Se estableció un experimento en invernadero durante tres épocas del año para determinar el efecto de la época de aplicación de AIB sobre el enraizamiento, características de las raíces formadas, así como del crecimiento posterior de las plantas enraizadas de Ficus benjamina L. Altos porcentajes de enraizamiento se obtuvieron en las tres épocas para todos los tratamientos, incluso el testigo. Sin embargo, valores ligeramente mayores para todas las variables se obtuvieron durante la época de primavera. El tratamiento con 1,500 ppm de AIB produjo el máximo porcentaje de enraizamiento y el mayor volumen de raíces formadas. La dosis de 10,000 ppm de AIB, al igual que el testigo fue ligeramente inhibitoria para las variables estudiadas. Al término de 50 días los tratamientos de 1,500 y 3,000 ppm de AIB favorecieron el crecimiento en altura y número de hojas formadas. No existen diferencias en los contenidos de azúcares solubles totales en las tres épocas del año.
Se determinó la existencia de hibridación natural entre P. oocarpa Schiede ex Schlechtendal y P. pringlei Shaw analizando una muestra de 30 árboles que habitan naturalmente en simpatría y que incluyó individuos catalogados como híbridos putativos. El área de hibridación se localiza en el municipio de Ario de Rosales, Michoacán. Se utilizó un análisis discriminante de variables canónicas con 19 características morfológicas y anatómicas de acículas, conos y semillas. Los árboles inicialmente catalogados como híbridos putativos resultaron con valores intermedios en la mayoría de las características estudiadas. De éstas las que mayormente contribuyeron en las funciones de discriminación canónica y en la separación de las especies progenitoras de los árboles híbridos en estudio, resultaron ser el número de acículas por fascículo, el de hileras de estomas en la cara dorsal, el de canales resiníferos internos y septales, así como la longitud del pedúnculo y el diámetro del cono. Un segundo análisis discriminante fue realizado con muestras de diez árboles de P. oocarpa y siete de P. pringlei de rodales puros habitando alopátricamente y los de los 30 árboles del rodal en simpatría. El análisis gráfico indicó que los árboles de cada especie se aglomeraron opuestamente unos de otros, aunque los individuos de los rodales puros lo hicieron ligeramente hacia los extremos, lo que puede indicar un posible proceso de infiltración genética en el área de hibridación en estudio.
En el presente estudio se determinó la variación existente en características reproductivas y la germinación de la semilla cosechada en árboles de Pinus leiophylla Schiede ex Schltdl. & Cham. en un huerto semillero sexual. Se midió el peso seco de 20 estróbilos maduros por árbol. Se determinaron las variables: número potencial de semillas; porcentajes de semillas desarrolladas, de semillas llenas, de semillas vanas y de óvulos abortivos; peso promedio de la semilla; índice de endogamia y eficiencia reproductiva por estróbilo. La semilla se germinó sobre tela en cajas de plástico, en una cámara de ambiente controlado. Se encontraron diferencias significativas entre árboles para las variables evaluadas. El peso seco promedio del estróbilo fue 15.2 g. El número potencial promedio de semillas por estróbilo fue 108, de las cuales 63.7, 17.7 y 18.6 % fueron óvulos abortivos, semillas llenas y semillas vanas, res-pectivamente. El peso promedio de una semilla fue 11.6 mg (86 200 semillas por kg). El índice promedio de endogamia (0.50) indica un alto nivel de autofecundación. Se encontró una eficiencia reproductiva de 15.9 mg de semilla por gramos de estróbilo. La semilla mostró calidad alta en: capacidad germinativa (82.5 %), valor pico promedio (8.5) y el valor germinativo promedio (29.4). Los árboles contribuyeron con 41.3, 29.3 y 36.9 % de la variación total para capacidad germinativa, valor pico y valor germinativo, lo que indica una alta variación genética en las características reproductivas y parámetros genéticos. A pesar del alto índice de endogamia y del bajo porcentaje de semillas llenas por estróbilo, los árboles de P. leiophylla en el huerto semillero produjeron semilla llena de alta calidad, de acuerdo con los resultados de germinación.
One of the most important things in the field of adult hippocampal neurogenesis (AHN) is the identification of the newly generated cells. The immunodetection of thymidine analogs (such as 5-Bromo-2'-deoxyuridine (BrdU)) is a standard technique used for visualizing these newly generated cells. Therefore, BrdU is usually injected in small animals intraperitoneally, so the thymidine analog gets incorporated into dividing cells during DNA synthesis. Detection is performed by immunohistochemical analysis of brain slices. Every research group that has been using this technique can appreciate that it requires special attention to minute details to achieve a successful stain. For instance, an important step is DNA denaturation with HCl, which allows it to reach the cell nucleus to stain it. However, the existing scientific reports describe very few of such steps in detail. Therefore, standardizing the technique is challenging for new laboratories as it can take several months to yield positive and successful outcomes. The purpose of this work is to describe and elaborate the steps to obtain positive and successful outcomes of the immunostaining technique in detail when working with the thymidine analog BrdU. The protocol includes the reagent preparation and setup, administration of thymidine analog in a rodent, transcardial perfusion, tissue preparation, peroxidase immunohistochemical reaction, use of avidin-biotin complex, immunofluorescence, counterstaining, microscopy imaging, and cell analysis.
Con el propósito de determinar la variación inter e intraespecífica en la tasa de crecimiento en altura y el número de ciclos, se evaluó una plantación experimental de 18 meses de edad, establecida en Patoltecoya, Puebla, durante un periodo de 12 meses. La plantación incluye un total de 30 procedencias de 4 especies de Pinus (10 de P. greggii Engelm., 11 de P. patula Schl. et Cham., 4 de P. tecunumanii (Schw.) Eguiluz et Perry y 5 de P. maximinoi H. E. Moore), en un diseño experimental de parcelas divididas en bloques completos al azar, en dos sitios contiguos. No se encontró un efecto significativo del sitio, ni de la interacción genotipo*ambiente en ninguna de las variables del crecimiento evaluadas, pero sí variación significativa ( p ≤ 0.01) entre y dentro de especies en estas características. La variación entre especies fue de 2 a 6 veces mayor que la variación intraespecífica, por lo que existe un mayor potencial de selección a nivel de especies que de procedencias en dichas características. Pinus maximinoi y P. tecunumanii presentaron las mayores tasas de crecimiento en altura (2.50 y 2.44 m /año respectivamente), mientras que P. patula presentó las menores (1.92 m/año). En P. tecunumanii y P. patula la tasa de crecimiento se asoció en forma negativa ( r = -98) en la primera y en forma positiva (r = 0.75) en la segunda con el número de ciclos de crecimiento, mientras que en P. greggii y P. maximinoi no se encontró una correlación significativa entre estas características. Además, se detectaron diferencias importantes entre las especies en el patrón de variación geográfica en las características del crecimiento evaluadas.
