting eDNA were recently proposed to eliminate and/or prevent different microbial infections associated with biofilm formations. Streszczenie Znaczna część infekcji występujących u ludzi jest związana z powstawaniem biofilmu. Biofilm stanowi złożoną strukturę składającą się z drobnoustrojów oraz wyprodukowanej przez nie polimerowej macierzy, umożliwiającej adhezję do różnych powierzchni. Drobnoustrojowe oraz ludzkie DNA, pochodzące odpowiednio z mikroorganizmów i komórek gospodarza, uwalniane w miejscu infekcji działa jako czynnik promujący rozwój biofilmu. Uważa się, że zewnątrzkomórkowe DNA (eDNA), dzięki zwiększeniu integralności konstrukcji macierzy, jest niezbędnym elementem biorącym udział w adhezji bakterii do podłoża i wczesnym formowaniu się biofilmu. Wyniki badań wykorzystujących plazmidy, chromosomy oraz eukariotyczne DNA wykazały, że wpływ eDNA na rozwój biofilmu jest niespecyficzny i niezwiązany bezpośrednio ze źródłem materiału genetycznego. DNaza I hamuje powstawanie biofilmu. Postuluje się, że wykorzystanie eDNA jako celu terapeutycznego może mieć istotne znaczenie w zapobieganiu i leczeniu zakażeń związanych z formowaniem się biofilmu.
The increased prevalence of antibiotic-resistant pathogens requires additional efforts to develop new antimicrobial agents and alternative methods to prevent and treat infections. In response to this challenge, a variety of nanotechnology-based tools are currently being designed and thoroughly investigated. To date, a considerable number of studies have reported increased activity of antibiotic-conjugated polymeric nanoparticles against bacteria and fungi associated with various infections, including those caused by drug-resistant pathogens. Importantly, high biocompatibility of these structures coupled with enhanced biological activity and improved pharmacokinetic properties supports the potential of these nanosystems as new tools to treat infections. In this review, we summarize the synthesis of polymer-based nanoparticles and describe their mechanism of action. We also highlight the recent advances in the application of antibiotic-conjugated polymeric nanoparticles as novel antimicrobial agents. StreszczenieStale narastająca lekooporność drobnoustrojów wymusza konieczność rozwoju alternatywnych metod profilaktyki i terapii zakażeń, a także uzasadnia prace nad nowymi lekami o aktywności przeciwdrobnoustrojowej. Realizacja tego celu jest moż-liwa dzięki zastosowaniu osiągnięć z zakresu nanotechnologii. Zwiększająca się liczba doniesień literaturowych potwierdza znaczną aktywność przeciwdrobnoustrojową nanocząstek polimerowych skoniugowanych z klasycznymi antybiotykami, co może zostać wykorzystane w terapii infekcji o charakterze bakteryjnym i grzybiczym, również tych wywoływanych przez patogeny lekooporne. Wysoka biokompatybilność nanocząstek polimerowych, a także możliwość zwiększania aktywności przeciwdrobnoustrojowej i poprawy parametrów farmakokinetycznych stosowanych obecnie chemioterapeutyków, wskazuje na możliwość zastosowania nanostruktur w nowoczesnej terapii chorób infekcyjnych. W niniejszej pracy podsumowano metody syntezy nanosystemów opartych na nanocząstkach polimerowych, a także opisano mechanizm ich działania. Przedstawione zostały również najnowsze osiągnięcia w syntezie nanocząstek skoniugowanych z antybiotykami, pozwalające na ich zastosowanie w terapii infekcji o charakterze bakteryjnym i grzybiczym.
