The aim of the present study was to determine the prevalence and risk factors associated with Campylobacter fetus subsp. venerealis and Tritrichomonas foetus infection in cows from dairy herds in the state of Pernambuco, Brazil. In total, 383 samples of cervico-vaginal mucus were collected from cows on 21 herds in 19 districts. Genomic DNA was extracted from the samples and submitted for polymerase chain reaction analysis. An investigative questionnaire was used to analyze the risk factors, using questions related to reproductive and hygiene/sanitation management. A prevalence of 1.8% (0.8-3.9%; confidence interval (CI) 95%) and 33.4% (28.7-38.4%; CI 95%) was found for C. fetus subsp. venerealis and T. foetus, respectively. In terms of the number of foci, 28.6% of the herds contained at least one animal that was positive for C. fetus subsp. venerealis and 90.5% for T. foetus. The present study identified herds larger than 100 animals as a risk factor for bovine genital campylobacteriosis (OR = 7.2; CI 1.3-38.4%; p = 0.020) and the use of natural breeding as a risk factor for bovine trichomonosis (OR = 2.4; CI 1.1-5.9%; p = 0.041). In conclusion, C. fetus subsp. venerealis and T. foetus infections occurred in the region studied and high numbers of foci were found. Thus, prophylaxis and control measures, such as diagnosis, separation, and sexual rest for infected females, are suggested. An artificial insemination program with early rigorous sanitary care should be implemented on the properties in order to avoid the spread of agents in the herds.

Coxiella burnetii, an intracellular bacterium, is the agent of Q fever/coxiellosis, a worldwide zoonosis. Dairy animals are the primary reservoirs of C. burnetii, and although the disease is usually asymptomatic or subclinical, abortion is a serious clinical outcome among small ruminants. This study was conducted to investigate C. burnetii seroprevalence and infection In a flock of dairy goats in Brazil. Serum samples from 312 goats collected from a dairy goat flock with a history of reproductive failure were tested by a commercial ELISA (LSIVet Ruminant Q Fever - Serum/Milk; Thermo Fisher Scientific, Lissieu, France) for anti-C. burnetii IgG antibodies. Samples of cotyledons from 23 placentas were analyzed by nested PCR for the presence of the bacterial DNA. ELISA seroreactivity was found in 55.1% (172/312; 95% CI = 49.4%-60.7%) of the serum samples analyzed. C. burnetii DNA was detected in 8.7% (2/23) of the placental samples tested, where both animals were also seropositive. This study reports the first description of C. burnetii infection in an abortion outbreak in goats in Brazil. The results point out to the importance of including this disease in animal and public health surveillance programs as well as into the list of abortive diseases in goats in Brazil.

Objetivou-se com esse estudo avaliar a eficácia in vitro de desinfetantes comerciais utilizados no pré e pós- -dipping, frente a Staphylococcus spp. isolados do leite de vacas procedentes de propriedades leiteiras do Agreste e Zona da Mata do Estado de Alagoas. Foram utilizados iodo (0,57%), clorexidine (2,0%), cloro (2,5%) e composto de amônio quaternário (4,0%), nas concentrações indicadas, como desinfetantes comerciais usados convencionalmente no pré e pós-dipping. Analisou-se um total de 97 isolados de Staphylococcus spp. identificados como S. aureus (16), Staphylococcus coagulase positiva (7) e Staphylococcus coagulase negativa (74). Os desinfetantes foram avaliados em três tempos distintos (15", 30" e 60"). Observou-se que 56,3% de Staphylococcus aureus foram sensíveis ao iodo, 68,8% sensíveis ao cloro, 87,5% à clorexidine e 37,5% ao composto de amônia no tempo de 60". Quanto aos Staphylococcus coagulase positiva (SCP), 100% dos isolados foram resistentes ao clorexidine, 85,7% ao composto de amônio, 57,1% ao cloro, e 42,9 resistentes ao iodo no tempo de 60". Em relação aos Staphylococcus coagulase negativa (SCN) foi observado 91,9% de sensibilidade ao clorexidine, 70,3% sensíveis ao cloro, 66,2% ao iodo e 24,3% sensíveis ao composto de amônio no tempo de 60". Conclui-se com esse estudo que a maior atividade desinfetante in vitro foi verificada para clorexidine e cloro frente aos S. aureus, iodo e cloro para os SCP e clorexidine e cloro para os SCN. Devido às variações no perfil de sensibilidade e resistência encontradas, é necessária a avaliação regular da eficiência dos desinfetantes usados nas propriedades, com o intuito de observar a eficácia do produto e assim garantir o controle da mastite no rebanho.

Para isso, foram submetidas ao exame microbiológico 1260 amostras de leite e em cada propriedade foi aplicado um questionário contendo informações relacionadas às características raciais dos animais, dados gerais sobre as propriedades, manejo do rebanho e higiênico-sanitário durante a ordenha. Para a análise dos fatores de risco foi realizada uma regressão logística considerando como variável dependente o exame microbioló-gico. Ao exame microbiológico observou-se 477 (37,9%) The objective of this study was to analyze the risk factors for bovine mastitis in the micro-region of Garanhuns, Pernambuco, Brazil. For this, was submitted for microbiologic exam 1260 milk samples and on each farm was applied a questionnaire about characteristics of animal breeds, general data from the farm, handling of cattle and sanitary measures for milking. To analyze the risk factors, a logistic regression was made as a dependent variable of the microbiologic exam. The microbiologic exams showed 477 (37.9%) positive samples and 783 (62.1%) negatives ones. The risk factors for bovine mastitis were: the semi-intensive breeding system (OR=2.3, p<0.0001), feeding during milking (OR=1.3, p=0.0101), antimicrobial shift (OR=1.3, p=0.00770), and dry cow therapy (OR=1.5; p=0.0013). The results show that the main risk factors are associated with handling to which the cattle is submitted. The knowledge of these factors allow to adopt prevention and control measures to avoid mastitis to improve animal productivity and gain within the production system. The aim of this study was to detect the IS900 region of Mycobacterium avium subsp. paratuberculosis (MAP) in bovine milk samples using real-time polymerase chain reaction (qPCR) and conventional PCR, and to study the agreement between these tests. A total of 121 bovine milk samples were collected from herds considered positive for MAP, from the State of Pernambuco, Brazil. MAP DNA was detected in 20 samples (16.5%) using conventional PCR and in 34 samples (28.1%) using qPCR. MAP DNA was detected in all of the 6 animal farms studied. Moderate agreement was found between qPCR and conventional PCR results, where the sensitivity and specificity of conventional PCR in relation to qPCR were 50% and 96.6%, respectively. Thus, the IS900 region of MAP was found in bovine milk samples from the State of Pernambuco. To the best of our knowledge, this is the first report of MAP DNA found in bovine milk in Northeast Brazil. We also demonstrated the qPCR technique is more sensitive than conventional PCR with respect to detection of MAP in milk samples.
