The rapid development of cities in the recent 10 years caused a reduction in the cultivated land area, which only accounts for 14% of the total land area in China. Land development and reclamation have been regarded as an effective way to compensate farmland occupation. However, most of the newly reclaimed land has poor soil fertility and suitability; in some cases, the production capacity is only 10–30% of the occupied farmland. In order to ameliorate the soil quality of the newly reclaimed land, this study evaluated the effects of commercial organic fertilizer (0.75, 1.50, and 2.25 kg/m2), mushroom residue (1.50, 2.25, and 3.00 kg/m2), biogas slurry (150, 225, and 300 kg/m2), vegetable cake (0.30 and 0.60 kg/m2), and chemical compound fertilizer (37.50 g/m2) on the pH, moisture content and organic matter content (OMC), available phosphate, total nitrogen, alkaline hydrolysis nitrogen, microbial biomass carbon and nitrogen, and number of total bacteria and phosphate-solubilizing bacteria, as well as the growth of maize seedlings. The results from this study indicate that the soil quality (OMC is an indicator) was ameliorated by chemical and organic fertilizers, in particular commercial organic fertilizers, which caused a 9.35–16.35% increase in moisture content, a 11.56–18.72% increase in pH, a 1.73–2.15 fold increase in OMC, a 338.44–491.41% increase in available P, a 36.80–48.14% increase in total N, a 95.32–128.34% increase in alkaline hydrolysis N, a 92.57–178.38% increase in total bacterial numbers, and a 7.57–20.87 fold increase in microbial biomass carbon compared with the control. The pot experiment further indicated that soil amended with commercial organic fertilizers caused a 20.35–30.55% increase in the height and a 12.50–16.67% increase in the total dry weight of maize seedlings. In addition, representative strains with the ability to dissolve phosphorus and fix nitrogen were successfully isolated using the culture method, and were then identified based on colony morphological observation and 16S rDNA sequence analysis, which help us to not only understand why organic fertilizer has great effect on soil improvement, but also provides beneficial microbial resources for further study.
Bacteria belonging to the genus Paenibacillus were frequently isolated from legume nodules. The nodule-inhabiting Paenibacillus as a resource of biocontrol and plant growth-promoting endophytes has rarely been explored. This study explored the nodule-inhabiting Paenibacillus’ antifungal activities and biocontrol potentials against broad-spectrum important phytopathogenic fungi. We collected strains which were isolated from nodules of Robinia pseudoacacia, Dendrolobium triangulare, Ormosia semicastrata, Cicer arietinum, Acacia crassicarpa, or Acacia implexa and belong to P. peoriae, P. kribbensis, P. endophyticus, P. enshidis, P. puldeungensis, P. taichungensis, or closely related to P. kribbensis, or P. anseongense. These nodule-inhabiting Paenibacillus showed diverse antagonistic activities against five phytopathogenic fungi (Fusarium graminearum, Magnaporthe oryzae, Rhizoctonia solani, Sclerotinia sclerotiorum, and Botrytis cinerea). Six strains within the P. polymyxa complex showed broad-spectrum and potent activities against all the five pathogens, and produced multiple hydrolytic enzymes, siderophores, and lipopeptide fusaricidins. Fusaricidins are likely the key antimicrobials responsible for the broad-spectrum antifungal activities. The nodule-inhabiting strains within the P. polymyxa complex were able to epiphytically and endophytically colonize the non-host wheat plants, produce indole acetic acids (IAA), and dissolve calcium phosphate and calcium phytate. P. peoriae strains RP20, RP51, and RP62 could fix N2. P. peoriae RP51 and Paenibacillus sp. RP31, which showed potent plant colonization and plant growth-promotion competence, effectively control fungal infection in planta. Genome mining revealed that all strains (n = 76) within the P. polymyxa complex contain ipdC gene encoding indole-3-pyruvate decarboxylase for biosynthesis of IAA, 96% (n = 73) contain the fus cluster for biosynthesis of fusaricidins, and 43% (n = 33) contain the nif cluster for nitrogen fixation. Together, our study highlights that endophytic strains within the P. polymyxa complex have a high probability to be effective biocontrol agents and biofertilizers and we propose an effective approach to screen strains within the P. polymyxa complex.
