This work describes the mechanism of action of some reactive oxygen species (ROS) and reactive nitrogen species (RNS) in the oxidative stress of the human body, and their consequences on damage to DNA, RNA, proteins and lipids. It also illustrates the defense system of our organism against these ROS and RNS species. The action of nonenzymatic protection systems is reported, with emphasis on micromolecules like Q10 coenzyme, vitamin C, α-tocopherol, carotenoids and flavonoids. The importance of flavonoids is also emphasized, and their body protection mechanism is detailed.Keywords: oxidative stress; antioxidants; reactive species. INTRODUÇÃOAtualmente existe um grande interesse no estudo dos antioxidantes devido, principalmente, às descobertas sobre o efeito dos radicais livres no organismo. A oxidação é parte fundamental da vida aeróbica e do nosso metabolismo e, assim, os radicais livres são produzidos naturalmente ou por alguma disfunção biológica. Esses radicais livres cujo elétron desemparelhado encontra-se centrado nos átomos de oxigênio ou nitrogênio são denominados ERO ou ERN [1][2][3][4] . No organismo, encontram-se envolvidos na produção de energia, fagocitose, regulação do crescimento celular, sinalização intercelular e síntese de substâncias biológicas importantes. No entanto, seu excesso apresenta efeitos prejudiciais, tais como a peroxidação dos lipídios de membrana e agressão às proteínas dos tecidos e das membranas, às enzimas, carboidratos e DNA 5 . Dessa forma, encontram-se relacionados com várias patologias, tais como artrite, choque hemorrágico, doenças do coração, catarata, disfunções cognitivas, câncer e AIDS, podendo ser a causa ou o fator agravante do quadro geral 6 . O excesso de radicais livres no organismo é combatido por antioxidantes produzidos pelo corpo ou absorvidos da dieta. De acordo com Halliwell 3 "Antioxidante é qualquer substância que, quando presente em baixa concentração comparada à do substrato oxidável, regenera o substrato ou previne significativamente a oxidação do mesmo". Os antioxidantes produzidos pelo corpo agem enzimaticamente, a exemplo da GPx, CAT e SOD 2 ou, não-enzimaticamente a exemplo de GSH, peptídeos de histidina, proteínas ligadas ao ferro (transferrina e ferritina), ácido diidrolipóico e CoQH 2 . Além dos antioxidantes produzidos pelo corpo, o organismo utiliza aqueles provenientes da dieta como o a-tocoferol (vitamina-E), β-caroteno (pro-vitamina-A), ácido ascórbico (vitamina-C), e compostos fenólicos onde se destacam os flavonóides e poliflavonóides 4,7 . Dentre os aspectos preventivos, é interessante ressaltar a correlação existente entre atividade antioxidante de substâncias polares e capacidade de inibir ou retardar o aparecimento de células cancerígenas, além de retardar o envelhecimento das células em geral 8 . In the literature there are a considerable number of chemical and biochemical tests for evaluation of in vitro antioxidant activities of pure compounds or fractions and organic extracts. These tests are important tools for screening of synthetic and natural bioactive compound as well as they can be employed in food chemistry. INTRODUÇÃOO termo oxidação de uma substância era comumente definido como a incorporação de oxigênio em sua estrutura. Atualmente, pode ser mais precisamente definido como sendo a conversão de uma substância química em um derivado com menor número de elétrons. Oxidação, portanto, é a perda de um ou mais elétrons para outra substância e o procedimento inverso pode ser considerado como redução. 1 A transferência de elétrons é um dos processos químicos mais fundamentais para a sobrevivência das células. O efeito colateral dessa dependência é a produção de radicais livres e outras espécies reativas de oxigênio (ERO) que podem causar dano oxidativo. Radicais livres são átomos ou moléculas produzidos continuamente durante os processos metabólicos e atuam como mediadores para a transferência de elétrons em várias reações bioquímicas, desempenhando funções relevantes ao metabolismo.2 As principais fontes de radicais livres são as organelas citoplasmáticas que metabolizam oxigênio, nitrogênio e cloro, gerando grande quantidade de metabólitos. Os radicais livres possuem diferentes papéis no organismo e encontram-se envolvidos na produção de energia, fagocitose, regulação do crescimento celular, sinalização intercelular e síntese de substâncias biológicas importantes. Entretanto, seu excesso apresenta efeitos deletérios, tais como danos ao DNA, proteínas e organelas celulares, como mitocôndrias e membranas, provocando alterações na estrutura e funções celulares e, dessa forma, se encontram envolvidos em diversas patologias a exemplo de câncer, envelhecimento precoce, doenças cardiovasculares, degenerativas e neurológicas, choque hemorrágico, catarata, disfunções cognitivas, etc.4 Para combater os radicais livres os organismos vivos produzem substâncias que são capazes de regenerar ou prevenir os danos oxidativos, exercendo seu papel como antioxidante. Além destes, substâncias com habilidade de sequestrar radicais livres podem ser obtidas de fontes externas, como alimentos e bebidas. In this paper we report the geometries and properties of 24 structural isomers located on the MP2/6-311++g** potential energy surface of the water hexamer. At least 15 structural patterns are located within 3 kcal/mol of the most stable conformation, leading to a very complex potential energy surface, several isomers having significant contributions. A quadratic correlation between the distance from the proton to the center of the hydrogen bond with the distance between oxygen atoms for all clusters is reported. MP2/6-311++g** and CCSD(T)/aug-cc-pvdz//MP2/6-311++g** predict different stabilization orderings but are in good agreement for binding energies. Compact structures are energetically favored by electronic energies with zero point energy corrections, while noncompact cyclic structures are preferred when temperature and entropy are accounted for.
