A indústria tem forte interesse sobre a Genipa americana L. para produção de corante a partir do extrato ou suco do fruto. Atualmente, tem-se utilizado o extrato para formulação de agentes e loções corporais antioxidantes, tintura alimentícia e para tatuagem e produtos antileucemicos. Estas novas tecnologias estão embasadas em sua composição quimica com presença de flavonóides e, principalmente, iridóide genipin. O estudo teve por finalidade apresentar o estado da arte sobre a atividade biológica sobre o uso de extrato do jenipapeiro. Observou-se com o trabalho que não há produtos ou formulação relacionando a G. americana L com as atividades biológicas de cicatrização, antimicrobiana, antiparasitária, sendo notificada ação antioxidante e antileucemica do extrato do fruto. Não foi referenciada a produção de gel e microemulsão contendo extrato ou óleo de fruto, folha ou entrecasca de jenipapeiro. No entanto, o principal produto relatado foi a produção de corante, sendo as empresas latino-americanas as maiores depositárias de patentes. Com isto, o estudo mostra que há uma área ainda inexplorada sobre a potencialidade do jenipapeiro para diversos usos biológicos, não só para produção de corante a partir do pigmento azul do fruto.
Plants are important sources of secondary metabolites responsible for various biological activities. In this research, was evaluated the effect action ethyl acetate fraction of Syzygium cumini leaves (Sc-AcOEt) in the antimicrobial activity, contractile and electrical effects in isolated rat hearts and the chemical characterization. Antimicrobial activity was assessed using the disk-diffusion method and the determination of the minimum inhibitory concentration. The contractile and electrical effects were tested using the Langendorf method. The chemical characterization was evaluated through quantification and HPLC analyzes. P. aeruginosa was not viable in any concentration of the fraction (MIC 0,125 mg/mL), while S. aureus showed a MIC of 0.5 mg/mL. K. pneumoniae and E. coli had the highest MIC (1 mg/mL). A reduction in LVDP was observed after heart perfusion with 0.1 mg/mL of Sc-AcOEt. No change was observed in the duration of systole and diastole at all concentrations and in the electrocardiographic parameters. Total phenols, total flavonoids and total flavonols contents of Sc-AcOEt, calculated from the respective calibration curves, were 21.556µg/mg AGE, 617.222µg/mg QE and 315,222µg/mg RE, respectively. In addition, the HPLC analysis of Sc-AcOEt showed the presence of gallic acid, quercetin-3-glucoside, naringin and myricetin as main components. Thus, the Sc-AcOEt has important antimicrobial and cardiac activities possibly due to its chemical constituents.
Naringin is a flavonoid glycoside found in citrus fruits and grapes with a wide range of therapeutic actions. We aimed to study the inotropic effect of naringin on rat-isolated atria, dissecting intracellular mechanisms involved in this response. Concentration-response curves of naringin (0.003-6 mM) were obtained before and after pre-incubation with selective antagonists. Our results showed that naringin presented a biphasic inotropic response, with a positive inotropic effect at low and middle concentrations (0.003-2.0 mM), and a negative inotropic effect at high concentrations (above 3 mM). Pre-incubation with propranolol or atenolol (β-adrenergic receptors antagonists, β-AR), H89 (protein kinase-A inhibitor, PKA), nifedipine (L-type Ca2+ channel blocker), or ryanodine (ryanodine receptor inhibitor) fully abolished the positive inotropic effect induced by naringin. Pre-treatment of animals with reserpine, catecholamine-depleting drug, also prevented the increase of atrial contractility evoked by naringin. Altogether, we show that naringin causes a positive inotropic effect in isolated atria through β-AR/PKA-dependent pathway.
A celulose é um biopolímero com elevado potencial de aplicação em áreas como a têxtil, alimentos e farmacêutica. Porém, a baixa solubilidade em água dificulta tais aplicações, o que pode ser contornado via modificações na estrutura, incluindo a oxidação medida por N-oxil-2,2,6,6-tetrametilpiperidina (TEMPO). Além de aprimorar a solubilidade, as modificações abrem oportunidades de obtenção de novos materiais, dentre os quais os hidrogéis, que podem ser aplicados como adsorventes, curativos ou carreadores de fármacos. O presente trabalho avaliou a eficiência de adsorção de sulfato de gentamicina e a atividade antibacteriana dos hidrogéis a base de celulose microcristalina, previamente oxidada via mediação por TEMPO, reticulados quimicamente com epicloridrina, na ausência e na presença dos modificadores β-ciclodextrina e κ-carragena. Os resultados de espectroscopia no infravermelho evidenciaram tanto o sucesso da oxidação da celulose quanto do processo de reticulação, enquanto a difratometria de raios X mostrou o caráter predominantemente não cristalino dos hidrogéis. Imagens de microscopia eletrônica de varredura revelaram morfologias semelhantes a esponjas e, finalmente, a avaliação da atividade antibacteriana por meio do teste de difusão em ágar mostrou que os hidrogéis preparados apresentam grande potencial como possíveis candidatos a utilização como carreadores de fármaco.