Land reclamation may expand the supply of usable land for food security. Immature soil is not suitable for plant growth and needs to be amended by the addition of organic matter and plant growth-promoting (PGP) microorganisms. However, the effects of different PGP fungi on plant growth in immature soil are largely unexplored. In order to obtain beneficial soil microorganisms with a good PGP ability in new reclamation land, 162 fungal isolates were isolated from different abandoned wastelands, four isolates of which were obtained in this study by the screening of P solubilization, siderophore production, and indole acetic acid (IAA) production. The result of this study revealed that isolate HZ123 had the highest ability to solubilize P and produce siderophores and IAA, followed by HZ23, HZ10, and HZ06. Based on the results of morphological and molecular analyses, isolate HZ06 was identified as Penicillium oxalicum, isolates HZ23 and HZ10 were identified as Aspergillus brunneoviolaceus, and isolate HZ123 was identified as Aspergillus tubingensis. Furthermore, the results of in vivo PGP assays demonstrated that isolate HZ123 has a minimal negative effect on the growth of eggplant; however, the other three isolates, particularly isolate HZ06, caused the greatest increase in eggplant biomasses. Overall, these results indicate that isolate HZ06 has great potential as a PGP fungus to develop biofertilizer for application in eggplant production in immature soil from new reclamation land.
It is well known that soil fertility is poor in most newly reclaimed land, which has been developed as an effective way to compensate farmland occupation. In order to ameliorate the soil quality of the newly reclaimed land, this study investigated the effect of commercial organic fertilizer (COF) (0.45, 0.90, 1.35 and 1.80 kg/m2), sheep manure (SM) (0.45, 0.75, 1.05 and 1.35 kg/m2), mushroom residue (MR) (1.50, 2.25, 3.00 and 3.75 kg/m2), and chemical compound fertilizer (CCF) (0.075 kg/m2) on the growth of sweet potato, soil pH, organic matter content (OMC), available phosphate, total nitrogen, available potassium, exchangeable Ca and Mg, as well as bacterial and fungal microbial composition during 2019–2021. The results from this study indicated that the COF, SM, MR, and CCF did not significantly change the soil pH, but significantly increased the OMC, which has been regarded as the most significant soil quality parameter. This suggests that the soil amendments used in this study have great potential to improve the soil quality in newly reclaimed land. However, these soil amendments exhibited a differential effect on sweet potato biomass, nutrient elements and the microbial community of the newly reclaimed soil, which depend on the kind and concentration of organic/chemical fertilizer, the application time, as well as the plant and soil parameters. The change was also observed on the bacterial and fungal soil microbial community, which provides us with a microbial basis to understand why organic fertilizer has a great effect on soil improvement. Overall, our results suggest that soil amended with organic fertilizers has great potential for the production of sweet potato in immature soil from the new reclamation land.
To gain better insight into the regulatory networks of anthocyanin biosynthesis, an integrated analysis of the metabolome and transcriptome in purple and green leaves of Tetrastigma hemsleyanum was conducted. Transcript and metabolite profiles were archived by RNA-sequencing data analysis and LC-ESI-MS/MS, respectively. There were 209 metabolites and 4211 transcripts that were differentially expressed between purple and green leaves. Correlation tests of anthocyanin contents and transcriptional changes showed 141 significant correlations (Pearson correlation coefficient >0.8) between 16 compounds and 14 transcripts involved in the anthocyanin biosynthesis pathway. Some novel genes and metabolites were discovered as potential candidate targets for the improvement of anthocyanin content and superior cultivars.
